Alteration of Community Metabolism by Prebiotics and Medicinal Herbs.

Project description:Several studies have examined the impact of prebiotics on gut microbiota and associated changes in host physiology. Here, we used the in vitro cultivation of human fecal samples stimulated with a series of chemically related prebiotics and medicinal herbs commonly used in Ayurvedic medicine, followed by 16S rRNA sequencing. We applied a genome-wide metabolic reconstruction of enumerated communities to compare and contrast the structural and functional impact of prebiotics and medicinal herbs. In doings so, we examined the relationships between discrete variations in sugar composition and sugar linkages associated with each prebiotic to drive changes in microbiota composition. The restructuring of microbial communities with glycan substrates alters community metabolism and its potential impact on host physiology. We analyzed sugar fermentation pathways and products predicted to be formed and prebiotic-induced changes in vitamin and amino acid biosynthesis and degradation. These results highlight the utility of combining a genome-wide metabolic reconstruction methodology with 16S rRNA sequence-based community profiles to provide insights pertaining to community metabolism. This process also provides a rational means for prioritizing in vivo analysis of prebiotics and medicinal herbs in vivo to test hypotheses related to therapeutic potential in specific diseases of interest.

Mothers were given or not during gestation a diet enriched in the prebiotics GOS and inulin. Eight to ten weeks old male offspring were treated or not with 3 cycles of DSS in drinking water, one cycle consisting of 2 days with DSS and 5 days without DSS.

Dietary soyasaponin and prebiotics modulate intestinal health and function in the cleaner fish Ballan wrasse (Labrus bergylta)

Project description:This paper presents data from a 5-week feeding trial in which Ballan wrasse was fed either a reference diet, or the identical diet supplemented with i) the antinutrient soya saponin (0.7%) ii) a commercial prebiotic (Aquate™ SG, 0.4%) or iii) a combination of soya saponin and prebiotics. Blood, tissue and gut content from four separate intestinal segments were sampled from 6 fish per duplicate tank. Gut health and digestive functions were evaluated by various endpoint measurements employing biochemical and histomorphological tools as well as global gene expression profiling. No significant differences in fish growth were seen between the four dietary groups. Saponin supplementation, both alone and in combination with prebiotics, increased weight indices of two mid gut segments (IN2 and IN3) and decreased blood plasma glucose, cholesterol and total protein. Dry matter of intestinal content and activity of digestive enzymes were not affected by diet. Histomorphological analyses revealed clear structural alterations in the gut of fish fed saponin, both alone and in combination with prebiotics. The results indicated a progressing inflammation with increased infiltration by immune cells particularly into the distal parts of the intestine. Gene expression profiles obtained by RNA sequencing and quantitative PCR mirrored the histological and biochemical changes induced by the saponin load. The work has provided novel basic knowledge on the anatomy, digestive and immune function of the Ballan wrasse intestine. Additionally, the study demonstrated that Ballan wrasse gut health and digestive function may be markedly affected by diet composition.

Overlapping and distinct roles of LIN28 paralogs in reprogramming and stem cell metabolism

Project description:The RNA binding protein paralogs LIN28A and LIN28B play critical roles in embryonic development, tumorigenesis, and pluripotency. Somatic cells can be reprogrammed to induced pluripotent stem (iPS) cells by overexpression of OCT4 and SOX2 together with NANOG and LIN28A (OSNA), but the exact roles LIN28A/B play are still poorly understood. Here we show that LIN28B likewise promotes reprogramming, and that reactivation of both endogenous LIN28A and B loci are required for maximal reprogramming efficiency. The LIN28B locus has open chromatin and is activated early during reprogramming, whereas LIN28A is activated at later stages by OCT4, marking the transition to bona fide iPS cells. By proteomic and metabolomic analysis, we demonstrate that LIN28A chiefly regulates let-7 and protein translation, whereas LIN28B promotes embryonic metabolism by repressing oxidative phosphorylation and enhancing glycolysis. Thus, LIN28A and LIN28B play independent and cooperative roles in reprogramming and pluripotent stem cell metabolism. 8 samples in total were analyzed. 2 replicates for each phenotypes. Controls are the wild type (flox) without lin28a/b loss of function.
