Gene expression alteration after RBSDV infection

ABSTRACT: We investigated the alterations of genes expression in maize infected by RBSDV, and detected about 4000 genes alterations which were significant according to the identification standards influenced by RBSDV infection, including resistance related genes, cellulosesynthesis and development related genes. Expression alterations was varified in the same RNA samples by RT- PCR. Two sets samples, RBSDV-infected vs mock; Biological replicates: 4 RBSDV-infected, 4 mock; independently inoculated and harvested.

Proteomics of non-host response in barley

Project description:Nonhost resistance, a resistance of plant species against all nonadapted pathogens, is considered the most durable and efficient immune system in plants. To increase our understanding of the response of barley (Hordeum vulgare L.) plants to infection by powdery mildew, Blumeria graminis f. sp. Tritici, we used quantitative proteomic analysis (LC-MS/MS). We compared the response of two genotypes of barley cultivar Golden Promise, wild type (WT) and plants with overexpression of phytoglobin (previously hemoglobin) class 1 (HO), which has previously been shown to significantly weaken non-host resistance. A total of 8804 proteins were identified and quantified, out of which the abundance of 1044 proteins changed significantly in at least one of the four comparisons (‘i’ stands for ‘inoculated’)- HO/WT and HOi/WTi (giving genotype differences), and WTi/WT and HOi/HO (giving treatment differences). Among these differentially abundant proteins (DAP) were proteins related to structural organization, disease/defense, metabolism, transporters, signal transduction and protein synthesis. More proteins changed in abundance in WT than in HO after inoculation and most DAP increased in abundance.

Transcriptome profiling of Pythium graminicola-inoculated rice roots.

Project description:Contrary to the relative wealth of information regarding pathogen defense responses in aboveground plant parts, little is known about the mechanistic basis and regulation of plant immunity in root tissues. Aiming to further our fundamental understanding of root immune responses, we have investigated the interaction between rice and one of its major root pathogens, the oomycete Pythium graminicola. The specificic objectives of this study were twofold: i) to disentangle the molecular and genetic basis of the rice-Pythium interaction by comparing the transcriptome of rice roots at different times after inoculation with a highly virulent Pythium strains, and ii) to offer fundamental insights into the genetic architecture and regulation of rice disease resistance pathways operative in root tissue and to identify the molecular players controlling the possible nodes of convergence between these resistance conduits Comparison between P. graminicola- and mock-infected rice roots. Project description:This SuperSeries is composed of the following subset Series: GSE13254: Sclerotinia infected vs Mock infected controls in B. napus (Westar) GSE13256: Zhong You 821 Sclerotinia infected vs Mock infected controls in B. napus (Zhong You 821) The fungal pathogen Sclerotinia sclerotiorum infects a broad range of dicotyledonous plant species and is the causative agent of stem rot in Brassica napus. To elucidate the mechanisms underlying the defense response, we studied the patterns of gene expression in a partially resistant variety of ZhongYou 821 (ZY821) and a susceptible line from Westar over five time points, 6, 12, 24, 48, and 72 hours post-inoculation (hpi) using a B. napus oligonucleotide microarray. For each cultivar, a two-dye experiment was run comparing infected to mock-infected stem tissue. Identification of early response genes to Pratylenchus coffeae in resistant ramie by digital gene expression analysis

Project description:Digital gene expression (DGE) analysis was performed to study differentially expressed genes (DEGs) between CK and P. coffeae challenged (CH) roots of ramie. A total of 10.16 and 8.07 million clean reads were detected in the CK and CH libraries, respectively. A total of 137 genes were identified as DEGs, 117 and 20 of them were up- and down- regulated respectively, in response to P. coffeae infection. Roots samples of Pratylenchus coffeae infected(E2) and un-infected (E1) ramie were RNA-Seq sequenced to compare differented expressed genes.

Transcription profiling of Arabidopsis thaliana six days after infection with Agrobacterium tumefaciens

Project description:This study focuses on responses of the host plant to infection and transformation with Agrobacterium tumefaciens. Genome wide changes in gene expression were integrated with the alterations in metabolite levels six days after inoculation of agrobacteria. Plants were infected with the virulent strain C58, harboring a T-DNA, or with strain GV3101, containing a disarmed Ti-plasmid. This allows discrimination between signals which derive from the bacterial pathogen and the T-DNA encoded genes. Experiment Overall Design: The bases of Arabidopsis thaliana (WS-2) inflorescence stalks were wounded and immediately infected with Agrobacterium tumefaciens or mock-infected for six days. Stalks of intact plants were inoculated with the oncogenic strain C58 (C58 6dpi 1 to 3) or the non-virulent strain GV3101 (GV3101 6dpi 1 to 3) to provide conditions close to nature. The gene expression data of three independent experiments of infected material were compared with three non-infected samples (reference 6dpi 1 to 3). Differential gene expression was determined by applying the LIMMA package (Linear Models for Microarray Data; Smyth, G.K. (2004) Applic. Genet. Mol. Maize gene expression after infection of Ustilago maydis SG200 and SG200M-NM-^Ttin2

Project description:Anthocyanin induction in plant is considered a general defense response against biotic and abiotic stresses. The infection by Ustilago maydis, the corn smut pathogen, is accompanied with anthocyanin induction in leaf tissue. We revealed that anthocyanin is intentionally induced by the virulence promoting secreted effector protein Tin2. Tin2 protein functions inside plant cells where it interacts with cytoplasmic maize protein kinase ZmTTK1. Tin2 masks an ubiquitin-proteasome degradation motif in ZmTTK1 leading to a more stable active kinase. Active ZmTTK1 controls transcriptional activation of genes in the anthocyanin biosynthesis pathway rerouting phenylalanine away from lignin biosynthesis. Therefore, we performed microarray analysis to understand how maize gene transcription in phenylpropanoid pathway is differentially changed after infection with Ustilago maydis SG200 (wild type) and SG200Dtin2 (anthocyanin-inducing effector mutant). We prepared three biological replicates for mock-inoculated maize (control), SG200-infected maize and SG200M-NM-^Ttin2-infected maize. For 1 sample, we harvested the leaves (1-3cm below injection hole) from 20 plants and pooled them. At 4 days post inoculation, total RNA was extracted.
