Project description:To mimic the effect of ROS in contributing to ageing process in immune cells of PBMC. The PBMC cells were harvested from old human subjects (>50) and induced with H2O2 during 3 hour and 6 hour. The CEL file were normalized using RMA methods. We used microarray to explore the gene expression profiling before and after ROS. Gene related to cell cycle, apoptosis, metabolism and signal transduction were differentially expressed after inducing with H2O2 in compared to baseline.

Project description:Reactive oxygen species (ROS) are key signalling molecules that regulate growth and development and coordinate responses to biotic and abiotic stresses. ROS homeostasis is controlled through a complex network of ROS production and scavenging enzymes. Recently, the first genes involved in ROS perception and signal transduction have been identified and, currently, we are facing the challenge to uncover the other players within the ROS regulatory gene network. The specificity of ensuing cellular responses depends on the type of ROS and their subcellular production sites. Various experimental systems, including catalase-deficient plants, in combination with genome-wide expression studies demonstrated that increased hydrogen peroxide (H2O2) levels significantly affect the transcriptome of plants and are capable of launching both defence responses and cell death events. We used microarrays to assess differential gene expression provoked by H2O2 from plastid or peroxisomal origin, respectively.

A microsomal proteomics view of H2O2- and ABA-dependent responses

Project description:The plant hormone abscisic acid (ABA) modulates a number of plant developmental processes and responses to stress. In planta, ABA has been shown to induce reactive oxygen species (ROS) production through the action of plasma membrane-associated NADPH oxidases. Although quantitative proteomics studies have been performed to identify ABA- or hydrogen peroxide (H2O2)-dependent proteins, little is known about the ABA- and H2O2-dependent microsomal proteome changes. Here, we examined the effect of 50 µM of either H2O2 or ABA on the Arabidopsis microsomal proteome using tandem mass spectrometry and identified 86 specifically H2O2-dependent and 52 specifically ABA-dependent proteins that are differentially expressed. We observed differential accumulation of proteins involved in the TCA cycle notably in response to H2O2. Of these, aconitase 3 responded to both H2O2 and ABA. Additionally, over 30 proteins linked to RNA biology responded significantly to both treatments. Gene ontology categories such as ‘response to stress’ and ‘transport’ were enriched, suggesting that H2O2 or ABA directly and/or indirectly cause complex and partly overlapping cellular responses.

Project description:ROS once released from myeloid cells is quickly converted into H2O2 in lungs. We performed RNAseq of cultured pulmonary primary endothelial cells treated with or without H2O2. Pathway enrichment analysis revealed that some of the signaling pathways that are altered by H2O2 are related to AKT signaling. AKT signaling has a protective role in a murine model of ALI by preventing capillary leakage. Moreover, H2O2 stimulates AKT activation in endothelial cells to strengthen vessel barrier integrity
