Project description:Background. Radioiodide 131I is commonly used to treat thyroid cancer and hyperthyroidism, and 131I releases during nuclear accidents have resulted in increased incidence of thyroid cancer in children. To develop a better understanding of underlying cellular mechanisms behind 131I exposure and identify potential biomarkers, the aim of this work was to study the long-term dose-related effects of 131I exposure in thyroid tissue and plasma in young rats. Materials and methods. Male Sprague Dawley rats (5-week-old) were i.v. injected with 0.5, 5.0, 50 or 500 kBq 131I (Dthyroid ca 1–1000 mGy), and killed after nine months at which time the thyroid and blood samples were collected. Gene expression microarray analysis (thyroid samples) and LC-MS/MS analysis (thyroid and plasma samples) were performed to assess differential gene and protein expression patterns in treated and corresponding untreated control samples. Bioinformatics analyses were performed using the DAVID functional annotation tool and Ingenuity Pathway Analysis (IPA). Results. Nine 131I exposure-related biomarkers (Afp and RT1-Bb,ARF3, DLD, IKBKB, NONO, RAB6A, RPN2, and SLC25A5) were identified in thyroid tissue. Four dose-related biomarker candidates (APRT, LDHA) and (TGM3 and DSG4) were identified in thyroid and plasma, respectively. Candidate biomarkers for thyroid function were the upstream regulator PPARG and the proteins ACADL and SORBS2 (all activities), TPO and TG (low activities)). 131I exposure was shown to have a profound effect on metabolism, the immune system, apoptosis and cell death. Furthermore, several signalling pathways essential for normal cellular function (actin cytoskeleton signalling, HGF signalling, NRF2-mediated oxidative stress, integrin signalling, calcium signalling) were also significantly regulated. Conclusion. Exposure-related and dose-related effects on gene and protein expression were observed, thereby identifying several candidate genes that could be used as potential biomarkers for exposure, absorbed dose and thyroid function.

Project description:<p>Objective: Metabolite abnormalities are potentially implicated in pathogenesis of Hashimoto’s thyroiditis (HT). An in-depth study of the relationship between HT and small molecule metabolites, as well as its pathogenesis, can help enhance the diagnosis, prognosis, and treatment of HT patients.</p><p>Methods: We used metabolomics to analyze changes in serum metabolite levels in 20 HT patients and 20 healthy controls (HC) and the most significant differentially expressed metabolite was analyzed. Western blot and qPCR were used to measure the expression of histidine decarboxylase (HDC) and histamine receptor 1 (H1R). Increasing concentrations of histidine were used to treat neutrophils and observe the effect on neutrophil extracellular traps (NETs) synthesis. Histidine treated neutrophils were co-culture with thyroid follicular cells to study the protective effects of histidine on HT thyroid follicular cells and related mechanisms.</p><p>Results: A total of 48 differentially expressed metabolites were found. Histidine exhibited reduced expression levels in HT patients, displaying the most significant discrepancy (p<0.001). ROS and NETs were increased and HDC, H1R, and histamine were upregulated in neutrophils upon stimulated. These effects were corrected by addition of histidine, in a dose dependent manner. In addition, in co-culture experiments, histidine enhanced expression of SOD and suppressed production of inflammatory cytokines IL-6 and TNF-α and NF-κB pathway in thyroid follicular cells, thereby inhibiting inflammation and oxidative stress.</p><p>Conclusion: Histidine inhibits NETs synthesis and NF-κB signaling. Thus, histidine play an anti-inflammatory and antioxidant role by decreasing thyroid follicular cell inflammation in HT.</p>

Project description:Although most thyroid tumours are benign, thyroid cancer represents the most common malignancy of the endocrine system, comprising mainly follicular and papillary thyroid carcinomas (FTC and PTC, respectively). Previous studies have shed some light on the molecular pathogenesis of thyroid cancer but there have not been any comprehensive mass spectrometry-based proteomic studies to reveal protein expression differences between thyroid tumours and the molecular alterations associated with tumour malignancy. We applied a label-free quantitative mass spectrometry analysis to compare normal thyroid tissue with the three most common tumours of the thyroid gland: follicular adenoma, follicular carcinoma and papillary carcinoma.

Project description:Acrylamide (AA) is known to produce tumors in animals in different tissues including the thyroid where typically the tumors are found in follicular cells in the thyroid gland. Using transcriptomic tools, we examined global gene expression changes in the thyroid glands of RccHan Wistar rats that were sub-chronically exposed to a low dose of AA (3 mg/Kg). A transcriptomic approach was used to investigate changes in gene expression and their association with physiological responses (changes in plasma hormone levels) in rats treated with acrylamide (AA), exposed from gestational day 6 to post natal day 21. We hypothesized that 3 mg AA/Kg bw/day exposure would produce changes in plasma hormone levels associated with key genes and biochemical pathways involved with molecular actions of AA.

Project description:Bisphenol A (BPA) is primarily used to make polycarbonate plastic, with a global capacity of production exceeding 8 million tons per year. Biomonitoring studies with human urine, blood and tissue samples suggest that humans are subjected to widespread and continuous exposure to BPA. It has been well established that early life exposure to BPA predisposes the prostate gland to carcinogenesis later in life. However, it remains unknown if BPA exposure during adulthood induces benign or neoplastic pathology in the prostate. The main objective of the present study is to determine the effects of BPA exposures during adulthood on the prostate and to characterize the global transcriptional reprogramming underlying endocrine disruption by BPA. We elevated circulating levels of free BPA in Noble rats to the human-relevant internal dose range with BPA-filled Silastic implants while maintaining the physiological levels of testosterone (T) with T-filled implants. Cotreatment with T and 17β-estradiol (E2) was our reference regimen which induced preneoplastic and cancereous lesions. The T + low/high dose of BPA induced prostatic hyperplasia, low-grade prostate intraepithelial neoplasia (LGPIN) and intraepithelial infiltration of T-lymphocytes specifically in the lateral prostate (LP). Using microarray analysis, we delineated specific impacts of low and high dose of BPA (with the T-support) on the gene expression program in LPs. Hierarchical clustering revealed that the endocrine disrupting effects of T + low dose of BPA showed partial resemblance to those of T + high dose of BPA and T+E2. In contrast, the influence of T+ high dose of BPA on the LP transcriptome was completely different from those of T + E2. Further, IPA analysis of specific T+ low or high BPA gene signature identified a transcription factor, HNF4α, as a regulatory hub affecting a number of differentially expressed genes by BPA exposures. These findings suggest that the adult rat prostate is still venerable to the endocrine disrupting effects of BPA. Perhaps chronic exposure to low dose of BPA provides a niche comprising heightened cell proliferation, inflammatory responses and disrupted gene expression program, which favor the onset and development of prostatic benign or malignant diseases in men. Rat lateral prostates were collected from untreated control and treated groups [Testosterone (T) + Estradiol (E2), T + low dose Bisphenol A (low BPA) and T + high BPA] for RNA extraction and hybridization on Affymetrix microarrays. We sought to identify transcriptional signature of BPA exposures in the lateral prostate gland.

Project description:In the current study we use untargeted transcriptomic and metabolomic analyses to address the consequence of in utero and lactational exposure to a low dose of the model thyroid hormone system disrupting chemical propyl-thio-uracyl (PTU) in mice.We detected minor changes in gene expression in the brain, and of the metabolite content of the serum. Fatty acid and lipid metabolites were the most noteworthy markers in serum.
