Tripartite motif-containing 33 (Trim33) determines the pathogenic function of Th17 cells

Project description:We report that Tripartite motif-containing 33 (Trim33), a protein that was previously associated with TGF-beta signaling, determines the pathogenic function of Th17 cells. Trim33 deficiency in T cells resulted in resistance to an autoimmune disease model. Lack of Trim33 did not impact TGF-beta signaling in mediating Foxp3 gene expression but greatly reduced TGF-beta induction of IL-17 production during Th17 cell differentiation. Importantly, we found TGF-beta not only increased IL-17 but also suppressed IL-10 expression; absence of Trim33 or Smad2 but not Smad4 in T cells enhanced IL-10 expression. In a Smad2-dependent manner, Trim33 was recruited to Il17 and Il10 gene loci and was crucial in appropriate histone modification accompanying Th17 differentiation. Our study thus demonstrates that Trim33, a non-canonical branch of TGF-beta signaling, programs the pro-inflammatory function of Th17 differentiation by promoting IL-17 and suppressing IL-10 expression. As a critical switch of pathogenic versus regulatory phenotype of T cells, Trim33 may be targeted in treating human autoimmune diseases.

Project description:We demonstrated that TSPAN18 was overexpressed in prostate cancer (PCa) and markedly accelerated PCa cell migration and invasion in vitro and bone metastasis in vivo by inhibiting E3 ligase tripartite motif containing 32 (TRIM32)-mediated stromal interaction molecule ubiquitination and degradation. To further investigate the genes regulated by TSPAN18, we knockdown TSPAN18 by siRNA and performed high-throughput sequence. So we identify a series of genes regulated by TSPAN18.

Project description:Abstract: Gene expression is controlled by transcription factors (TFs) that bind cognate DNA motifs in cis-regulatory elements (CREs), however, the combinations of motifs that mediate cell type-specific gene regulation are undefined. Chromatin accessibility, TF footprinting, and H3K27ac-dependent DNA looping data were generated to identify 7,531 cell type-specific CREs across 15 normal human cell types. A co-enrichment framework nominated 838 cell type-specific, recurrent heterotypic DNA motif combinations (DMCs), which were functionally validated using massively parallel reporter assays. Compared to their normal counterparts, cancer cells induced DMCs linked to neoplasia-enabling processes that operate in normal cells while also engaging entirely new DMCs. These data identify a cis-regulatory combinatorial DNA motif lexicon for cell type-specific gene regulation in diverse normal and diseased human cells. Purpose: To identify a cis-regulatory combinatorial DNA motif lexicon mediating cell type-specific gene regulation in diverse normal and diseased human cells, matched RNA-seq, chromatin accessibility profiling via ATAC-seq, and enhancer-promoter looping via H3K27ac HiChIP was performed in 15 primary human cell types and 6 human cancer cell lines. Processed and raw data for all cell types can be found in this repository. Squamous cell carcinoma cell line raw and processed data can be found at Jung, et al. "XXX". All biological replicates (n=2) are merged.

The master energy homeostasis regulator PGC-1α couples transcriptional co-activation and mRNA nuclear export

Project description:PGC-1α plays a central role in maintaining the mitochondrial and energy metabolism homeostasis, linking external stimuli to the transcriptional co-activation of genes involved in adaptive and age-related pathways. The carboxyl-terminus encodes a serine/arginine-rich (RS) region and a putative RNA recognition motif, however potential RNA-processing role(s) have remained elusive for the past 20 years. Here, we show that the RS domain of human PGC-1α directly interacts with RNA and the nuclear RNA export factor NXF1. Inducible depletion of endogenous PGC-1α and expression of RNAi-resistant RS-deleted PGC-1α further demonstrate that the RNA-binding activity is required for nuclear export of co-activated transcripts and mitochondrial homeostasis. Moreover, a quantitative proteomics approach confirmed PGC-1α-dependent RNA transport and mitochondrial-related functions, identifying also novel mRNA nuclear export targets in age-related telomere maintenance. Discovering a novel function for a major cellular homeostasis regulator provides new directions to further elucidate the roles of PGC-1α in gene expression, metabolic disorders, ageing and neurodegenerative diseases.
