Transcription profiling of pig jejunum from animals fed an animal or vegetable protein diet

ABSTRACT: This study examines the evidence that dietary can regulate genes in the intestinal epithelium and it discusses the physiologic relevance of such alterations in gene expression. Experiment Overall Design: Seven RNA samples were analyzed using Affymetrix GeneChipÂ® Porcine Genome Array (Affymetrix, Santa Clara, CA, USA) containing 23,937 probe sets that interrogate approximately 23,256 transcripts from 20,201 S. scrofa genes.

Using Genome Arrayï¼We generated transcriptase profiles of intestine from the Landrace piglets at 0, 3, 8, 14ï¼21 days post natal (dpn). We identified 4014 differentially expressed transcripts during intestinal developing in Landrace piglets. Respectively, the Gene Ontology processes and the time sequence profile analysis of differentially expressed genes were constructed. In the course of growth , the mostly significant enriched GO category of profile mainly focus on folic acid transport, lipopolysaccharide-mediated signaling pathway, Momolybdopterin cofactor biosynthetic process,The down-regulation genes show the state of malnutritionplay at early growth in pig life. Project description:An early settlement of a complex gut microbiota can protect against gastro-intestinal dysbiosis, but the effects of neonatal microbiota colonization on the gut barrier upon the further encounter of favorable bacteria or not, are largely unknown. The jejunal transcriptome of differently perfused intestinal loops of 12 caesarian-derived pigs previously associated with microbiota of different complexity was studied. Pigs received pasteurized sow colostrum at birth (d0), 2 mL of starter microbiota (10^7 CFU of each Lactob. Amylovorus (LAM), Clostr. glycolicum, and Parabacteroides spp.) on d1-d3 of age and either a placebo inoculant (simple association, SA) or an inoculant consisting of diluted feces of an adult sow (complex association, CA) on d3-d4 of age. On days 26-37 of age, jejunal loops were perfused for 8 h with either enterotoxigenic E. coli F4 (ETEC), F4 fimbriae (F4), LAM or saline (CTRL) and jejunal samples were obtained from each piglet immediately afterwards. Association of Vegetable and Animal Flesh Intake with Inflammation in Pregnant Women from India.

Project description:In pregnant women, studies are lacking on the relationship of vegetable and animal flesh (poultry, red meat and seafood) intake with inflammation, especially in low- and middle-income countries. We conducted a cohort study of pregnant women receiving antenatal care at BJ Medical College in Pune, India. The dietary intake of pregnant women was queried in the third trimester using a validated food frequency questionnaire. Twelve inflammatory markers were measured in plasma samples using immunoassays. Only 12% of the study population were vegetarians, although animal flesh intake levels were lower compared to Western populations. In multivariable models, higher intakes of total vegetables were associated with lower levels of the T-helper (Th) 17 cytokine interleukin (IL)-17a (p = 0.03) and the monocyte/macrophage activation marker soluble CD163 (sCD163) (p = 0.02). Additionally, higher intakes of poultry were negatively associated with intestinal fatty-acid binding protein (I-FABP) levels (p = 0.01), a marker of intestinal barrier dysfunction and Th2 cytokine IL-13 (p = 0.03), and higher seafood was associated with lower IL-13 (p = 0.005). Our data from pregnant women in India suggest that a higher quality diet emphasizing vegetables and with some animal flesh is associated with lower inflammation. Future studies should confirm these findings and test if modulating vegetables and animal flesh intake could impact specific aspects of immunity and perinatal health.

An RNA Splicing System that Excises DNA Transposons from Animal mRNAs

Project description:All genomes harbor mobile genetic parasites called transposable elements (TEs). Here we describe a system, which we term SOS splicing, that protects C. elegans and human genes from DNA transposon-mediated disruption by excising these TEs from host mRNAs. SOS splicing, which operates independently of the spliceosome, is a pattern recognition system triggered by base- pairing of inverted terminal repeat elements, which are a defining feature of the DNA transposons. We identify three factors required for SOS splicing in both C. elegans and human cells; AKAP17A, which binds TE-containing mRNAs; the RNA ligase RTCB; and CAAP1, which bridges RTCB and AKAP17A, allowing RTCB to ligate mRNA fragments generated by TE excision. We propose that SOS splicing is a novel, conserved, and RNA structure-directed mode of mRNA splicing and that one function of SOS splicing is to genetically buffer animals from the deleterious effects of TE- mediated gene perturbation.
