Suboptimal prophylactic treatments with BDGR-49 in a lethal VEEV TC-83 mouse model

Project description:To study emergence of resistance to BDGR-49 during infection with Venezuelan equine encephalitis virus (VEEV), mice were treated suboptimal treatment regimen. C3H/HeN mice were prophylactically treated with BDGR-49 2 hours prior to infection for a total of 5 days. Mice were infected with a lethal challenge dose of VEEV TC-83. Following infection mice brains were euthanized on 3-, 5-, 7,9-, and 10-days post-infection and their brains collected and RNA-Seq performed on. RNA-Seq results were used to analyze for viral resistant mutants. Analysis revealed that only one of the mice contained a mutant that is potentially resistant to BDGR-49.

Characterization of young and aged lung tissue RNA 60 days post-influenza infection

Project description:To identify molecular characteristics of young and aged lungs post-influenza infection, we isolated RNA from lungs 60 days post-infection and examined by bulk RNAseq. We found a large number of genes remain upregulated in aged compared to young lungs. Comparison with expression of immune system-related genes from Nanostring expriments indicated this was largely infection-induced rather than baseline differences between age. This indicates that aged lungs fail to return to homeostasis following a viral respiratory infection

NanoString nCounter assay was performed to measure the expression of neuroinflammatory genes in the brains of cPIIRS mice (C57BL/6Tac, 21 day post-infection) compared to cPIIRS mice treated with Ruxolitinib, and naïve mice.

Real-time quantitative PCR analysis of inflammation-related genes of organs and tissues from mice in response to Coxsackievirus A2 (CVA2) infection

Project description:For in vivo experiments, mice were inoculated a lethal dose of CVA2 strain (104 TCID50/mouse) or equal volume of culture supernatant of RD cells that were uesd to cultivate virus. Total RNA was extracted from brains, lungs, hearts and skeletal muscle of mice at 3 days post infection (dpi), 5 dpi and corresponding controls using TRIzol reagent to quantitate inflammation-related genes from the CVA2 infected mice and control. qPCR gene expression profiling. Brains, lungs, hearts and skeletal muscles from four groups were treated separately. Equal amount total RNA from each organ or tissue of mice was harvested to analyze inflammation-related genes.

Project description:Mucosal-Associated Invariant T (MAIT) cells are an innate-like subset of alpha/beta-T cells expressing semi-invariant TCRs that recognize conserved vitamin B2 metabolite-based antigens from microbes, thus contributing to protection against several bacterial pathogens. To investigate the transcriptomic changes induced during systemic Francisella tularensis infection, liver MAIT cells were recovered from naïve mice and mice at days 13 and 48 post infection.

Differential cellular transcriptional profiles during virus-induced neuroinflammation identified new microglial- and macrophage-specific genes

Project description:Goal: Determine differences in gene expression profile between microglia and macrophages isoalted from the brains of mice during viral-induced neuroinflammation. Methods: microglia and macrophages were obtained by FACS cells sorting (based on CD45 and CD11b level of expression) from brains of C57BL/6J mice, 6 days post i.c infection with TMEV or injection with PBS. RNA was extracted with TRYzol and precipitated using glycogen. polyA mRNA was enriched using oligo d(T)25 magnetic beads Smarter Stranded RNA-Seq Kit was then utilized to prepare cDNA library for RNA-seq. Results: Using RNA-seq and later RT-qPCR to validate our candidates, we found genes that are speifically expressed by activated microglia or infiltrating macrophages during viral-induced neuroinflamamtion

Project description:Seasonal influenza causes significant morbidity and mortality worldwide, with 70-90% of these deaths occurring in individuals aged 65 or older. To explore the mechanisms behind this age-related vulnerability, we investigated immune responses and inflammation in the lungs of young and old mice during influenza A virus (IAV) infection. Young (12-week) and old (70-week) C57BL/6J mice were intranasally infected with IAV PR8, and immune responses were assessed at 7 days post-infection (dpi) using single-cell RNA sequencing (scRNA-seq).

Spatial Transcriptomics of Brains Infected with VEEV

Project description:Venezuelan equine encephalitis virus (VEEV) is a neurotropic alphavirus that causes neurological disease in both humans and equine. In this study we paired spatial transcriptomics and a mouse model of VEEV to generate a transcriptomic profile of infected or control brains at 6 days post infection. We identified regionally distinct myeloid and lymphocyte populations, contributing to our understanding of the neuroinflammation caused by VEEV.
