Proteomic analysis of sheep primary testicular cells infected with bluetongue virus.

Project description:Bluetongue virus (BTV) causes a non-contagious, arthropod-transmitted disease in wild and domestic ruminants, such as sheep. In this study, we used iTRAQ labeling coupled with LC-MS/MS for quantitative identification of differentially expressed proteins in BTV-infected sheep testicular (ST) cells. Relative quantitative data were obtained for 4455 proteins in BTV- and mock-infected ST cells, among which 101 and 479 proteins were differentially expressed at 24 and 48 h post-infection, respectively, indicating further proteomic changes during the later stages of infection. Ten corresponding genes of differentially expressed proteins were validated via real-time RT-PCR. Expression levels of three representative proteins, eIF4a1, STAT1 and HSP27, were further confirmed via western blot analysis. Bioinformatics analysis disclosed that the differentially expressed proteins are primarily involved in biological processes related to innate immune response, signal transduction, nucleocytoplasmic transport, transcription and apoptosis. Several upregulated proteins were associated with the RIG-I-like receptor signaling pathway and endocytosis. To our knowledge, this study represents the first attempt to investigate proteome-wide dysregulation in BTV-infected cells with the aid of quantitative proteomics. Our collective results not only enhance understanding of the host response to BTV infection but also highlight multiple potential targets for the development of antiviral agents.

Project description:The gene expression in conventional and plasmacytoid dendritic cells (cDC and pDC respectively) during Bluetongue virus (BTV) infection in sheep depends on the lymphoid compartment and suggest that these cell types have a role in the physiopathology We analyzed the gene expression in lymph node pDC and cDC from 3 control and 3 BTV infected sheep at day 6 post infection, in blood pDC from 2 control and 2 BTV infected sheep at day 6 post infection, and in spleen cDC from 3 control and 2 BTV infected sheep at day 6 post infection.

Project description:Bluetongue virus causes infections in wild and domesticated ruminants, with the potential for causing significant morbidity, mortality and economic harm in both the developing and developed world. While vaccines have been developed, no treatment for acute infections exists. In this regard, a possible approach is to determine host-cell factors utilised by Bluetongue virus. We thus proceeded to characterize the phosphoproteome of BTV infected HeLa cells to elucidate the intracellular signalling pathways and identify critical host factors activated during Bluetongue virus infection.

Transcriptome profiling of Hu sheep testicular development associated with dietary energy restriction and during compensatory compensation

Project description:To investigate the effect of energy levels on sheep testicular development, we systematically investigated mRNAs using high-throughput sequencing at two dietary energy levels of Hu sheep (maintenance energy and 55% energy levels) chrysophanol alone treatment and combination treatment as well as revealed that the combination treatment the most defence-related genes induced expression changes. We identified numerous differentially expressed genes that exhibited distinct expression patterns, and defence-related genes that were highly differentiated between the physcion and chrysophanol alone treatment and combination treatment. These genes have known or potential roles in defense response and were enriched in functional gene categories potentially responsible for cucumber resistance. Therefore, they are appealing candidates for further investigation of the gene expression and associated regulatory mechanisms related to defense response.
