Project description:Full-term pregnancy (FTP) at an early age confers long-term protection against breast cancer, in this study, we evaluated gene expression changes induced by parity in the breast of premenopausal women. Gene expression profiling of normal breast tissue from 30 nulliparous (NP) and 79 parous (P) premenopausal volunteers were performed using Affymetrix microarrays. In addition to a discovery/validation analysis, we conducted analysis of gene expression differences in P vs. NP women as a function of time since last FTP.

Project description:Maternal obesity is becoming a major health consideration for successful pregnancy outcomes. There is growing proof that maternal obesity has a negative influence on placental development and function, thereby adversely influencing offspring programming and health outcomes. However, the molecular mechanisms underlying these processes are so far poorly understood. We set out to analyse term placenta whole transcriptome in obese (n=5) and normoweight women (n=5), using Affymetrix microarray platform compromising of 50,000 probe sets. Our analysis shows that the placental transcriptome differs between normoweight and obese women. Different processes and pathways among placenta from obese women were dysregulated, including inflammation and immune responses, lipid metabolism, cell death and survival and cancer pathways, vasculogenesis and angiogenesis, and glucocorticoid receptor signaling pathway. Together, this global gene expression profiling approach demonstrates and confirms that maternal obesity creates a unique in utero environment that impairs placental transcriptome.

Project description:Obesity represents a global burden with an increasing worldwide prevalence, especially in women of reproductive age. Obesity in women, defined as a body mass index (BMI) > 30 kg/m2, has a worldwide prevalence ~21%, however, it exceeds 30% in countries such as Chile, Mexico, United States, and the United Kingdom. Growing evidence support the notion that pre-gestational obesity confers an increased risk for the development of diabetes, obesity and chronic inflammatory diseases in the offspring later in life. In fact, maternal obesity during gestation is associated with an increased risk of asthma and wheezing during early or late infancy, in terms of medication and need for hospital admission. Here we report changes in DNA methylation at a genomic level in monocytes isolated by adherence from cord blood of neonates born to women with obesity and integrate these changes to reveal the epigenetic programming of immune signaling pathways resulting from maternal obesity.

Project description:Activation of inflammatory pathways is one plausible mechanism underlying the association between obesity and increased breast cancer risk. However, macrophage infiltration and local biomarkers of inflammation in breast adipose tissue have seldom been studied in association with obesity. Gene expression profiles of normal breast tissue from reduction mammoplasty patients were evaluated by whole genome microarrays to identify patterns associated with obesity status (normal-weight, body mass index (BMI) <25; overweight, BMI 25-29.9; obese, BMI > or equal to 30). The presence of macrophage-enriched inflammatory loci with immunopositivity for CD68 protein was evaluated by immunohistochemistry (IHC). After adjusting for confounding by age, 760 genes were differentially expressed (203 up and 557 down; FDR = 0.026) between normal-weight and obese women. Gene ontology analysis suggested significant enrichment for pathways involving IL-6, IL-8, CCR5 signaling in macrophages and RXRalpha and PPARalpha activation, consistent with a pro-inflammatory state and suggestive of macrophage infiltration. Gene set enrichment analysis also demonstrated that the genomic signatures of monocytes and macrophages were over-represented in the obese group with FDR of 0.08 and 0.13, respectively. Increased macrophage infiltration was confirmed by IHC, which showed that the breast adipose tissue of obese women had higher average macrophage counts (mean = 8.96 vs. 3.56 in normal-weight women) and inflammatory foci counts (mean = 4.91 vs. 2.67 in normal-weight women). Obesity is associated with local inflammation and macrophage infiltration in normal human breast adipose tissues. Given the role of macrophages in carcinogenesis, these findings have important implications for breast cancer etiology and progression. 72 normal breast tissue samples from patients undergoing reduction mammoplasty. Project description:Transcription profiling of chicken development The experiment were perfomed as a part of our Vertebrate Evo-Devo project. The aim of the project is to compare transcription profiles of normal (unmanipulated, wild-type, whole embryo) vertebrate embryos. Total RNA was collected from wild type G.gallus whole embryos at 15 different stages (Stages:HH1,2,4,6,8,9,11,14,16,19,24,27,32,34,38), and hybridized to the Affymetrix Chicken Genome Array. All the stages contains data from two biological replications. Each staged-samples consists of pooled total RNA from several whole embryos.

Project description:This study aimed to compare the proteomic profile of stimulated and unstimulated saliva sam-ples from pregnant women with/without obesity and periodontitis. Initially, 126 pregnant women were recruited and, after exclusions, were allocated into: with obesity and periodontitis (OP); with obesity but without periodontitis (OWP); with normal BMI but with periodontitis (NP); with normal BMI and without periodontitis (NWP). Stimulated saliva (SS) and unstimulated saliva (US) samples were collected, and salivary proteins were extracted and individually processed by proteomic analysis (nLC-ESI-MS/MS). Proteins involved with immune response process, antiox-idant activity, and retina homeostasis were decreased or absent in SS samples from all groups (i.e., Antileukoproteinase, Lysozyme C, Alpha-2-macroglobulin-like protein 1, Heat shock proteins – 70kDa 1-like, 1A, 1B, 6, Heat shock-related 70 kDa protein 2, Putative Heat shock 70 kDa protein 7, Heat shock cognate 71 kDa). Also, proteins related to carbohydrate metabolic process and gly-colytic and glucose metabolic process were absent in SS, mainly from OP and OWP (i.e., Fru-tose-bisphosphate aldose A, Glusoce-6-phosphate isomerase, Pyruvate kinase). Saliva stimulation decreased important proteins involved with immune response and inflammation process in all groups. Unstimulated salivary samples seem to be the best choice to proteomic approach in pregnant women.

Project description:Aim: This study aimed to analyze the salivary proteomic profile related to obesity and periodontitis in women during pregnancy and after delivery. Materials and methods: The sample assisted during the 3rd trimester of pregnancy (T1) and after childbirth (T2) was divided into: with obesity and periodontitis (OP = 8); with obesity and without periodontitis (OWP = 10); with normal BMI but with periodontitis (NP = 10); with normal BMI and without periodontitis (NWP = 10). The proteins were extracted and processed individually by label-free proteomics (nLC-ESI-MS/MS). ProteinLynx GlobalServer software was used to process and search the data, and the proteins were analyzed by their access number by UniProt catalog. Results: The up-regulation of Heat shock protein (HSP) 70 kDa 1A, 1B and 1-like were related to both obesity and periodontitis, separately. Albumin and thioredoxin were up-expressed in periodontitis cases, while Cystatins (mainly S, SA, SN) and Lactotransferrin were down-expressed. The high abundance of Submaxillary gland androgen-regulated protein 3B, S100A8, Matrix metalloproteinase-9 (MMP9), HSP 70 kDa 2 and 6, Putative HSP 70 kDa 7, HSP 71 kDa, Haptoglobin and Plastin-1 were significant in the combination of obesity and periodontitis. Conclusion: Obesity and periodontitis remarkably altered the proteome of the saliva during pregnancy with substantial alterations after delivery. Proteins related to humoral immune response and antibacterial humoral response were highly expressed in women with periodontitis, while proteins related to glycolytic, carbohydrate catabolic, glucose metabolic, organophosphate metabolic and carbohydrate derivative metabolic processes were more expressed in individuals with obesity.
