Project description:To analyze early transcriptional events in cardiac tissue after infarction and evaluate the genetic expression profile of post-infarction mesenchymal cells of the heart, we induced myocardial infarction in rats by ligation of the left coronary artery. 24 hours after surgery, the affected area was harvested for RNA isolation and cell culture. We then performed a gene expression profile analysis using data obtained from RNA sequencing of 3 different postinfarction tissues and cells. Healthy tissues and cells of the left ventricle of the heart of sham-operated rats were used as controls.

Neferine improves cardiac remodeling induced by hypertension by modulating multiple signaling pathways --heart

Project description:The goals of this study is to compare the differently expressed genes in heart tissue of WKY and SHR with or withoutneferine treatment. The SHRs were randomly divided into 2 groups: SHR, and SHR +neferine (10mg/kg/d) groups . WKY rats were set as control. Rats in WKY and SHR groups were intragastrically with double distilled water (dd H2O), while rats in SHR + neferine (10mg/kg/d) group were intragastrically with 10 mg/kg/d of neferine for 10 weeks. Then the heart tissues were used to identify differentially expressed genes among different groups.

[E-MTAB-366] Transcription profiling by array of chicken embryos at 15 different stages

Project description:Transcription profiling of chicken development The experiment were perfomed as a part of our Vertebrate Evo-Devo project. The aim of the project is to compare transcription profiles of normal (unmanipulated, wild-type, whole embryo) vertebrate embryos. Total RNA was collected from wild type G.gallus whole embryos at 15 different stages (Stages:HH1,2,4,6,8,9,11,14,16,19,24,27,32,34,38), and hybridized to the Affymetrix Chicken Genome Array. All the stages contains data from two biological replications. Each staged-samples consists of pooled total RNA from several whole embryos.

Environmentally Induced Epigenetic Transgenerational Inheritance of Somatic Transcriptomes

Project description:Environmentally induced epigenetic transgenerational inheritance of adult onset disease involves a variety of phenotypic changes suggesting a general alteration in genome activity. Investigation of eleven different tissue transcriptomes in male and female F3 generation vinclozolin versus control lineage rats demonstrated all tissues examined had unique transgenerational transcriptomes. Common cellular pathways and processes were identified among the tissues. A bionetwork analysis identified gene modules with coordinated gene expression and each had unique gene networks regulating tissue specific gene expression and function. A large number of statistically significant over-represented clusters of differentially expressed genes were identified and termed “Epigenetic Control Regions”. Combined observations demonstrate that all tissues derived from the epigenetically altered germ line develop transgenerational transcriptomes unique to the tissue, but common epigenetic control regions in the genome appear to in part coordinately regulate these tissue specific transcriptomes. This systems biology approach provides insight into the molecular mechanisms involved in the epigenetic transgenerational inheritance of a variety of adult onset disease phenotypes. We used microarrays to determine genes expressed differentially in rats 11 male or female smatic tissues -male heart, kidney, liver, testis, prostate, seminal vesicles; female heart, kidney, liver, ovary, uterus - due to Vinclozolin treatments of their grand-grandmothers.
