Project description:This data analyzes gene expression patterns in Arabidopsis whitening mutants, wn1, with defects in chloroplast development. We found that wn1 was a T-DNA insertion line of Arabidopsis AtPTAC10/PAP3 (At3g48500) and exhibited a lethal, albino-like phenotype in the seedling stages. pTAC10 was known as a component of RNA polymerase complex called plastid-encoded RNA polymerase, PEP, in mature chloroplasts. Because wn1 mutants were lethal, we extracted RNA from cotyledons of 7-day-old seedlings from wn1 mutants and Col-0. RNA seq analysis was commissioned by NICEM in Seoul National University, Korea and transcriptome resequencing was confirmed in Macrogen, Korea. Through analysis, we compared gene expression patterns of wn1 mutant and Col-0 seedlings.

Project description:We used microarrays to detail the global programme of gene expression underlying cellularisation and identified distinct classes of up-regulated genes in fbn5-1 T-DNA insertion Arabidopsis mutant. Fibrillins are lipid-associated proteins in plastids and are ubiquitous in plants. Fibrillins are known to accumulate and sequester carotenoids in chromoplasts during the development of flowers and fruits; however, little is known about the function of fibrillins in leaf tissues. Here we identified a novel protein fibrillin5 (FBN5) that is essential for plastoquinone-9 (PQ-9) biosynthesis in Arabidopsis thaliana. We showed that homozygous fbn5-1 plants were seedling-lethal, and XVE:FBN5-B transgenic plants expressing low levels of FBN5-B showed a lower growth rate and were smaller than wild-type. In chloroplasts, FBN5-B physically interacts with solanesyl-diphosphate synthase (SPS) 1 and 2, which synthesize the solanesyl moiety of PQ-9. Plants containing defective FBN5-B accumulated less PQ-9 and its cyclized product, plastochromanol-8, which is consistent with mutant phenotypes showing lower photosynthetic performance and higher levels of hydrogen peroxide. The role of FBN5-B in the FBN5-B/dimeric SPS complex is discussed with respect to biosynthesis of the solanesyl moiety. 4 replicats of wild-type as control and fbn5-1 T-DNA insertion mutants were analized.

Project description:Aim: To identify regulatory factors that control: (1) chloroplast protein importand (2) chloroplast-to-nucleus signalling. This project is a joint proposal from the Jarvis lab which is interested in chloroplast protein import [1] and the Moller lab which is interested in plastid-to-nucleus signalling [2]. Background: The majority of chloroplast proteins are encoded in the nucleus and imported post-translationally into chloroplasts. The abundance of chloroplast proteins may therefore be regulated at multiple levels. It is well documented that the nuclear gene expression is responsive to (largely unknown) signals from the chloroplast [23] and evidence is now emerging that protein import is also a regulated process [1]. Protein import into chloroplasts is mediated by protein complexes in the outer and inner envelope membranes called Toc and Tic respectively. Biochemical studies of pea chloroplasts identified several Toc/Tic components. These proteins are mechanistic or structural components of the import apparatus. Arabidopsis homologues of the pea Toc/Tic proteins were identified by the AGI. Pea Toc34 is represented in Arabidopsis by two genes "Toc33 and Toc34" and pea Toc75 is represented by three genes. These different Tocs have different expression patterns and are proposed to have different precursor protein recognition specificities. The factors that regulate Toc expression in concert with the needs of plastids in developmentally different cells are unknown. Proposal: Two Arabidopsis mutants will be analysed. The ppi1 mutant is null for the putative precursor protein receptor Toc33 [1]and the ppi3 mutant is null for a putative component of the protein import channel Toc75-IV (on chromosome IV). ppi1 plants are yellow-green in appearance but remarkably healthy and grow only slightly more slowly than wild type. Project description:Amino acids (AA) are essential molecules for life because they are precursors for the synthesis of proteins and other organic compounds. Their metabolism in plants involves more than one cellular compartment, among which the chloroplast plays a key role. From the 20 essential AA, ten (Arg, Lys, Thr, Leu, Ile, Val, Trp, Phe, Tyr and His) are only synthesized in the plastids and seven (Asp, Cys, Gln, Glu, Gly, Ser and Met) can be synthesized in both chloroplast and other parts of the cell. Despite knowing the roles of AA in plant physiology, little has been assessed in plants with albino phenotypes, lacking chloroplasts. To assess the effect of chloroplast biogenesis disturbances on AA metabolism in albino and variegated somaclonal variations of Agave angustifolia Haw., a comparative proteomic based TMT- synchronous precursor selection (SPS)-MS3 strategy was used to determine the status of AA biosynthetic pathways. Besides the 20 essential AA was quantified to underpin proteomics finding. A total of 2,442 different proteins were identified in the proteome of the somaclonal variants, from which 82 correspond to enzymes participating in AA biosynthesis. From these 82, 32 proteins were differentially accumulated. On the other hand, the concentration of 16 and 12 of the AA synthesized by the chloroplast were over-accumulated in the variegated and albino somaclonal variant, respectively. In plantlets devoid of functional chloroplasts, there is a surprising increase of AA and key enzymes of the AA biosyntethic pathways, compared to green plantlets. This could suggest que la activation of AA biosynthesis is key to sustaining albino and variegated plantlets survival that lack chloroplasts.

Project description:Genetic recoding, a process that modifies genome templated protein sequence, plays an important role especially in the small genomes of viruses and bacteria. Chloroplasts have evolved from cyanobacterial ancestry and mainly retain the prokaryotic gene expression machinery. I-iota, a plastome mutant of Oenothera (the evening primrose), carries a single adenine insertion in a polyA stretch near to the 5’-end of the atpB gene, encoding the β-CF1 subunit of the ATP synthase This causes formation of a truncated protein. Surprisingly, a full-length AtpB protein is detectable in I-iota leaves, suggesting the presence of genetic recoding operating in I-iota. In order to analyze this phenomenon in more detail, several tobacco transplastomic lines were generated inducing and destroying genetic recoding of atpB. We identified that a two adenine (+2A) insertion was more efficiently compensated in plants than +1A, -1A or -2A mutations at the same location. Furthermore, a homopolymeric composition of the polyA stretch is essential for successful recoding: plants carrying a destroyed polyA stretch have an albino-phenotype, indicating the absence of genetic recoding. Here we show evidence for genetic recoding acting in the chloroplast that plays presumablya compensatory function and enables correction of indels in an essential gene for autotrophic growth.

Project description:Survival of plants depends on their ability to adapt to ever-changing environmental conditions. Chloroplasts sense different stimuli and respond to ion fluxes in the cytosol triggering systemic responses under stress conditions. Indeed, changes in calcium and magnesium ion concentration in the chloroplast stroma impact the immune response against pathogens and adapt the photosynthetic machinery under fluctuating light conditions. Here we investigated the link between di-valent cations, calcium, magnesium and manganese and the protein kinase activity in Arabidopsis chloroplasts. Our results show that overall, manganese ions are the strongest activator of kinase activity in chloroplasts followed by magnesium ions, whereas calcium ions have seemingly no effect. Additionally, when comparing the phosphorylation pattern from Arabidopsis wild type and a cmt1 mutant, which is defective in manganese import into chloroplasts, the phosphorylation of specific protein bands is strongly reduced in the mutant chloroplasts supporting the notion that chloroplasts contain manganese-dependent protein kinases. These findings provide insights for the future characterization of chloroplast protein kinases activity regarding in-vitro assays and potential target proteins.

Project description:The effect Ds insertion mutation in Ds13-2198-1 line on the gene expression profiles was investigated. The genes for photosynthesis and some transcriptional factors were upregulated while genes for metabolism were downregulated. Experiment Overall Design: To investigate the effect of an strong albino mutation, the genome wide gene expression profiles of plants were examined. Segregants with normal phenotype and albino phenotype obtained from the heterotalic parental line were grown on the same 1/2 MS plate for 3 weeks.
