Project description:Spondyloarthropathies (SpA) are a family of rheumatic disorders that could be divided into axial (axSpA) and peripheral (perSpA) sub-forms depending on the disease clinical presentation. The chronic inflammation is believed to be driven by innate immune cells such as monocytes, rather than self-reactive cells of adaptive immune system. The aim of the study was to investigate the micro-RNA (miRNA) profiles in monocyte subpopulations (classical, intermediate and non-classical subpopulations) acquired from SpA patients or healthy individuals in search for prospective disease specific and/or disease subtype differentiating miRNA markers. Several SpA-specific and axSpA/perSpA differentiating miRNAs have been identified that appear to be characteristic for specific monocyte subpopulation. For classical monocytes, upregulation of miR-567 and miR-943 was found to be SpA-specific, whereas downregulation of miR-1262 could serve as axSpA-differentiating, and the expression pattern of miR-23a, miR-34c, mi-591 and miR-630 as perSpA-differentiating markers. For intermediate monocytes, expression levels of miR-103, miR-125b, miR-140, miR-374, miR-376c and miR-1249 could be used to distinguish SpA patients from healthy donors, whereas the expression pattern of miR-155 was identified as characteristic for perSpA. For non-classical monocytes, differential expression of miR-195 was recognized as general SpA indicator, while upregulation of miR-454 and miR-487b could serve as axSpA-differentiating, and miR-1291 as perSpA-differentiating markers. Our data indicate for the first time that in different SpA subtypes, monocyte subpopulations bear disease-specific miRNA signatures that could be relevant for SpA diagnosis/differentiation process and may help to understand SpA etiopathology in the context of already known functions of monocyte subpopulations.

T helper lymphocyte- and monocyte-specific type I interferon (IFN) signatures in autoimmunity and viral infection.

Project description:This study demonstrates quantitative and qualitative differences between type I IFN signatures in autoimmunity and viral infection using purified CD4pos T cells and CD16pos- and CD16neg-monocyte subsets. We were able to discriminate between cell-specific viral response signatures and the pathogenically amplified IFN signatures observed in autoimmunity. The differences were of both a qualitative and quantitative nature, as the signatures in the patients with SLE were characterized by much more complexly compiled gene patterns with increased absolute gene expression levels.

Human dermal fibroblast subpopulations display distinct gene signatures related to cell behaviours and matrisome.

Project description:The upper papillary and deeper reticular dermis differ structurally and functionally. Although the papillary and reticular fibroblasts produced distinct extracellular matrix (ECM), the matrisome of these two fibroblast subpopulations has not been defined. Therefore we performed a transcriptomic analysis of papillary and reticular fibroblasts freshly isolated from skin of young donors around 20, at a time they produced high level of ECM. Bioinformatics analysis delivered 230 upregulated and 139 downregulated transcripts in papillary vs reticular fibroblasts. Expression of various selected genes was validated by q-PCR. The papillary fibroblasts were characterized by a higher expression of genes involved in the defense function, in the regulation of cell motility and proliferation and in the MAPK cascade whereas reticular fibroblasts showed higher expression of genes related to the development of connective tissues. Papillary fibroblasts were characterized by the expression of a high number of matrisome-associated genes whereas reticular fibroblasts gene signature mainly related to the core matrisome and ECM regulators. The regulation of selected genes was validated at protein level attesting to the robustness of the transcriptome analysis. Altogether, our data brought new insights into an ECM signature that is coherent with the organization and function of the papillary and reticular dermis.
