Role of Cryptococcus neoformans Sre1-pathway in low oxygen gene expression


ABSTRACT: Comparison of wild-type serotype A C. neoformans cells grown at 21% and 3% oxygen for 2 hours in rich medium. Also comparison of wild-type and Sre1-pathway mutant strains grown at 3 % oxygen for 2 hours in rich medium. Two-condition experiment. Each comparison was performed in two biological replicates, each containing duplicate probes/array (=4 data points total per comparison).

In the human fungal pathogen Cryptococcus neoformans, the SREBP orthologue Sre1 is important for adaptation and growth in nutrient-limiting host tissues. In this study, we characterize the C. neoformans serotype A Sre1 and its activating protease, Stp1. We demonstrate that Stp1 is a functionally conserved orthologue of the mammalian Site-2 protease and that Stp1 cleaves Sre1 within its predicted first transmembrane segment.
