Project description:The JAK1/2 inhibitor ruxolitinib is a cornerstone of management for some subsets of BCR-ABL negative myeloproliferative neoplasms (MPNs), but some patients respond suboptimally to ruxolitinib.Here, we evaluated the efficacy of micheliolide (MCL) alone or in combination with ruxolitinib in JAK2V617F mutated MPN cell lines, MPN patient primary samples and Jak2V617F knock-in mouse model and found that MCL was able to exert effective therapeutic effects in MPNs. To gain insights of the molecular mechanisms by which MCL exerts effects, we then performed gene expression profiling analysis using data obtained from RNA-seq of UKE1 cells following treatment with MCL and/or ruxolitinib.

TRIM59 promotes gliomagenesis by inhibiting TC45 dephosphorylation of STAT3

Project description:Although the oncogenic signalings driven by amplification and mutations of EGF receptor (EGFR) gene play a major role in glioblastoma pathogenesis, the responsible downstream mechanisms remain less clear. Here we demonstrate that tripartite motif-containing protein 59 (TRIM59), acting as a new downstream effector of EGFR signaling, regulates STAT3 activation in glioblastoma. EGFR signaling leads to TRIM59 upregulation through SOX9 that results in enhancing TRIM59 interaction with STAT3 in nucleus, and inhibiting STAT3 association with TC45 (the nuclear form of T cell protein tyrosine phosphatase TC-PTP), thereby maintaining STAT3 phosphorylation and activation and promoting tumorigenesis. Silencing TRIM59 suppresses cell proliferation, migration, and orthotopic xenograft brain tumorigenesis of GBM cells. Moreover, evaluation of GBM patient samples reveals an association among EGFR activation, TRIM59 expression, STAT3 phosphorylation, and poor prognoses. Our study identifies TRIM59 as a new regulator of oncogenic EGFR-STAT3 signaling and a potential therapeutic target for GBM patients with EGFR activation.

Project description:Using an unbiased RNA sequencing approach of primary CD4+ T cells infected with three wild type primary HIV-1 isolates and selective mutants thereof, Langer et al. show that the HIV-1 accessory protein Vpu exerts broad immunosuppressive effects, inhibiting the induction of innate and adaptive immune responses. Transcription factor network analyses reveal that Vpu suppresses the expression of NF‑κB rather than IRF3 target genes. As a result, Vpu impairs the production of pro-inflammatory cytokines such as IFNβ, CXCL10 and IL-6.

Quantitative proteomics reveals the protective effects of Huangqi decoction against intrahepatic cholestasis by inhibiting NF-κB/IL-6/STAT3 signaling pathwa

Project description:Intrahepatic cholestasis (IC) is a common symptom of liver diseases but with limited treatment. Huangqi decoction (HQD), a classic herbal medicine, has shown protective effects against IC. In this study, the iTRAQ-based quantitative proteomics was performed to investigate the potential mechanism of HQD on alpha-naphthylisothiocyanate (ANIT) induced IC, resulting in 2,796 quantified proteins across all the samples, including 270 differentially expressed proteins under HQD treatment. Fuzzy c-means (FCM) clustering analysis of these 270 proteins revealed that the pro-inflammatory proteins, such as LCN2, SAA1, FGG, FGA, and FGB, were assigned in the Cluster 1 (up-regulated by ANIT, and down-regulated by HQD). Following functional bioinformatics analysis and protein-protein interaction (PPI) network analysis represented that these pro-inflammatory proteins were involved in the STAT3 signaling pathway. Further real-time PCR and Western blot experiments confirmed that the expression of these proteins was consistent with the proteomic results. Moreover, HQD treatment decreased the phosphorylation of STAT3 that induced by ANIT. And Western blot experiments also revealed that HQD could decrease phosphorylation of NF-κB and down-regulate the expression inflammatory genes IL-6, and therefore inhibit IL-6/STATA3 signaling pathway. In summary, the present study suggested that HQD treatment may ameliorate intrahepatic cholestasis via inhibiting the NF-κB/IL-6/STAT3 signaling pathway.

PekUSZ_Development of Lysine Crotonyl-mimic Probe to Covalently Identify H3K27Cr Interacting Proteins

Project description:we developed a novel H3g27Cr peptide probe mimicking Histone lysine crotonylation, which could more effectively recognize Histone Kcr-interacting proteins through covalent proximity-induced bioconjugation. We initially validated the orthogonality and cysteine reactivity of crotonyl-like group through protein-ligand binding models of PDZΔRGS3 and PLCγ1-c SH2, and further showed the feasibility of H3g27Cr to covalently bind the known Kcr “eraser” HDAC1. With the H3g27Cr probe in hands, we successfully identified and validated one unknown H3K27Cr-interacting protein, the transcriptional activator STAT3, from AP-MS experiment in A549 cell lysates. The candidate protein STAT3 was found that directly bound to the Histone H3K27Ac/Cr in XL-MS and pull-down assays, and this interaction might reduce the H3K27Ac/Cr levels.
