Project description:To respond to the world around them, animals rely on the input of a network of sensory organs distributed across the body. A single individual will generally express several distinct classes of sensory organ, each bearing features that appear to enhance the detection of specific stimuli, such as strain, pressure, or light. The features that underlie this specialization relate both to the neurons housed within a sensory organ and to the various accessory cells that they are composed of. This diversity of cell types, both within and between sensory organs, raises two fundamental questions: (1) How is this diversity generated during development? And (2) What makes these cell types distinct from one another? To address these questions, we performed single-cell RNA-sequencing on a region of the pupal male Drosophila melanogaster foreleg that displays a wide variety of functionally and structurally distinct sensory organs: the first tarsal segment. Our work characterizes the cellular landscape in which the sensory organs reside; identifies a novel glia cell type that appears to contribute towards the construction of the neural lamella; resolves and validates a combinatorial transcription factor code unique to each of mechanosensory, sex comb, and campaniform sensilla neurons, as well as four distinct gustatory receptor neuron classes; matches receptors and membrane channels to specific neurons; and characterizes the transcriptomic differences between and within sensory organ support cell types. Collectively, our work identifies core genetic features of a variety of sensory organs and provides a rich, annotated resource for studying their development and function.

Adult male and female antennae (a3+arista), front legs (from distal tibia down), and proboscises were dissected at 1-2 hours after eclosion. Total RNA was extracted and the mRNA fraction was amplified by reverse transcription followed by in vitro transcription with T7 RNA polymerase.

In the crustacean Parhyale, we examine the fidelity of leg regeneration using cellular composition and cell molecular profiles. Single-nuclei sequencing shows that regenerated and uninjured legs are indistinguishable in terms of cell type composition and transcriptional profiles.

Project description:In this study, we have elucidated the DNA methylation patterns in different organs of a cultivated chickpea genotype ICC 4958 (leaf, root, flower and young pod) and leaf of wild chickpea PI 489777 using bisulphite sequencing. Approximately 108 million read pairs were analyzed per sample. The extent of methylation along-with the context and genomic location of methylated Cs was identified. Further processing was performed to identify the differentially methylated regions among samples with leaves of ICC 4958 as the reference sample. The high resolution methylome maps of different organs and differentially methylated regions will serve as reference for understanding the epigenetic regulation in chickpea.

Project description:Our ability to sense and move our bodies relies on proprioceptors, sensory neurons that detect mechanical forces within the body. Proprioceptors are diverse: different subtypes detect different features of joint kinematics, such as position, directional movement, and vibration. However, because they are located within complex and dynamic peripheral tissues, the underlying mechanisms of proprioceptor feature selectivity remain poorly understood. Here, we investigate molecular and biomechanical contributions to proprioceptor diversity in the Drosophila leg. Using single-nucleus RNA sequencing, we found that different proprioceptor subtypes express similar complements of mechanosensory and other ion channels. However, anatomical reconstruction of the proprioceptive organ and connected tendons revealed major biomechanical differences between proprioceptor subtypes. We constructed a computational model of the proprioceptors and tendons, which identified a putative biomechanical mechanism for joint angle selectivity. The model also predicted the existence of a goniotopic map of joint angle among position-tuned proprioceptors, which we confirmed using calcium imaging. Our findings suggest that biomechanical specialization is a key determinant of proprioceptor feature selectivity in Drosophila. More broadly, our discovery of proprioceptive maps in the fly leg reveals common organizational principles between proprioception and other topographically organized sensory systems.
