Human engineered skeletal muscle of hypaxial origin from pluripotent stem cells with advanced function and regenerative capacity

Project description:Human pluripotent stem cell derived muscle models show great potential for translational research. Here, we describe novel developmentally inspired methods for the derivation of skeletal muscle cells and their utility in three-dimensional skeletal muscle organoid formation as well as skeletal muscle tissue engineering. Three-dimensional organoid and tissue engineered models exhibit organotypic maturation and function and regenerative responses, recapitulating canonical properties of bona fide skeletal muscle in vivo. Key steps include the directed differentiation of human pluripotent stem cells to embryonic muscle progenitors of hypaxial origin followed by primary and secondary fetal myogenesis with development of a satellite cell pool and evidence for innervation in vitro. Regenerative competency was demonstrated in a cardiotoxin injury model with evidence for satellite cell activation as underlying mechanism.

Project description:The in vitro directed differentiation of pluripotent stem cells (PSCs) through stimulation of developmental signaling pathways can generate mature somatic cell types for basic laboratory studies or regenerative therapies. We used microarrays to detail the global transcriptomes of mouse embryonic stem cells differentiated in vitro into putative thyroid vs lung epithelial lineages using serum-free media supplemented with either BMP4+FGF2 (thyroid media) or BMP4+Wnt3a (lung media.) Differentiated cells carried an Nkx2-1mCherry knock-in reporter to allow sorting of mCherry + vs - cells in each condition on day 14 of differentiation for global transcriptomic profiling.

Thermal stability variation during transtion to pluripotency

Project description:To investigate functional differences between pluripotent stem cells and somatic cells, we measured protein thermal stability and expression after transitions between cell types using allogenic cell lines. To this purpose, we reprogrammed hFF into human iPSCs and initiated undirected differentiation through formation of EBs during 21 days. We also included human embryonic stem cells (ESC) in order to validate our approach and colon cancer cell line RKO.
