Langdon or T. turgidum ssp. carthlicum) and diploid wheat progenitor Aegilops tauschii (KU2025). Project description:We have employed whole genome microarray expression profiling as a discovery platform to identify genes to alter the transcript accumulation levels in hybrid necrosis-showing plants, which are triploid hybrids crossed between tetraploid wheat and diploid wheat progenitor Aegilops tauschii. Of the up-regulated genes, defense-related genes were most frequently found, whereas photosythesis-related genes down-regulated in the type I necrosis line. To validate the microarray data, RT-PCR and quantitative RT-PCR analyses for 22 selected genes were performed. Of the examined 15 up-regulated and 7 down-regulated genes, the expression pattern of only one down-regulated gene was inconsistent with the microarray data. Together with cytological analysis of the necrotic tissues, the microarray analysis strongly suggests that an autoimmune response might be triggered by intergenomic incompatibility between the AB and D genomes in type I necrosis, and that genetically programmed cell death could be regarded as a hypersensitive response-like cell death similar to that observed in Arabidopsis intraspecific and Nicotiana interspecific hybrids. Expression patterns were compared between the two synthetic hexaploid lines showing the wild-type phenotype (as a reference) and hybrid necrosis. Total RNA samples were isolated from the 3-week-old seedling leaves. Two independent experiments were conducted.

Project description:Modern wheat cultivars are increasingly sensitive to ground level ozone, with 7-10% mean yield reductions in the northern hemisphere. In this study, three of the genome donors of bread wheat, Triticum urartu (AA), T. dicoccoides (AABB), and Aegilops tauschii (DD) along with a modern wheat cultivar (T. aestivum 'Skyfall'), a 1970s cultivar (T. aestivum 'Maris Dove'), and a line of primary Synthetic Hexaploid Wheat were grown in 6 L pots of sandy loam soil in solardomes (Bangor, North Wales) and exposed to low (30 ppb), medium (55 ppb), and high (110 ppb) levels of ozone over 3 months. Measurements were made at harvest of shoot biomass and grain yield. Ae. tauschii appeared ozone tolerant with no significant effects of ozone on shoot biomass, seed head biomass, or 1000 grain + husk weight even under high ozone levels. In comparison, T. urartu had a significant reduction in 1000 grain + husk weight, especially under high ozone (-26%). The older cultivar, 'Maris Dove', had a significant reduction in seed head biomass (-9%) and 1000 grain weight (-11%) but was less sensitive than the more recent cultivar 'Skyfall', which had a highly significant reduction in its seed head biomass (-21%) and 1000 grain weight (-27%) under high ozone. Notably, the line of primary Synthetic Hexaploid Wheat was ozone tolerant, with no effect on total seed head biomass (-1%) and only a 5% reduction in 1000 grain weight under high ozone levels. The potential use of synthetic wheat in breeding ozone tolerant wheat is discussed.

Study aims to understand regulation of gene expression in synthetic and natural wheat allohexaploids (Triticum aestivum), that combines the AB genome of T. turgidum and the D genome of Aegilops tauschii; and which we have recently characterized as genetically stable. We conducted a comprehensive genome-wide analysis of gene expression that allowed us to compare the effect of variability of the D genome progenitor, the trans-generation stability as well as comparison to natural wheat allohexaploid. We used the Affymetrix GeneChip® Wheat Genome Array, on which 55,049 transcripts are represented. Additive expression was shown to represent majority of expression regulation in the synthetic allohexaploids, where expression for more than 93% of transcripts was equal to the one evaluated from equal mixture of parental RNA. This leaves ~2000 (~7%) transcripts, which expression was non-additive. No global gene expression bias or dominance towards any of the progenitor genomes was observed whereas high trans-generational stability and low effect of the D genome progenitor variability were revealed. Our study suggests that gene expression regulation in wheat allohexaploids is early established upon allohexaploidization and highly conserved over generations, as demonstrated by the high similarity of expression with natural wheat allohexaploids. Of the up-regulated genes, defense-related genes were most frequently found in the leaves and shoot apical meristem (SAM) of type II necrosis line with low temperature. On the other hand, cell cycle-related genes were highly down-regulated in SAM of type II necrosis line under low temperature. To validate the microarray data, RT-PCR and quantitative RT-PCR analyses for 37selected genes were performed in total. Of the examined 21 up-regulated and 16 down-regulated genes, more than 75% of expression pattern were consistent with the microarray data. Together with cytological analysis of the necrotic tissues, the microarray analysis strongly suggests that an autoimmune response-like reaction and repression of cell division might be triggered by intergenomic incompatibility between the AB and D genomes in type II necrosis. Expression patterns were compared between the two synthetic hexaploid lines showing the wild-type phenotype (as a reference) and cold-induced hybrid necrosis. Total RNA samples were isolated from the 3-week-old seedling leaves. In addition, total RNA were also extracted from the seedling leaves and shoot meristem with 12- and 6-weeks of low temperature, respectively. Two independent experiments were conducted in each exprement.

Project description:As the diploid progenitor of common wheat, Aegilops tauschii Cosson (DD, 2n = 2x = 14) is considered to be a promising genetic resource for the improvement of common wheat. In this work, we demonstrated that the efficiency of transferring A. tauschii segments to common wheat was clearly improved through the use of synthetic octaploid wheat (AABBDDDD, 2n = 8x = 56) as a "bridge." The synthetic octaploid was obtained by chromosome doubling of hybrid F1 (A. tauschii T015 × common wheat Zhoumai 18). A set of introgression lines (BC1F8) containing 6016 A. tauschii segments was developed and displayed significant phenotype variance among lines. Twelve agronomic traits, including growth duration, panicle traits, grain traits, and plant height (PH), were evaluated. And transgressive segregation was identified in partial lines. Additionally, better agronomic traits could be observed in some lines, compared to the recurrent parent Zhoumai 18. To verify that the significant variance of those agronomic traits was supposedly controlled by A. tauschii segments, 14 quantitative trait loci (QTLs) for three important agronomic traits (thousand kernel weight, spike length, and PH) were further located in the two environments (Huixian and Zhongmou), indicating the introgression of favorable alleles from A. tauschii into common wheat. This study provides an ameliorated strategy to improve common wheat utilizing a single A. tauschii genome.

Project description:Synthetic hexaploid wheat (SHW) can serve as a bridge for the transfer of useful genes from Aegilops tauschii and tetraploid wheat (Triticum turgidum) into common wheat (T. aestivum). The objective of this study was to evaluate 149 SHW lines and their 74 tetraploid parents for their genetic diversity, breeding values and inter-genomic interactions for resistance to Fusarium head blight (FHB). The genetic diversity analysis was performed based on the population structure established using 4,674 and 3,330 polymorphic SNP markers among the SHW lines and tetraploid parents, respectively. The results showed that all T. carthlicum and most T. dicoccum accessions formed different clusters and subpopulations, respectively, whereas all the T. durum, T. polonicum, T. turgidum, and T. turanicum accessions were clustered together, suggesting that T. durum was more closely related to T. polonicum, T. turgidum, and T. turanicum than to T. dicoccum. The genetic diversity of the SHW lines mainly reflected that of the tetraploid parents. The SHW lines and their tetraploid parents were evaluated for reactions to FHB in two greenhouse seasons and at two field nurseries for 2 years. As expected, most of the SHW lines were more resistant than their tetraploid parents in all environments. The FHB severities of the SHW lines varied greatly depending on the Ae. tauschii and tetraploid genotypes involved. Most of the SHW lines with a high level of FHB resistance were generally derived from the tetraploid accessions with a high level of FHB resistance. Among the 149 SHW lines, 140 were developed by using three Ae. tauschii accessions CIae 26, PI 268210, and RL 5286. These SHW lines showed FHB severities reduced by 21.7%, 17.3%, and 11.5%, respectively, with an average reduction of 18.3%, as compared to the tetraploid parents, suggesting that the D genome may play a major role in reducing disease severity in the SHW lines. Thirteen SHW lines consistently showed a high level of FHB resistance compared to the resistant check, Sumai 3, in each environment. These SHW lines will be useful for the development of FHB-resistant wheat germplasm and populations for discovery of novel FHB resistance genes.
