CD4+T cells activation

ABSTRACT: Aanlysis of human resting CD4+T cells and cells activated by two different methods: CD4+ T cells unstimulated (be susceptible but not support to HIV-1) and stimulated either by CD3/CD28 costimulation (reversed susceptibility and resisted to HIV-1) or by PHA/IL-2 for six days (be susceptible and support to HIV-1) to investigate potential mechanism of reversing susceptible to HIV-1. We measured gene expression in resting human CD4+T cells and cells activated by two methods for six days which could induce different effects to HIV-1. Three independent experiments were performed at each treatments using different donors for each experiment.

Substrate induced gene expression was measured when culture was grown on glucose, fructose, mannose, galactose, arabinose, xylose, galactomannan, xylan and CMC to mid-logarithmic phase.

Global Microarray Analysis of Bacillus sp. N16-5 Salt Stress Adaptation

Project description:The alkaliphilic halotolerant bacterium Bacillus sp. N16-5 often faces salt stress in its natural habitats. One-color microarrays was used to investigate transcriptome expression profiles of Bacillus sp. N16-5 adaptation reactions to prolonged grown at different salinities (0%, 2%, 8% and 15% NaCl) and the initial reaction to suddenly alter salinity from 0% to 8% NaCl. Salt induced gene expression was measured when culture was grown on different salinities (0%, 2%, 8% and 15% NaCl) to mid-logarithmic phase. And salt induced gene expression was also measured at 0 min, 10 min, 30 min, 60min, 120min after a sudden change salinity from 0% to 8% NaCl.

Determination of Sigma 54 reglons in Bacillus thuringiensis HD73 strain

Project description:To determine Sigma 54 (SigL) reglons in Bacillus thuringiensis HD73 strain, A sigLmutant, HD(M-NM-^TsigL::kan), was constructed with insertion of kanamycin resistance gene cassete. We have employed whole genome microarray expression profiling as a discovery platform to identify the difference of gene expression between mutant and wild-type strains. 2 ml samples were separately harvested from B. thuringiensis HD73 and HD(M-NM-^TsigL::kan) strains grown in SchaefferM-bM-^@M-^Ys sporulation medium (SSM) at stages T7 of stationary phase (7 hours after the end of the exponential phase). Three independent repeats were performed for each stain.

Circulating miRNAs spectra in plasma of multiple myeloma patients compared with healthy individuals

Project description:MicroRNAs (miRNAs) are a class of small non-coding single-stranded RNAs whose dysregulation of expression plays an important role in cancer development. Circulating miRNAs are novel biomarkers in several cancers. Thus, we explored whether the miRNAs in plasma could be useful clinical biomarkers for multiple myeloma (MM) patients. The expression levels of four miRNAs in plasma were upregulated while eight miRNAs were downregulated in MM patients compared with healthy controls according to microarray. MiRNA microarray was conducted to determine deregulated miRNAs in plasma of 9 MM patients and 7 healthy controls.
