Time-course in rice leaves of the temperature shift experiment

ABSTRACT: Time-course transcriptional profiling of rice leaf of the temperature shift experiment in a growth chamber. This experiment was performed to validate the results of field transcriptomic modeling. Rice leaves (cv. Norin8) were collected at 2:00 and 14:00 on the day -1, 2, 4 and 6 from the temperature shift (diurnal cycle temperature condition to continuous temperature condition). Two biological replicates for each sampling time point.

Project description:Time-course transcriptional profiling of rice leaf in the field in 2009. This experiment was performed to validate the results of field transcriptomic modeling. Using 461 field transcriptome data obtained in 2008 (GSE36040; GSE36042; GSE36043; GSE36044; GSE18685) and the corresponding meteorologicla dara, we perfomred statistical modeling of transcriptome. Rice leaves (Norin8 vs. osgi) in the paddy field were collected on Aug. 10 - 12, 24 - 25, 31, Oct. 8 - 9 in 2009.

Project description:Tissue-specific transcriptional profiling of the abscission layer (AL) at the base of young flower in rice using laser micro-dissection: NIL(qSH1) vs. Nipponbare. We used two rice varieties, NIL(qSH1) and Nipponbare. NIL(qSH1) is a nearly isogenic line containing the seed shattering gene qSH1. Seed shattering is easy in NIL(qSH1), but it is not in Nipponbare. So, we used some stages of young flower in NIL(qSH1) and some in Nipponbare. Four regions: 1. abscission layer region of NIL(qSH1), 2. upper abscission region of NIL(qSH1), 3. lower abscission layer region of NIL(qSH1), and 4. abscission layer region of Nipponbare. Sample experiments: NIL(qSH1) AL vs. Nipponbare AL, NIL(qSH1) AL vs. NIL(qSH1) upper region of AL, and NIL(qSH1) AL vs. NIL(qSH1) lower region of AL.

OsNOA1 functions in a threshold-dependent manner to regulate chloroplast proteins in rice at lower temperatures

Project description:While our earlier microarray analysis identified a number of possible candidate genes, which may be involved in OsNOA1 function (Yang et al., 2011a), a complementary study of the rice proteome profiles under the same condition has the potential to yield new insights into the biological functions of NOA1 and molecular mechanism of NOA1 associated regulation. In this study, we utilized previously-generated OsNOA1 RNAi lines (Yang et al., 2011a) and newly-generated OsNOA1 overexpression lines to further investigate the function and mechanism of NOA1 in rice. Here we report the proteome changes which include important candidate proteins involved in chloroplast ribosome biogenesis and several classes of proteins with significant changes in response to various levels of OsNOA1 under two different temperature conditions. Our results suggest that OsNOA1 can function in a threshold-dependent manner to regulate the biosynthesis of chloroplast proteins in rice at lower temperatures, and this regulation may be achieved via interactions between NOA1 and chloroplast ribosomes.

Transcription profiling by array of rice plants grown in different light and temperature conditions

Project description:Rice (Oryza sativa, spp. Indica, cv. 93-11) plants were grown in a Conviron PGR 15 growth chamber using precise control of temperature, light, and humidity.<br>Diurnal (driven) conditions included 12L:12D light cycles and 31C/20C thermocycles in three different combinations. These were: photocycles (LDHH), 12 hrs. light (L)/12 hrs. dark (D) at a constant temperature (31C; HH); photo/thermocycles (LDHC): 12 hrs. light (L) /12 hrs. dark (D) with a high day temperature (31C) and a low night temperature (20C); and thermocycles (LLHC): continuous light (LL) with 12 hrs. high/12 hrs. low temperature (31C, day; 20C, night). Light intensity and relative humidity were 1000 micromol m-2s-2 and 60%, respectively.<br>Three-month-old rice plants were entrained for at least one week under the respective condition prior to initiation of each experiment. Leaves and stems from individual rice plants were collected every four hours for 48 hrs in driven (diurnal) conditions followed by a two day freerun spacer under continuous light/temperature followed by two additional days of sampling under the same continuous free run condition.
