Project description:Gene expression profiling of primary stromal cell cultures isolated from human endometrium and ovarian endometriosis. Samples are derived from the endometrium of 6 healthy patients and the endometriomas of 6 diseased patients. The results indicate the gene expression differences between these two cell populations. Stromal cells were obtained from human normal endometrial tissues and ovarian endometriomas. The tissues were digested enzymatically, and pure stromal cell populations were established.

Project description:Goal: to profile gene expression change that occur when cancer-associated firboblast precursor cells are co-cultured with normal epithelial cells (IOSE4) or epithelial ovarian cancer cells (HeyA8) MSCs and INOF cells were co-cultured with IOSE4 cells or Hey.A8 EOC cells. 1000 stromal cells were plated into 6 well plates and 10,000 epithelial cells plated into porous inserts (pore size 1μm, Greiner BioOne). Project description:RNA was extracted from normal human granulosa cells from IVF patients (hGC1 and hGC2 samples) and from adult-type ovarian granulosa cell tumor samples (H1, H8, H20, H23, H24, H28, H30, H33, H4, H18) as described in Jamieson et al, 2010. RNA from all samples was linearly amplified using the Whole Transcriptome Amplification kit (Sigma), starting from 300ng of RNA, and with 12 amplifications cycles. cDNA was purified on columns and sent to the Nimblegen platform for hybridization and transcriptional profiling. The FOXL2 locus was gentoyped in tumor samples, and all samples were found positive for the recurrent somatic mutation p.Cys134Trp which is present in >95% of adult-type ovarian granulosa cell tumors (Shah et al, 2009).

Project description:Ascites or solid tumour from patients with ovarian cancer was collected and grown in culture as ex vivo models. Each sample has a mixture of tumour and stromal cells which were separated into individual cultures. Therefore each patient has tumour and stromal cultures originating from the same tissue collection. Variant calling (exome-seq) analysis was performed on these matched models to establish tumour specific mutations. Stromal cells were used to rule out germline mutations.

Project description:The etiology of ovarian cancer is poorly understood, mainly due to the lack of an appropriate experimental model for studying the onset and progression of this disease. We have created a mouse model termed ERalpha d/d in which a conditional deletion of estrogen receptor alpha (ERalpha) gene occurred in the anterior pituitary, but ERalpha expression remained intact in the hypothalamus and the ovary. The loss of negative-feedback regulation by estrogen (E) at the level of the pituitary led to elevated production of luteinizing hormone (LH) by this tissue. Hyperstimulation of ovarian cells by LH resulted in increased steroidogenesis, leading to high circulating levels of progesterone, testosterone and E. The ERalpha d/d mice exhibited formation of palpable ovarian epithelial tumors starting at 5 months of age, and by 12 months, most mice carrying these tumors died. Besides proliferating epithelial cells, these tumors also contained an expanded population of stromal cells, which express P450 aromatase suggesting that these cells acquired the ability to synthesize E. In ERalpha d/d mice, in response to the E produced by the stromal cells, the ERalpha signaling is accentuated in the ovarian epithelial cells, triggering increased ERalpha-dependent gene expression, abnormal cell proliferation, and tumorigenesis. The ERalpha d/d animal model of ovarian epithelial tumorigenesis will serve as a powerful tool for exploring the involvement of E-dependent signaling pathways in the etiology of ovarian cancer. To identify aberrantly regulated genes in epithelial ovarian tumors, we performed gene expression profling of ovarian tumor tissue isolated from ERaplha d/d mice and normal ovary tissue isolated from ERaplha f/f control mice. The ERalpha d/d mouse model was created via conditional deletion of ERalpha by employing the Cre-LoxP strategy. Transgenic mice expressing Cre recombinase under the control of progesterone receptor (PR) promoter, termed PR-cre mice, were crossed with mice harboring the M-bM-^@M-^XfloxedM-bM-^@M-^Y ERalpha gene (ERalpha f/f) to create the ERalpah d/d mice in which the ERalpha gene is deleted in cells expressing PR. Ovarian tumor tissue was isolated from ERalpha d/d mice and normal ovary tissue was isolated from ERalpha f/f mice at 5 months and 10 months of age. Tissues were snap frozen and total RNA was isolated. Total RNA was pooled from 3 mice for each sample subjected to microarray analysis.

Project description:We report the application of RNA-sequencing for high-throughput profiling of gene expression in hedgehog-responsive stromal cells in normal mouse prostate and mouse prostate tumors. By using the Gli1-GFP knock-in reporter mouse line, we isolated the subset of mouse prostate stromal cells undergoing hedgehog signaling to compare the transcriptomes between PB-MYC prostate tumor and normal prostate in mice at the age of about 45 weeks.
