Project description:Mitogen-Activated Protein 3 Kinase 1 (MAP3K1) is a dynamic signaling molecule with myriad cell-type specific functions not yet fully understood. Here we describe a role of MAP3K1 in the development of female reproductive tract (FRT). MAP3K1 kinase domain deficient females (Map3k1ΔKD) exhibit imperforate vagina, labor failure and sterility. The defects correspond to shunted Müllerian duct (MD), precursor of the FRT, in embryos, and contorted caudal vagina and abrogated vagina-urogenital sinus fusion in newborns. Although MAP3K1 acts through JNK and ERK to activate WNT in epithelial cells, it is dispensable for epithelial but crucial for mesenchymal WNT activity at the caudal embryonic MD in vivo. Moreover, conditional media derived from MAP3K1-compentent, but not -deficient, epithelial cells activate TCF-luciferase reporter in fibroblasts, suggesting MAP3K1 induces secreted activators from epithelial cells to activate WNT in fibroblasts. Correspondingly, the expression of Wnt7b is high in wild type, but low in Map3k1 knockout caudal MD epithelium and MAP3K1-deficient keratinocytes. Our data reveal a temporal-spatial paracrine MAP3K1-WNT crosstalk in the regulation of caudal MD elongation and FRT development.

Project description:Hox genes are key regulators of development. In mammals, the study of these genes is greatly confounded by their large number, overlapping functions, and their interspersed shared enhancers. In this report, we describe a novel recombineering strategy that was used to introduce simultaneous frameshift mutations into the flanking Hoxa9, Hoxa10, and Hoxa11 genes, as well as their paralogs on the HoxD cluster. The resulting mutant mice displayed dramatic homeotic transformations of the reproductive tracts, with uterus anteriorized towards oviduct and the vas deferens anteriorized towards epididymis. The Hoxa9,10,11 mutant mice provided a sensitized genetic background that allowed the discovery of Hoxd9,10,11 reproductive tract patterning function. Both shared and distinct Hox functions were defined. The HoxD genes played a crucial role in the regulation of the uterine immune function. Non-coding nonpolyadenylated RNAs were among the key Hox targets. In addition, we observed a surprising anti-dogmatic posteriorization of the uterine epithelium. Reproductive tracts were collected from WT and Hox mutant mice (n=3/genotype) aged 3-7 months in order to characterize the molecular changes caused by mutation of Hoxa9,10,11 and Hoxd9,10,11. Female mice were staged and collected in diestrus.

Project description:Fertility depends, in part, on interactions between male and female reproductive proteins inside the female reproductive tract (FRT) that mediate postmating changes in female behavior, morphology, and physiology. Coevolution between interacting proteins within species may drive reproductive incompatibilities between species, yet the mechanisms underlying postmating-prezygotic isolating barriers remain poorly resolved. Here, we used quantitative proteomics in sibling Drosophila species to investigate the molecular composition of the FRT environment and its role in mediating species-specific postmating responses. We found that (1) FRT proteomes in D. simulans and D. mauritiana virgin females express unique combinations of secreted proteins and are enriched for distinct functional categories, (2) mating induces substantial changes to the FRT proteome in D. mauritiana but not in D. simulans, and (3) the D. simulans FRT proteome exhibits limited postmating changes irrespective of whether females mate with conspecific or heterospecific males, suggesting an active female role in mediating reproductive interactions. Our study suggests that divergence in the FRT extracellular environment and postmating response contribute to previously described patterns of postmating-prezygotic isolation and the maintenance of species boundaries.

Project description:In internally fertilizing organisms, mating involves a series of highly coordinated molecular interactions between the sexes that occur within the female reproductive tract. In species with promiscuous mating systems, traits involved in postcopulatory interactions are expected to evolve rapidly, potentially leading to postmating-prezygotic (PMPZ) reproductive isolation between diverging populations. Here, we use a novel study design to investigate the postmating transcriptional response of Drosophila mojavensis female reproductive tracts following copulation with either conspecific or heterospecific (D. arizonae) males at three time points postmating. Relatively few genes (15 total) were transcriptionally regulated in the female lower reproductive tract in response to conspecific mating. Heterospecifically-mated females exhibited significant perturbations in the expression of the majority of these genes, and also downregulated transcription of a number of others, including several involved in mitochondrial function. These striking regulatory differences indicate failed postcopulatory molecular interactions between the sexes consistent with the strong PMPZ isolation observed for this cross. We also report, for the first time, the transfer of male mRNA transcripts to females during copulation. These included transcripts from male accessory-gland proteins (ACPs), a finding with potentially broad implications for understanding postcopulatory molecular interactions between the sexes. Project description:Precursors for the male and female reproductive tracts (also known as the Wolffian duct and the Müllerian duct) co-exist in a mammalian embryo before sexual differentiation. The Wolffian duct and Müllerian duct are surrounded by their own mesenchymes in the mesonephros, which can be distinguished by their specific expression of Gli1 and Amhr2, respectively. In the XX embryo, during sexual differentiation, the Wolffian duct degenerates. In this study, by tracking the fate of the Gli1+ Wolffian duct mesenchyme in the tamoxifen inducible genetic lineage tracing model Gli1-CreER; Rosa-Tomato, we demonstrate that the Wolffian duct mesenchyme does not regress along with the Wolffian duct epithelium in the XX embryo. Instead, the Wolffian duct mesenchyme is repurposed and becomes a unique mesenchymal population in the female reproductive tract. Profiling the transcriptomes and chromatin accessibilities of the Gli1+ Wolffian duct mesenchyme in XX and XY during sexual differentiation (E14.5 and E16.5) shows signaling pathways and transcriptional factors that potentially involves the female and male fate differentiation of the Gli1+ Wolffian duct mesenchyme. Comparing the transcriptomes between the Gli1+ Wolffian duct mesenchyme and the Amhr2+ Müllerian duct mesenchyme from the neonatal uterus reveals similarities and differences between the two mesenchymal populations in the female reproductive tract. Ex vivo ablation of the Wolffian duct mesenchyme stunted the growth of the fetal female reproductive tract. In summary, our discovery of the contribution of the Wolffian duct mesenchyme to the female reproductive tract provides new insights into our understanding of sexual differentiation of reproductive tracts.
