Project description:Tumor cell radioresistance is a major challenge in radiotherapy. By contrast, radiation response heterogeneity of tumor cells is poorly understood. Here, we performed scRNA-seq and scATAC-seq of 6 Gy γ-ray treated human lung adenocarcinoma cells to dissect cellular radiation response heterogeneity from perspective of transcriptional and regulatory cell states. Notably, we observed heterogeneous radioresponsiveness in purified lung adenocarcinoma cells by qualified cellular radiation response based on transcriptional landscape. There was DNA repair and apoptosis related transcriptional cell state appeared post-irradiation. We identified gene markers in this radiation-associated cell state and screened promising targets for LUAD radiosensitization based on 107 transcriptomic data of radiotherapy treated LUAD patients from TCGA database. Besides, transcriptional cell states transition was obvious accompanied with radiation-induced cell cycle arrest in which cell-cell communication was a potential regulatory mechanism. Finally, we identified radiation-associated regulatory cell state and analyzed the regulatory network of key transcriptional factors in cellular radiation response.

Project description:Radiation is the frontline treatment for malignant gliomas. Intra-tumoral heterogeneity has been proposed to grant cancer cells a superior trajectory and survival advantage to avoid therapeutic interventions including radiation. However, direct evidence to support the hypothesis via the transcriptome dynamics of glioma during radiation therapy is limited. The current study aim to measure the functional subpopulation dynamics before and after radiation treatment that assist the radiation resistance at single cell resolution. We investigate the single cell transcriptome and biological pathways of primary glioma mouse model and post-radiation early/late time point. Specifically, we used the RCAS mouse model for gliomas, which overexpress PDGFRA as the model. Using single cell transcriptome, for the first time, we confirmed the proneural classification of PDGFRA RCAS glioma mouse model and its heterogeneity. We found that recurrent dominant subpopulations are featured with elevated proliferation rate and hypoxia. In addition, we identified a subpopulation of radiation resistant cells in at early time points with elevated stemness. Lastly, the subpopulations composition undergoes large changes at late time point when the tumor recurred. Single cell transcriptome profiling of radiation treated mouse glioma mouse model identified tumor cell subpopulations dynamics. It provides novel insights into the molecular phenotype and biological functions of radiation resistant tumor cell population.

Project description:The biomedical consequences of space radiation pose a significant concern for astronauts engaged in deep space. However, the effects of long-term low dose-rate exposures in space environments remain elusive. In this study, we simulated the space radiation environment by exposing human bronchial epithelial cells to low dose-rate (0.0067 Gy/day) α-particles, and continuously irradiated them multiple times to achieve cumulative total doses of 0.2 Gy, 0.4 Gy, and 0.5 Gy, respectively. At the same time, the cells were irradiated with the same total dose in a single exposure to investigate the potential of low dose-rate alpha particles to induce malignant transformation of human bronchial epithelial cells. A comprehensive suite of assays was employed to assess tumorigenic potential, including tumor formation in NOD/SCID mice, immunohistochemistry, CCK-8 proliferation assay, invasion assay, and the evaluation of multicellular spheroid formation during subsequent passages post-irradiation. Moreover, we dissected differential malignant mechanisms in tumor evolution ecosystem induced by the two distinct irradiation modes from systems biology views based on scRNA-seq technology. Our results showed that exposure to α-particles, whether through a single acute exposure or long-term low dose-rate exposures, induced the occurrence and development of tumors. Long-term low dose-rate exposures to α-particles increase the malignancy of induced tumors, but not the risk of carcinogenesis, compared to a single acute exposure with the same total dose. In addition, through scRNA-seq, we found that long-term low dose-rate exposures triggered more copy number variation (CNV) and epithelial-mesenchymal transition (EMT) events, and the activation of DNA damage repair pathways occurred significantly later than with a single acute exposure and involved more specific changes in cellular communication dynamics. In conclusion, our findings provide emerging yet convincing evidence that not only sheds light on why cells exposed to long-term low dose-rate exposures exhibit heightened malignancy, but also offers valuable insights into the genetic determinants driving tumor evolution and heterogeneity.

Project description:To study how radiation alters the different types of cells during tumor recurrence in high-risk MYC-amplified MB, we performed scRNA-seq using the 10X Genomics scRNA-seq platform to compare the primary untreated tumors, residual tumors immediately following radiotherapy, and the spontaneously recurrent tumors 7-8 weeks post-irradiation using OLIG2-high MB002 PDX. Project description:The immunologic effects of radiation (RT) are influenced by dose and each may be optimized over a unique dose range. We hypothesized that delivering a heterogenous dose of RT would enhance anti-tumor immunity compared to homogenous RT in combination with immune checkpoint blockade (ICI). Using a point source, we delivered a spectrum of dose (~2-32 Gy) to the primary tumor and observed superior systemic immunity in models of metastatic cancer compared to low (2 Gy), moderate (8 Gy), or high (20 Gy) dose homogenous RT. Heterogeneity in RT dose resulted in spatial heterogeneity in gene expression and immune cell infiltration. Moreover, heterogenous RT optimally engaged multiple immune signaling pathways with diverse dose response relationships whereas responses to homogenous RT treatment groups were limited. Single cell RNAseq demonstrated unique enrichment of immune cell populations between each homogenous RT dose group, with several of these dose-dependent changes also represented in the heterogeneously treated tumors, with engagement of both lymphoid and myeloid compartments. This optimized activation of multiple immune mechanisms with distinct dose-dependent profiles within a single tumor may underlie a greater capacity of RT heterogeneity to augment anti-tumor immune response when combined with ICI, as compared to homogenous dose RT.
