Project description:We report the use of transcriptome sequencing in the intervention of luteolin or quercetin in adipogenesis. We constructed the adipogenic differentiation model of mouse embryonic fibroblasts and obtained the differential genes of luteolin or quercetin regulating adipocyte differentiation by transcriptomic sequencing. We identified 2085 differentially expressed lipogenesis response genes, 181 luteolin response genes and 574 quercetin response genes. This study provides a framework for the network regulation mechanism of luteolin or quercetin inhibiting preadipocyte differentiation.

Project description:We reported the application of transcriptome sequencing in high-throughput analysis of metformin intervention in adipogenesis. By obtaining differentially expressed genes from transcriptome, we constructed the mRNA expression profile in mouse embryonic fibroblast adipose differentiation model. We found that metformin can effectively distinguish expressed, ready to express or stably inhibited genes in the process of adipose differentiation, which reflects the cell state and lineage potential. Using an optimized data analysis workflow, we identified 5362 different expression adipogenesis responsive genes and 2539 metformin responsive genes. This study provided a framework for the application of comprehensive transcriptome analysis to the network regulation mechanism of metformin inhibiting preadipocyte differentiation

Project description:To determine if any altered miRNA expression is actually involved in the growth inhibition and/or cell death induction by luteolin and/or gefitinib, we performed miR-array analysis using RNA from PC-3 cells after 24h of treatment with 60Î¼M luteolin and/or 60Î¼M gefitinib. PC-3 cells were treated for 24h with DMSO, 60Î¼M luteolin (Lut), 60Î¼M gefitinib (Gef) or their co-administration (Lut+Gef). Each sample was run in duplicate.

Luteolin exerts anti-tumor immunity in hepatocellular carcinoma by accelerating CD8+ T lymphocyte infiltration

Project description:We observed that luteolin effectively restrained the growth of solid tumors in the established mouse model of HCC. Expression spectrum chip sequencing showed that treatment with luteolin could promote T cell activation, cell chemotaxis and cytokine production. Furthermore, we explored the influence of luteoin on the phenotypic and functional alterations of T cells. Our findings revealed that luteolin maintained a high ratio of CD8+ T cells in the peripheral blood, spleen, and tumor tissues. Additionally, luteolin restored the cytotoxicity of tumor-infiltrating CD8+ T cells in H22 tumor-bearing mice. Moreover, CD8+ T cells showed stronger activation phenotypes and more production of IFN-γ, TNFα, and Granzyme B in serum. Furthermore, the co-administration of luteolin and PD-1 mAb resulted in potentiated anti-tumor effects in H22 tumor-bearing mice.These findings indicate that luteolin could exert an anti-tumor immune response by inducing CD8+ T cell infiltration and enhance the anti-tumor effects of PD-1 mAb in HCC treatment in H22 tumor-bearing mice.
