Project description:Pouteria lucuma Ruiz and Pav., known as the 'Gold of the Incas' or 'lucuma', is a subtropical fruit belonging to the Sapotaceae family, with a very sweet flavor, used to prepare cakes, ice creams as well as in the baking and dairy industries. To date, the content of primary metabolites is known, but little information is reported about the composition in specialized metabolites. Moreover, no study is reported on skin which represent an important agricultural waste due to the high demand for lucuma. In order to have a preliminary metabolite profile of Pouteria lucuma, the extracts of pulp and skin have been analyzed by LC-ESI/LTQOrbitrap/MS/MS in negative ion mode. The careful analysis of the accurate masses, of the molecular formulas and of the ESI/MS spectra allowed to identify specialized metabolites belonging to phenolic, flavonoid and polar lipid classes. The LC-MS/MS analysis guided the isolation of compounds occurring in the pulp extract whose structures have been characterized by spectroscopic methods including 1D- and 2D-NMR experiments and ESI-MS analysis. Furthermore, the phenolic content of the extracts along with the antioxidant activity of extracts and isolated compounds was evaluated.

Project description:In this study, we determined the complete plastome sequence of Pouteria campechiana (Kunth) Baehni (Sapotaceae) (NCBI acc. no. KX426215). This is the first time a plastome from the Sapotaceae has been sequenced. The gene order and structure of the P. campechiana plastome are collinear with those of the typical plastome of land plants. The complete plastome size is 157,922?bp in length and consists of a large single-copy region of 87,122?bp and a small single-copy region of 18,559?bp, which are separated by a pair of 26,120?bp-long inverted repeat regions. The overall A-T content of the plastome sequence is 63.2%. The plastome contains 113 genes, of which 79 are protein-coding genes, 30 are tRNA genes, and 4 are rRNA genes. Sixteen genes contain one intron and two genes have two introns. A total of 91 simple sequence repeat loci were identified within the genome. Phylogenetic analysis revealed that P. campechiana is a sister group of the Primulaceae-Ebenaceae clade with 100% bootstrap support.

Project description:Natural products are vital in drug discovery and the search for anticancer agents has been significant importance to the researchers for a long time. In the present study, aqueous leaf extract of Pouteria sapota (P.sapota) was evaluated for its cytotoxic activity. The leaf extract was preliminarily screened for antioxidant activity using DPPH method for Radical Scavenging Activity, Hydrogen Peroxide Scavenging Activity and Reducing Power Activity. Further, the aqueous leaf extract was screened for cytotoxic activity against breast cancer cell lines (MCF-7) in vitro. The results of the study showed that aqueous extract of the P.sapota leaf was rich in phytochemicals, antioxidant activity and showed a significant anti-cancer activity against tested MCF-7 cell lines. The present study was designed to evaluate the anticancer potential of P.sapota leaf. The antioxidants present in P.sapota have strong cytotoxic activity suggests that it can be considered for anti-cancer treatment.

Project description:Abiu fruit (Pouteria caimito [Ruiz & Pavon.] Radlk.) is endemic to the Amazonian region of South America, the fruit is also called yellow star apple, blueberry pie fruit and cauje. In this study, the chloroplast genome sequence of P. caimito was assembled and characterized using Illumina sequencing. The whole chloroplast genome of the wild species of abiu fruit is 158,916 bp, composed of four regions: a large single-copy region (88,096 bp) and a small single copy (18,620 bp) region, separated by two inverted repeat regions (26,100 bp), and the GC content is 36.83%. A total of 133 genes were annotated, including 88 that encoded proteins, eight that encoded rRNA genes and 37 that encoded tRNA genes. A maximum likelihood tree was constructed based on the sequences of chloroplast genome, the results showed that the wild species of P. caimito is the most closely related to Pouteria campechiana. This study provided abundant genomics data for the research and development of P. caimito.
