Project description:During the over 300 million years of co-evolution between herbivorous insects and their host plants, a dynamic equilibrium of evolutionary arms race has been established. However, the co-adaptation between insects and their host plants is a complex process, often driven by multiple evolutionary mechanisms. We found that various lepidopteran pests that use maize as a host exhibit differential adaptation to the plant secondary metabolites, benzoxazinoids (BXs). Notably, the Spodoptera genus, including Spodoptera frugiperda (fall armyworm) and Spodoptera litura (cotton leafworm), demonstrate greater tolerance to BXs compared to other insects. Through comparative transcriptomic analysis of the midgut, we identified four candidate genes potentially involved in BXs detoxification in S. frugiperda. Subsequently, we confirmed two UGT genes, Sfru33T10 and Sfru33F32, as key players in BXs detoxification using CRISPR/Cas9 gene-editing technology. Phylogenetic analysis revealed that Sfru33T10 evolved independently within the Noctuidae family and is involved in the glycosylation of HDMBOA, while Sfru33F32 evolved independently within the Spodoptera genus and functions as a key detoxification enzyme responsible for the glycosylation of both DIMBOA and HMBOA. Our study demonstrates that the UGT gene family plays a crucial role in the adaptation of noctuid insects to maize, with multiple independent evolutionary events within the Noctuidae family and the Spodoptera genus contributing significantly to host adaptation.

Project description:Insect gut microbiota plays important roles in acquiring nutrition, preventing pathogens infection, immune responses, and communicating with the environment. Gut microbiota can be affected by some external factors such as foods, temperature, and antibiotics. Spodoptera frugiperda (Lepidoptera: Noctuidae) is an important destructive pest of grain crops all over the world. The function of gut microbiota in S. frugiperda remains to be investigated. In this study, we fed the S. frugiperda with the antibiotic mixture (penicillin, gentamicin, rifampicin, and streptomycin) to perturb the gut microbiota, and further examined the effect of dysbiosis in gut microbiota on the gene expression of S. frugiperda by RNA sequencing. We found the composition and diversity of the gut bacterial community were changed in S. frugiperda after antibiotics treatmen, and the expression of genes related to energy and metabolic process were affected after antibiotics exposure in S. frugiperda. Our work will help understand the role of gut microbiota in insects.

Project description:We wished to investigate if adaptation to host-plant diet is the basis of differentiation for two strains of Spodoptera frugiperda (Lepidoptera:Noctuidae). We performed reciprocal transplant experiments in laboratory conditions, feeding each strain (sf-C and sf-R) with artificial diet, corn plants or rice plants. RNA-Seq was performed on pooled 4th instar larvae from this experiment. We compared this transcriptional response with that of individual 4th instar larvae collected in corn and grass fields in Florida.

Project description:Fall armyworm (FAW; Spodoptera frugiperda), an exotic moth which recently invaded Africa, is a highly destructive pest of cereals especially maize a highly valued staple crop in Nigeria. The use of natural enemies such as predators or parasitoids for FAW control is more economically viable and environmentally safer than currently recommended synthetic insecticides. Natural enemies to combat the pest have not yet been reported in Nigeria. An exploration for the pests' natural enemies was undertaken by collecting FAW eggs and larvae from maize fields. These were reared in the laboratory for emergence, identification and efficacy as natural enemies. This yielded Euplectrus laphygmae (Hymenoptera: Eulophidae); Telenomus remus (Hymenoptera: Platygastridae) and Trombidium sp. (Acari.: Trombidiidae). Cotesia or Apanteles spp. were inferred to occur since Stictopisthus sp. (Hym.: Ichneumonidae), a secondary parasitoid, that attacks cocoons of Microgasterinae (e.g. Cotesia, Apanteles etc.) also emerged. Species of yet-to-be identified predators were also observed in various niches of maize plants. A positive relationship was found between FAW instar and the number of E. laphygmae eggs/instar ranging, on average, from 1.5 on second instar to 5.5 on fourth instars hosts. Parasitism rate of T. remus on FAW eggs was 100%. Parasitic mite infestation resulted in increasing paleness, reduced feeding, growth and movement as well as death of FAW 1st instars. Thus, the occurrence of FAW natural enemies in Nigeria calls for advocacy campaign to incorporate their use into integrated pest management strategies that attract and allow natural enemies to thrive for FAW management.

Project description:The aim of this experiment was to compare the transciptome of the fall armyworm (Spodoptera frugiperda) strain SUS (a laboratory insecticide-susceptible standard) with organophosphate (OP) and pyrethroid (PYR) resistant strains were collected in cornfields located in Minas Gerais and Mato Grosso States, Brazil, in 2008 and maintained in the laboratory under selection with chlorpyrifos (at increasing discriminating doses from 100 up to 400 µg of insecticide/g of insect in a topical bioassay) or lambda-cyhalothrin (from 8.4 up to 27 µg of insecticide/g of insect) respectively The custom microarray used in this study was designed using the Agilent eArray platform (Agilent Technologies). A SurePrint HD (8×15k) expression array was designed using the base composition and the best probe methodologies to design sense orientation 60-mer probes with a 3′ bias. The FAW EST database (SPODOBASE) was used as the reference transcriptome (Negre, Hotelier et al. 2006). These sequences are derived from 8 cDNA libraries as follows: Sf1F: Fat body, Sf1H: Hemocyte, Sf1M: Midgut, Sf1P: Pool of various tissues, Sf2H: Immune Challenged hemocytes, Sf2L: Sf21 Cell lines sequences, Sf2M: Xenobiotic Induced Midgut and Sf9L: Sf9 cell lines sequences. All assembled contigs and singlets were kindly sent by the website maintainers. The BLAST2GO software v.2.3.1 (http://www.blast2go.org) was used to annotate the EST database. 60-mer probes were designed for all 7,552 assembled contigs and 5,519 annotated singlets (BlastX), totaling 13,071 sequences. For contigs encoding detoxification enzymes (P450s, GSTs and CEs) three probes were designed. Additional probe groups for 15 control genes were also included. Project description:Purpose: We aimed to use RNA-Seq and de novo transcriptome assembly to produce transcriptomes and analyze the changes in transcription regulation associated with cold and heat treatment in Spodoptera frugiperda. A detailed differential expression analysis revealed a number of candidate genes that may be associated with FAW cold and heat tolerance. To validate the RNA-seq results, we used qRT-PCR. We hoped to provide a foundation for the adaptive mechanism as well as a rich resource for the discovery and detection of novel genes involved in cold and heat stress responses of Spodoptera frugiperda. Methods: Spodoptera frugiperda was reared at lab condition, and incubated at 2 different temperature( 4 and 40°C) for 16 hours as temperature treatment groups. RNA was extracted using Trizol reagent and Illumina sequencing was performed at Macrogen. Illumina short reads were quality-filtered and Illumina-based de novo transcriptome assembly was performed. Differential Gene Expression Analysis was studied for different tempearture conditions. Results: A total of 211,967 unigenes were collected, at least 14,338 of which were annotated with gene descriptions, gene ontology terms, and metabolic pathways. There were 50 GO functional sub-groups and 21 EggNOG words as a result. DEGs with log2FC≥2 were identified and compared at various temperatures. In comparison to the 25°C treated group, we discovered 199 and 1248 individual DEGs co-regulated at 4 and 40°C, respectively. Comparing transcriptome profiles for differential gene expression revealed a number of DE genes, including cytochrome P450, odorant binding proteins (OBP), and immune system genes previously implicated in cold and high temperature resistance. The enrichment pathways were identified using KEGG analysis, and heatmaps of similar unigenes from both treatment groups (T4 and T40) were plotted. We used qPCR to confirm the RNA-seq data on 10 up- and down-regulated DEGs. Conclusions: In conclusion, we used RNA-Seq technology focused on high-throughput sequencing to compare the transcriptomes of Spodoptera frugiperda under high- and low-temperature stresses. This research was the first to identify a large number of genes that were significantly up-regulated at high and low temperatures. Many genes were discovered through comparative transcriptome analysis, and a significant number of improvements in metabolic pathways were discovered through GO and KEGG enrichment analysis. Our findings will help future molecular research and genomic studies. These newly found genes may be important and necessary to FAW harsh environment tolerance and its behavior for adaptation in new environment as well as quarantine area.

Project description:The fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), spread rapidly in Africa and Asia recently, causing huge economic losses in crop production. Fall armyworm caterpillars were first detected in South Korea and Japan in June 2019. Here, the migration timing and path for FAW into the countries were estimated by a trajectory simulation approach implementing the insect's flight behavior. The result showed that FAWs found in both South Korea and Japan were estimated to have come from eastern China by crossing the Yellow Sea or the East China Sea in 10-36 h in three series of migrations. In the first series, FAW moths that arrived on Jeju Island during 22-24 May were estimated to be from Zhejiang, Anhui and Fujian Provinces after 1-2 nights' flights. In the second series, it was estimated that FAW moths landed in southern Korea and Kyushu region of Japan simultaneously or successively during 5-9 June, and these moths mostly came from Guangdong and Fujian Provinces. The FAW moths in the third series were estimated to have immigrated from Taiwan Province onto Okinawa Islands during 19-24 June. During these migrations, southwesterly low-level jets extending from eastern China to southern Korea and/or Japan were observed in the northwestern periphery of the western Pacific Subtropical High. These results, for the first time, suggested that the overseas FAW immigrants invading Korea and Japan came from eastern and southern China. This study is helpful for future monitoring, early warning and the source control of this pest in the two countries.
