Project description:Metastasis accounts for the leading cause of treatment failure in nasopharyngeal carcinoma (NPC). However, the exact mechanisms underlying metastasis remain elusive. S18 and S26 are a pair of NPC cell lines with high and low metastasis abilities, respectively. To evaluate the expression profiles of mRNA in S18 and S26 cells will help to demonstrate genes that contribute the metastasis of NPC. We performed microarray sequencing to clarify the genes that differentially expressed in S18 and S26 cells.

Project description:The development of therapeutic resistance and metastatic dissemination takes place in 20-30% patients with nasopharyngeal carcinoma (NPC), resulting in poor survival. Hence, a better understanding of the underlying molecular mechanisms will help improve clinical outcome. We have identified a novel circRNA, circIPO7, as a promoter of metastasis and cisplatin chemoresistance in NPC cells. High-throughput RNA sequencing analysis was performed in the human nasopharyngeal carcinoma cell line S18 after transfection with control siRNA or circIPO7 siRNA.

However, the function and mechanisms of circRNAs in nasopharyngeal carcinoma (NPC) metastasis remain largely unknown. In this study, we identified circWDR37 as a metastasis related circRNA. To further explore the underlying mechanism of circWDR37 promoting NPC metastasis, we performed RNA sequencing analysis of NPC cell line S18 upon circWDR37 knockdown.

Long non-coding RNAs and mRNAs in nasopharyngeal carcinoma metastasis

Project description:Increasing evidence has demonstrated a significant role for long non-coding RNAs (lncRNAs) in tumorigenesis. However, their functions in nasopharyngeal carcinoma (NPC) metastasis remain largely unknown. In this study, a model compared high and low metastatic NPC cell lines (5-8F vs. 6-10B and S18 vs. S26)was constructed to determine the expression profile of lncRNAs using the microarray analysis, and we found 167 lncRNAs and 209 mRNAs were differentially expressed. Validationof 26 significantly dysregulated lncRNAs by qRT-PCR showed the expression patterns of 22 lncRNAs were in accordance with the microarray data. Furthermore, the expression level of ENST00000470135, which was the most upregulated lncRNA in high metastatic cell lines, was significantly higher in NPC cell lines and tissues with lymph node metastasis（LNM）and knocking down ENST00000470135 suppressed the migration, invasion and proliferation of NPC cells in vitro. In conclusion, our study revealed expression patterns of lncRNAs in NPC metastasis. The dysregulated lncRNAs may act as novel biomarkers and therapeutic targets for NPC.

Expression profiling of nasopharyngeal carcinoma cell lines

Project description:To identify the deregulated genes which may involved in the carcinogenesis of nasopharyngeal carcinoma (NPC). To identify potential tumor suppressor genes / oncogenes of NPC, we have employed whole genome microarray expression profiling as a discovery platform to analyze two NPC cell lines, TW01 and HONE1. For the purpose to filter out differential genes may not related to carcinogenesis of NPC, a gene expression database for premalignant nasopharyngeal epithelial cells was obtained from NP460hTert cells, and we used the commercial universal human RNA (stratagene) as common reference for all microarray experiments. A total of 1272 genes were commonly up-regulated (fold change > 2) and 1810 genes were commonly down-regulated (fold change < 0.5) in TW01 and HONE1 cell lines, whereas not deregulated in NP460hTert cells.
