Project description:We aimed to explore the effect of the 3-day dry immersion - a model of physical unloading - on mitochondrial function, transcriptomic and proteomic profiles in a slow-twitch m. soleus of six healthy females. We registered that a marked reduction (25-34%) in the ADP-stimulated respiration in permeabilized muscle fibers was not accompanied by a decrease in the content of mitochondrial enzymes (mass spectrometry-based quantitative proteomics), hence it is related to the disruption in regulation of respiration. We detected a widespread change in the transcriptomic profile (RNA-seq) upon dry immersion. Down-regulated mRNAs were strongly associated with mitochondrial function, as well as with lipid metabolism, glycolysis, insulin signaling, and various transporters. Despite the substantial transcriptomic response, we found no effect on the content of highly abundant proteins (sarcomeric, mitochondrial, chaperon and extracellular matrix-related, etc.) that may be explained by long half-life of these proteins. We suggest that during short-term disuse the content of some regulatory (and usually low abundant) proteins such as cytokines, receptors, transporters, transcription regulators is largely determined by its mRNA concentration. These mRNAs revealed in our work may serve as putative targets for the future studies aimed at developing the approaches for the prevention of muscle deconditioning-induced by disuse.

Project description:This study assessed the effects of weekly disuse (dry immersion; 10 males) on the strength and aerobic performance of the ankle plantar flexors and knee extensors, mitochondrial function in permeabilized muscle fibers, and the proteomic (quantitative mass spectrometry-based analysis) and transcriptomic (RNA-sequencing) profiles of the soleus muscle and vastus lateralis muscle. Dry immersion-induced decreases in maximal voluntary contraction in both muscles were comparable and occurred without a decrease in the relative content of contractile proteins, which appears to be due to disruption of neuromuscular mechanisms rather than a decrease in muscle mass. The decrease in maximum ADP-stimulated mitochondrial respiration was also comparable in both muscles and occurred without a decrease in the relative content of mitochondrial proteins/respiratory enzymes, which appears to be associated with dysregulation of respiration. Meanwhile changes in the transcriptome were significantly more pronounced (2.5–4 times, in terms of number of differentially expressed genes) in the “slow” soleus muscle than in the vastus lateralis muscle. This suggests that longer-term disuse will lead to a change (primarily a decrease) in the relative content of a number of highly abundant proteins in Sol and an accelerated decline in the functional capacity of the ankle plantar flexors.

The result of the choice of the object of research fell on dry blood spots, since the technique of capillary blood sampling is less invasive and allows sampling with a greater frequency. This allows researchers to analyze the changes caused by dry immersion in stages and observe the unfolding of the adaptive response taking into account the time of exposure to the experimental conditions.

Project description:The dry period in cow are critical to mammary health and the success of the next lactation. The expression of proteins the dry period progresses need to be characterized to provide information on involution and immune functions during the dry period. Total protein and lactoferrin concentrations rose significantly by days 10 and 21 after cessation of lactation. Using the day 21 dry secretions we examined their ability to inhibit bacterial growth. Bacterial growth inhibition was not correlated with lactoferrin for 4 coliforms tested nor for all 7 mastitis pathogens. Label free mass spectroscopy was used to quantify changes in whey proteins from milk/dry secretions collected on days 0, 3,10, and 21 of the dry period. 776 proteins were identified. 109 proteins were upregulated at one or more times in the dry period. The most significate enriched GO functional terms for the up-regulated proteins were immune function and stress related. 68 proteins were down regulated in the dry period. The most significant enriched GO functional terms for the down-regulated proteins were stress and immune function related. 12 proteins were significantly correlated with coliform growth. The dry secretion composition changes we found provide information for understanding the dry period. The sample code for the MS raw files is as follows. The 3 technical replicates are denoted as a letter A, B and C. The number following is the cow identification number for 11 cows used. The final two-digit number after the underscore is the day sampled where _01 = day 1, _03 = day 3, _10 = day 10 and _21 = day 21 of dry period. For example, A1313_01 is technical replicate A for cow 1313 collected on day 1. B1313_03 is technical replicate B for cow 1313 collected on day 3.

Project description:Low-intensity neuromuscular electrical stimulation (NMES) is often used as an alternative to exercise and high-intensity electrical stimulation to prevent the loss of muscle mass, strength, and endurance in spaceflight and in patients with severe chronic diseases. This study assessed the efficiency of low-intensity (~10% of maximal voluntary contraction) combined (low- and high-frequency) electrical stimulation in preventing the negative effects of weekly disuse (dry immersion without [DI, see a related dataset GSE271607] and with [DI+NMES] daily stimulation; 10 males in each group) on the strength and aerobic performance of the ankle plantar flexors and knee extensors, mitochondrial function in permeabilized muscle fibers, and the proteomic (quantitative mass spectrometry-based analysis) and transcriptomic (RNA-sequencing) profiles of the soleus muscle and vastus lateralis muscle. Application of electrical stimulation during dry immersion prevented a decrease in the maximal strength and a slight reduction in aerobic performance of knee extensors, as well as a decrease in maximal ADP-stimulated mitochondrial respiration and changes in the expression of genes encoding mitochondrial, extracellular matrix, and membrane proteins in the vastus lateralis muscle. In contrast, for the ankle plantar flexors/soleus muscle, electrical stimulation had a positive effect only on maximal mitochondrial respiration, but accelerated the decline in the maximal strength and muscle fiber cross-sectional area, which appears to be associated with the activation of genes regulating the inflammatory response. The data obtained open up broad prospects for the use of low-intensity combined electrical stimulation to prevent the negative effects of disuse for “mixed” muscles, meanwhile, the optimization of the stimulation protocol is required for “slow” muscles.

Project description:Tears are a biological fluid that has diagnostic potential for ocular diseases. Extracellular vesicles (EVs), wildly detected in various biofluids including tears, are nanoparticles released by living cells and considered as promising detection sources for non-invasive liquid biopsy. Understanding the roles of tears and tear-EVs in ocular diseases such as dry eye can facilitate the studies of clinical diagnosis, which usually entails detecting such liquid objects with a rapid and effective method. In this study, we utilized a mass spectrometry based strategy to analyze peptidome/proteome profiles of tear and EVs for rapid dry eye diagnosis. Nano-sized EVs were isolated from tears of either healthy control (HC) individuals or dry eye syndrome (DES) patients, and the tear compositions were further analyzed by tracking their fingerprints with matrix-assisted laser desorption ionization/time-of-flight mass spectrometry (MALDI-TOF-MS). The fingerprints of tear-EVs could be observed in a dose-dependent manner as well as tears, allowing comparing their discriminant peaks between tears and EVs. By analyzing these peaks, the fingerprints of both tear and tear-EVs were showed to have a capability of distinguishing DES patients from HC donors, and providing an efficient way for screening potential DES biomarkers. The proposed tear and EV fingerprinting approach is expected to be a potential tool in rapid diagnosis of ocular disease and in-depth researches of pathogenesis.
