Project description:6-8 weeks male LysCre/+ OXCT1fl/fl (OXCT1mKO or mKO for short) and its littermates Lys+/+ OXCT1fl/fl (control or ctrl) mice were used to build HCC implantation model (Hep1-6 in Portal Vein injection). RNA was extracted from tumor associated macrophages of OXCT1mKO and their littermate control mice for transcriptome analysis of macrophage function.

Project description:miRNA played an important role in the process of carcinogenesis in HBV related hepatocellular carcinoma. Therefore, we performed miRNA microarray to evaluate the miRNAs that expressed differentially between HCC tumor versus non-tumor liver tissues. RNA was extracted from snap fresh tissue collected from resected HCC tumor and adjacent non-tumor liver tissues. All HCC tumors were HBV-associated HCC.

Neuregulin 4 suppresses NASH-associated liver cancer through restraining tumor-prone liver microenvironment

Project description:The mammalian liver comprises heterogeneous cell types within its tissue microenvironment that respond to physiological cues and undergo pathophysiological reprogramming in disease states, such as nonalcoholic steatohepatitis (NASH). Patients with NASH are at increased risk for the development of hepatocellular carcinoma (HCC). However, the molecular and cellular nature of liver microenvironment remodeling that links NASH to liver carcinogenesis remains obscure. Here we show that diet-induced NASH is characterized by induction of tumor-associated macrophage (TAM)-like macrophages and exhaustion of cytotoxic T cells in mouse liver. The adipose-derived endocrine factor Neuregulin 4 (NRG4) serves as a hormonal checkpoint that restrains this pathological reprogramming during NASH. NRG4 deficiency exacerbates the induction of tumor-prone liver immune microenvironment and NASH-associated HCC, whereas transgenic NRG4 overexpression elicits protective effects in mice. In a therapeutic setting, recombinant NRG4 protein exhibits remarkable efficacy in inhibiting HCC in mice with NASH, thereby paving the way for future therapeutic development.

Project description:A large fraction of HCC in Peru arises in younger, non-cirrhotic patients. Global DNA methylation analysis of the HCC and non-tumor liver (NTL) tissues demostrate a profound overhaul of the developmental DNA methylation programme culminating in an global hyper-methylated pattern in HCC. We used microarrays to compare global DNA methylation between tumor and non-tumor liver tissues and identified CpGs and ontologies that are distinctively differentially methylated between subsets.

The Role of Tumor-associated Macrophages in Colorectal Liver Metastases

Project description:Colorectal cancer is a major cause of mortality worldwide. Most patients develop colorectal liver metastases (CLM), and for such patients hepatectomy combined with chemotherapy may be curative. Nevertheless, in the era of precision medicine there is a critical need of prognostic markers to cope with the heterogeneity of CLM patients. Tumor-associated macrophages (TAMs) pave the way to tissue invasion and intravasation providing a nurturing microenvironment formetastases. The quantification of immune landscape of tumors has provided novel prognostic indicators of cancer progression, and the quantification of TAMs might explain the heterogeneity of CLM patients. Here, we will investigate the development of a new diagnostic tool based on TAMs with the aim to define the causative role of TAMs in CLM patients. This will open new clinical scenarios both for the diagnosis, therapy and prognosis, leading to the refinement of the therapeutic output in a personalized medicine perspective.
