Transcription profiling of pig jejunum from animals fed an animal or vegetable protein diet

ABSTRACT: This study examines the evidence that dietary can regulate genes in the intestinal epithelium and it discusses the physiologic relevance of such alterations in gene expression. Experiment Overall Design: Seven RNA samples were analyzed using Affymetrix GeneChipÂ® Porcine Genome Array (Affymetrix, Santa Clara, CA, USA) containing 23,937 probe sets that interrogate approximately 23,256 transcripts from 20,201 S. scrofa genes.

Project description:This paper describe the whole genome-wide analysis of differences in postnatal intestinal from 0 day to 21 days of piglets. Using Genome Arrayï¼We generated transcriptase profiles of intestine from the Landrace piglets at 0, 3, 8, 14ï¼21 days post natal (dpn). We identified 4014 differentially expressed transcripts during intestinal developing in Landrace piglets. Respectively, the Gene Ontology processes and the time sequence profile analysis of differentially expressed genes were constructed. In the course of growth , the mostly significant enriched GO category of profile mainly focus on folic acid transport, lipopolysaccharide-mediated signaling pathway, Momolybdopterin cofactor biosynthetic process,The down-regulation genes show the state of malnutritionplay at early growth in pig life. Early microbial colonisation affects transcriptome of pig jejunum perfused with E. coli F4, F4 fimbria or Lact. amylovorus

Project description:An early settlement of a complex gut microbiota can protect against gastro-intestinal dysbiosis, but the effects of neonatal microbiota colonization on the gut barrier upon the further encounter of favorable bacteria or not, are largely unknown. The jejunal transcriptome of differently perfused intestinal loops of 12 caesarian-derived pigs previously associated with microbiota of different complexity was studied. Pigs received pasteurized sow colostrum at birth (d0), 2 mL of starter microbiota (10^7 CFU of each Lactob. Amylovorus (LAM), Clostr. glycolicum, and Parabacteroides spp.) on d1-d3 of age and either a placebo inoculant (simple association, SA) or an inoculant consisting of diluted feces of an adult sow (complex association, CA) on d3-d4 of age. On days 26-37 of age, jejunal loops were perfused for 8 h with either enterotoxigenic E. coli F4 (ETEC), F4 fimbriae (F4), LAM or saline (CTRL) and jejunal samples were obtained from each piglet immediately afterwards. 5 Day Oral Study of A-998679 in Male Sprague Dawley Rats (Jejunum)

Project description:Male Sprague-Dawley rats [Crl:CD®(SD)IGS BR], weighing ~250 g at study initiation were obtained from Charles River Laboratories, Inc. (Wilmington, MA). Rats were housed singly in ventilated, stainless steel, wire-bottom hanging cages and fed non-certified Rodent Chow (Harlan Labs, Madison, WI) and water ad libitum and acclimated for at least 5 days after arrival. Rats were randomly assigned to various treatment groups (3 rats/group) and were dosed once daily by oral gavage with vehicle (0.2% hydroxypropylmethylcellulose at a dose volume of 10 ml/kg) or with 30, 100, or 200 mg/kg of A-998679. All rats were fasted overnight after their last dose, weighed and sacrificed under isoflurane anesthesia. Liver and small intestine (jejunum) were flash frozen in liquid nitrogen and stored at −80°C until processing for gene expression profiling on the Affymetrix platform. total of 11 samples, 3 biological replicates (rats) from each of 3 treatments groups 1. vehicle control 2. 30mg/kg A-998679 3. 100mg/kg A-998679 and 2 biological replicates from treatment group 4. 200mg/kg A-998679

Expression data from proximal jejunum of WT vs. TLR2 KO mice after methotrexate (MTX) treatment

Project description:The role of innate immunity in modulating severity of chemotherapy-induced complications is so far unclear. The aim of this study was to determine how TLR2 may influence MTX-induced mucositis in the small intestine in mice. We used microarrays to assess gene expression profiles in proximal jejunum of WT vs. TLR2 KO mice after systemic treatment with MTX. Mucositis was induced by i.p. injection of MTX [40mg/kg BW/d] for 4 days in WT or TLR2 knockout (KO) mice. On day 7, mice were sacrificed and RNA was extracted from proximal jejunum (n=3 mice/group) and hybridized on Affymetrix microarrays.
