Project description:we reproduced the clonal plants of Fix (Fixation of rice heterozygosity) to T4 generation with 1.9~3.2% clonal seeds rate of Fix-T01th, T12th, T23th and T34th. The descendant clonal plants Fix-T0, T1, T2, T3 and T4 exhibited no discernible differences on plant morphology, clonal seeds rate (3.7~4.3%), genome, methylome and transcriptome. Unexpectedly, few aneuploids were identified in each generation. The visible morphology changes of aneuploids might be caused by chromosome segments elimination. Together, the synthetic apomixis was heritable through multiple generations and few aneuploids were induced when using the synthetic apomixis system.

Project description:Previous studies have shown that exposure to ionising radiation (IR) results in DNA methylation changes in plants. However, to the best of our knowledge, this phenomenon is yet to be studied across multiple generations. Furthermore, the exact role of these changes in the IR-induced plant response is still far from understood. Here, we study the effect of gamma radiation on DNA methylation and its effect across generations in young Arabidopsis plant. A multigenerational set-up was used in which three generations of seven-day old Arabidopsis thaliana plants were exposed to either of the different radiation treatments (30 mGy h-1, 60 mGy h-1, 110 mGy h-1, or 430 mGy h-1) or to natural background radiation (control condition) for 14 days. The parental generation consisted of previously non-exposed plants, whereas generation 1 and generation 2 plants had already received a similar irradiation in the previous one or two generations, respectively. Directly after exposure the entire methylomes were analysed with UPLC-MS to measure whole genome methylation levels. Whole genome bisulphite sequencing (WGBS) was used to identify differentially methylated regions (DMRs), including their methylation context in the three generations in three different radiation conditions (control, 30 mGy/h, and 110 mGy/h). Both intra- and intergenerational comparisons of the genes and transposable elements (TEs) associated with the DMRs were made across the gamma radiation exposure conditions. It was apparent that there was a higher number of changes in CpG methylation, whereas only limited changes in CHG methylation occurred and no changes in CHH methylation were observed. A clear increase in IR-induced DMRs was seen over the three generations that were exposed to the lowest dose rate, where Gen 2 had a markedly higher number of DMRs than the previous two generations (Parent and Gen 1). Counterintuitively, we did not see significant differences in the plants exposed to the higher dose rate. In addition, a large number of DMRs associated with TEs were found, the majority of them being hyper methylated, likely leading to more genetic stability. Next to that, a significant number of DMRs were associated with genes (either in their promoter associated region or gene body). A functional analysis of these genes (not included in with this data) showed an enrichment for genes related to development as well as various stress responses, including DNA repair, RNA splicing, and (a)biotic stress responses. These observations indicate a role of DNA methylation in the regulation of these genes in response to IR exposure and shows a possible role for epigenetics in adaptation to IR in plants.

The key biological features of apomixis are the failure of meiosis (i.e., apomeiosis), the differentiation of unreduced embryo sacs and eggs, and their autonomous development in functional embryos through parthenogenesis and the formation of viable endosperm either via fertilization-independent means or following fertilization with a sperm cell (Koltunow and Grossniklaus 2003). Recently gained biological information has shown that H. perforatum is an attractive model system for the study of a naturally occurring form of asexual reproduction called aposporous apomixis, This research describes a global gene expression analysis of H. perforatum ovaries collected from sexual and aposporous plant accessions for the purpose of identifying genes and processes potentially associated with aposporous apomixis in this model species.

Project description:Unstable couple relationships and high rates of repartnering have increased the share of U.S. families with stepkin. Yet data on stepfamily structure are from earlier periods, include only coresident stepkin, or cover only older adults. In this study, we use new data on family structure and transfers in the Panel Study of Income Dynamics (PSID) to describe the prevalence and numbers of stepparents and stepchildren for adults of all ages and to characterize the relationship between having stepkin and transfers of time and money between generations, regardless of whether the kin live together. We find that having stepparents and stepchildren is very common among U.S. households, especially younger households. Furthermore, stepkin substantially increase the typical household's family size; stepparents and stepchildren increase a household's number of parents and adult children by nearly 40 % for married/cohabiting couples with living parents and children. However, having stepkin is associated with fewer transfers, particularly time transfers between married women and their stepparents and stepchildren. The increase in the number of family members due to stepkin is insufficient to compensate for the lower likelihood of transfers in stepfamilies. Our findings suggest that recent cohorts with more stepkin may give less time assistance to adult children and receive less time assistance from children in old age than prior generations.

Project description:Aflatoxins are carcinogenic fungal secondary metabolites. Levels of aflatoxins in agricultural commodities are stringently regulated by many countries. A cluster of genes is responsible for aflatoxin biosynthesis by Aspergillus flavus and other closely related species. Expression of the clustered aflatoxin genes is governed by a complex network of regulatory mechanisms. To better understand the molecular events that are associated with aflatoxin production, transcription profiling by microarray analyses which compared three independent aflatoxigenic A. flavus strains to individual isogenic progenies that no longer produced aflatoxins after serial transfers was carried out. Twenty-two significantly differentially expressed features were identified. After physical mapping using the A. oryzae genome sequence as the reference, the number of unique genes was reduced to 16. Compared to the parental strains, changes in the aflatoxin gene expression levels in the progenies were not significant, which suggests that the inability to produce aflatoxins is not caused by decreased expression. The only gene showing higher expression levels in the progenies is homologous to glutathione S-transferease genes. Overexpression of this gene, named hcc, at six- to nine-fold in an aflatoxigenic A. flavus did not cause discernible changes in colony morphology or aflatoxin production. Loss of aflatoxin production after serial transfers may not result from a single event but caused by multiple factors. Strains 459B-20-2, 474A-20, and 499A-20 were nonaflatoxigenic isolates obtained after 20 serial transfers of the parental strains on potato dextrose agar slants (Horn and Dorner 2002). Comparsions in each experiment consisted of one aflatoxigenic parental strain and one nonaflatoxigenic progeny, compared after 48- or 72-hr growth. Each comparison was repeated with duplicate dye-flip.

The synthetic libraries are variants of the glucocorticoid receptor binding sites (GBS). The "GBS half site" library contains 8 consecutive randomized nucleotides within the core binding sites, while the "Cgt/Sgk library" contains 5 consecutive randomized nucleotides on the flank of the GBS.
