Project description:Metals, including copper (Cu) and nickel (Ni) are among the most common contaminants in soils in Europe. Although their effects are relatively well known regarding survival and reproduction of soil invertebrates, their modes of action in these organisms are still poorly studied. Enchytraeus albidus has been used in soil ecotoxicology for many years, and more recently has a gene library and an oligonucleotide microarray for this species which allowed gene expression studies. This has potentiated the means to study further in depth the mechanisms of response to stressors. The main aim of this study is to understand the mechanisms of response of E. albidus to Cu and Ni. For that we have 1) assessed and compared the transcriptomic profile of E. albidus in response to Cu and Ni and 2) compared the Cu, Ni, Cd and Zn transcriptomic profiles. For the microarray hybridizations, E. albidus were exposed to the reproduction effect concentrations EC50 and EC90 of Cu and Ni during 4 days. Results indicate that Cu and Ni have to some extent, similar mechanisms of toxicity and that have already been identified in other species, indicating cross-species conserved mechanisms. Based on hierarchical clustering, it was possible to observe a clear separation of Cd treatments from all other metals. This separation strongly correlates with the available information regarding the toxicokinetics of the tested metals, in which Ni shows properties similar to essential metals.

Project description:Given that transition metals are essential cofactors in central biological processes, misallocation of the wrong metal ion to a metalloprotein can have resounding and often detrimental effects on diverse aspects of cellular physiology. Therefore, in an attempt to characterize unique and shared responses to chemically similar metals we have reconstructed physiological behaviors of Halobacterium NRC-1, an archaeal halophile, in sub-lethal levels of Mn(II), Fe(II), Co(II), Ni(II), Cu(II) and Zn(II). Over 20% of all genes responded transiently within minutes of exposure to Fe(II), perhaps reflecting immediate large scale physiological adjustments to maintain homeostasis. At steady state, each transition metal induced growth arrest, attempts to minimize oxidative stress, toxic ion scavenging, increased protein turnover and DNA repair, and modulation of active ion transport. While several of these constitute generalized stress responses, up regulation of active efflux of Co(II), Ni(II), Cu(II), and Zn(II), down regulation of Mn(II) uptake and up regulation of Fe(II) chelation, confer resistance to the respective metals. We have synthesized all these discoveries into a unified systems level model to provide an integrated perspective of responses to six transition metals with emphasis on experimentally verified regulatory mechanisms. Finally, through comparisons across global transcriptional responses to different metals we provide insights into putative in vivo metal selectivity of metalloregulatory proteins and demonstrate that a systems approach can help rapidly unravel novel metabolic potential and regulatory programs of poorly studied organisms. Project description:Polymer chemical recycling to monomers (CRM) is important to help achieve a circular plastic economy, but the "rules" governing catalyst design for such processes remain unclear. Here, carbon dioxide-derived polycarbonates undergo CRM to produce epoxides and carbon dioxide. A series of dinuclear catalysts, Mg(II)M(II) where M(II) = Mg, Mn, Fe, Co, Ni, Cu, and Zn, are compared for poly(cyclohexene carbonate) depolymerizations. The recycling is conducted in the solid state, at 140 °C monitored using thermal gravimetric analyses, or performed at larger-scale using laboratory glassware. The most active catalysts are, in order of decreasing rate, Mg(II)Co(II), Mg(II)Ni(II), and Mg(II)Zn(II), with the highest activity reaching 8100 h-1 and with >99% selectivity for cyclohexene oxide. Both the activity and selectivity values are the highest yet reported in this field, and the catalysts operate at low loadings and moderate temperatures (from 1:300 to 1:5000, 140 °C). For the best heterodinuclear catalysts, the depolymerization kinetics and activation barriers are determined. The rates in both reverse depolymerization and forward CHO/CO2 polymerization catalysis show broadly similar trends, but the processes feature different intermediates; forward polymerization depends upon a metal-carbonate intermediate, while reverse depolymerization depends upon a metal-alkoxide intermediate. These dinuclear catalysts are attractive for the chemical recycling of carbon dioxide-derived plastics and should be prioritized for recycling of other oxygenated polymers and copolymers, including polyesters and polyethers. This work provides insights into the factors controlling depolymerization catalysis and steers future recycling catalyst design toward exploitation of lightweight and abundant s-block metals, such as Mg(II).

Project description:This objective of this study was to identify gene expression and biological mechanisms related to trace mineral metabolism, central to inflammatory mediation and disease development, through the evaluation of the whole blood transcriptome in post‐weaned cattle receiving different mineral treatments. Here, 56 high‐risk cross-bred beef cattle (~250 kg) were assigned to receive ad libitum access to supplements containing 1 of 3 treatments over a 60-day period: 1) sulfate sources of Cu, Co, Mn, and Zn in feed (INR; custom blend; n= 40); 2) organic complexed source of Cu, Mn, Co, and Zn in feed (AAC; Availa4; n = 40); or 3) organic complexed source of Cu, Mn, Co, and Zn in feed and administration of a drench product [ProFusion] upon arrival (COMBO; n = 40). Whole blood RNA from 26 cattle (~4 calves/pen) were matched and utilized to assess the host transcriptome at days 0, 13, 28, 45, and 60 for the assessment of host transcriptome responses over time.

Project description:Six trinuclear CuIIMIICuII compounds (M = Cu, Ni, Co, Fe, Mn, Zn) derived from the Schiff base ligand, H2L (2 + 1 condensation product of salicylaldehyde and trans-1,2-diaminocyclohexane) are reported in this investigation. The composition of the metal complexes are [{CuIIL(ClO4)}2CuII(H2O)]·2H2O (1), [{CuIIL(ClO4)}{NiII(H2O)2}{CuIIL}]ClO4·CH3COCH3 (2), [{CuIIL(ClO4)}{CoII(CH3COCH3)(H2O)}{CuIIL(CH3COCH3)}]ClO4 (3) and isomorphic [{CuIIL(ClO4)}2MII(CH3OH)2] (4, M = Fe; 5, M = Mn; 6, M = Zn). Two copper(ii) ions in 1-6 occupy N2O2 compartments of two L2- ligands, while the second metal ion occupies the O(phenoxo)4 site provided by the two ligands, i.e., the two metal ions in both CuIIMII pairs are diphenoxo-bridged. Positive ESI-MS of 1-6 reveals some interesting features. Variable-temperature and variable-field magnetic studies reveal moderate or weak antiferromagnetic interactions in 1-6 with the following values of magnetic exchange integrals (H = -2JS 1 S 2 type): J 1 = -136.50 cm-1 and J = 0.00 for the CuIICuIICuII compound 1; J 1 = -22.16 cm-1 and J = -1.97 cm-1 for the CuIINiIICuII compound 2; J 1 = -14.78 cm-1 and J = -1.86 cm-1 for the CuIICoIICuII compound 3; J 1 = -6.35 cm-1 and J = -1.17 cm-1 for the CuIIFeIICuII compound 4; J 1 = -6.02 cm-1 and J = -1.70 cm-1 for the CuIIMnIICuII compound 5; J = -2.25 cm-1 for the CuIIZnIICuII compound 6 (J is between two CuII in the N2O2 compartments; J 1 is between CuII and MII through a diphenoxo bridge).

Project description:We describe a novel solid phase sorbent that was synthesized by coupling graphene oxide (GO) to ethylenediamine (EDA). This nanomaterial (referred to as GO-EDA) is capable of adsorbing the ions of iron, cobalt, nickel, copper, zinc and lead. The ethylenediamine-modified graphene oxide was characterized by X-ray photoelectron spectroscopy, scanning electron microscopy and Fourier transform infrared spectroscopy. The analytical procedure relies on (a) sorption of metal ions on GO-EDA dispersed in aqueous samples; (b) filtering, and (c) direct submission of the filter paper to energy-dispersive X-ray fluorescence spectrometry. This kind of dispersive micro-solid phase extraction was optimized with respect to pH values, concentration of GO-EDA, contact time, and the effects of interfering ions and humic acid on recovery of determined elements. Under optimized conditions, the recoveries of spiked samples range from 90 to 98 %. The detection limits are 0.07, 0.10, 0.07, 0.08, 0.06 and 0.10 ng mL-1 for Fe(III), Co(II), Ni(II), Cu(II), Zn(II) and Pb(II), respectively. The method has a relative standard deviation of <6 %, and its accuracy was verified by analysis of two standard reference materials [LGC6016 (estuarine water) and BCR-610 (groundwater)]. It was successfully applied to the determination of trace amounts of these metal ions in water samples. Graphical AbstractGraphene oxide was coupled to ethylenediamine in order to obtain an effective sorbent (GO-EDA) for preconcentration of Fe(III), Co(II), Ni(II), Cu(II), Zn(II) and Pb(II) from environmental water samples.

Project description:The catalysed ring opening copolymerizations (ROCOP) of carbon dioxide/epoxide or anhydride/epoxide are controlled polymerizations that access useful polycarbonates and polyesters. Here, a systematic investigation of a series of heterodinuclear Mg(II)M(II) complexes reveals which metal combinations are most effective. The complexes combine different first row transition metals (M(II)) from Cr(II) to Zn(II), with Mg(II); all complexes are coordinated by the same macrocyclic ancillary ligand and by two acetate co-ligands. The complex syntheses and characterization data, as well as the polymerization data, for both carbon dioxide/cyclohexene oxide (CHO) and endo-norbornene anhydride (NA)/cyclohexene oxide, are reported. The fastest catalyst for both polymerizations is Mg(II)Co(II) which shows propagation rate constants (kp ) of 34.7 mM-1 s-1 (CO2 ) and 75.3 mM-1 s-1 (NA) (100 °C). The Mg(II)Fe(II) catalyst also shows excellent performances with equivalent rates for CO2 /CHO ROCOP (kp =34.7 mM-1 s-1 ) and may be preferable in terms of metallic abundance, low cost and low toxicity. Polymerization kinetics analyses reveal that the two lead catalysts show overall second order rate laws, with zeroth order dependencies in CO2 or anhydride concentrations and first order dependencies in both catalyst and epoxide concentrations. Compared to the homodinuclear Mg(II)Mg(II) complex, nearly all the transition metal heterodinuclear complexes show synergic rate enhancements whilst maintaining high selectivity and polymerization control. These findings are relevant to the future design and optimization of copolymerization catalysts and should stimulate broader investigations of synergic heterodinuclear main group/transition metal catalysts.
