Comparison of gene change in lymphoma cells after co-culture with endothelial cells

ABSTRACT: Mouse lymphoma cells were co-cultured with endothelial cells in serum/cytokine-free condition. To identify specific genetic changes, we compared lymphoma cells cultured in medium containing 10% fetal bovine serum with lymphoma cells co-cultured with endothelial cells. The experiment compared genetic change in lymphoma cells caused by co-culture with endothelial cells.

Project description:Kaposi sarcoma is the most common cancer in AIDS patients and is typified by red skin lesions. The disease is caused by the KSHV virus (HHV8) and is recognizable by its distinctive red skin lesions. The lesions are KSHV infected spindle cells, most commonly the lymphatic endothelial and blood vessel endothelial cells (LEC and BEC), plus surrounding stroma. Here we examine KSHVs modulation of Notch signaling using wild-type LEC cells co-cultured with DLL4 and JAG1 expressing LEC cells. Experiment Overall Design: There are 1) n=2 of LEC control cells co-cultured with pSIN expressing LEC, 2) n=3 of LEC co-cultured with pSIN-Dll4 expressing LEC, and 3) n=3 of LEC co-cultured with pSIN-JAG1 expressing LEC.

Comparison of human embroynic stem cell derived vascular cells to mature human vascular and hematopoietic cells

Project description:The pathways involved in hierarchical differentiation of human embryonic stem cells (hESC) into abundant and durable endothelial cells (EC) are unknown. We employed an EC-specific VE-cadherin promoter driving GFP (hVPr-GFP) to screen for factors that augmented yields of vascular-committed ECs from hESCs. In phase 1 of our approach, inhibition of TGFb, precisely at day 7 of hESC differentiation, enhanced emergence of hVPr-GFP+ ECs by 10-fold. In the second phase, TGFb-inhibition preserved proliferation and vascular identity of purified ECs, resulting in net 36-fold expansion of homogenous EC-monolayers, and allowing transcriptional profiling that revealed a unique angiogenic signature defined by the VEGFR2highId1highVE-cadherin+EphrinB2+CD133+HoxA9- phenotype. Using an Id1-YFP hESC reporter line, we showed that TGFb-inhibition sustained Id1 expression in hESC-derived ECs, which was required for increased proliferation and preservation of EC commitment. These data provide a multiphasic method for serum-free differentiation and long-term maintenance of authentic hESC-derived ECs, establishing clinical-scale generation of transplantable human ECs. The experiment compared 6 sets of biological duplicates which included human embryonic stem cell derived and mature vascular cell types.

Project description:We studied the crosstalk between tumor and endothelial cells to explore the role of tumor microenvironment on cancer growth and progression. As part of our investigation, we showed that contact-dependent interaction of endothelial cells with breast tumor cells triggered the differential expression of a large number of genes in endothelium. Endothelial and MDA-MB231 cells were co-cultured together under serum- cytokine-free environment for 5 days followed by sorting endothelial cells and extracting RNA for microarray analysis. Role of TGFb1 and Wnt6 in FGF2 and BMP4-driven endothelial differentiation of murine embryonic stem cells [3h]

Project description:In this work we investigated the molecular mechanisms that sustain the endothelial differentiation of murine embryonic stem cells (ES). When ES cells are co-cultured with the stromal PA6 cells in serum-free medium they differentiate mainly into neurons, thanks to the neural inducing activity exerted by the stroma. The addition of exogenous FGF2 allows also the differentiation of endothelial cells whereas, in presence of exogenous BMP4, ES cells differentiate exclusively into endothelium. A pivotal role in ES cells differentiation is played by the stromal PA6 cells.To analyze the early signals released by PA6 cells in the medium of the co-cultures in response to FGF2 and BMP4, monolayers of PA6 cells were treated with recombinant FGF2 or BMP4 for 3 hours and the gene expression analysis of treated PA6 cultures versus the untreated ones was performed. Metabolomics revealed the influence of breast cancer on lymphatic endothelial cell metabolism, metabolic crosstalk, and lymphangiogenic signaling in co-culture

Project description:<p>Breast cancer metastasis occurs via blood and lymphatic vessels. Breast cancer cells 'educate' lymphatic endothelial cells (LECs) to support tumor vascularization and growth. However, despite known metabolic alterations in breast cancer, it remains unclear how lymphatic endothelial cell metabolism is altered in the tumor microenvironment and its effect in lymphangiogenic signaling in LECs. We analyzed metabolites inside LECs in co-culture with MCF-7, MDA-MB-231, and SK-BR-3 breast cancer cell lines using 1H nuclear magnetic resonance (NMR) metabolomics, Seahorse, and the spatial distribution of metabolic co-enzymes using optical redox ratio imaging to describe breast cancer-LEC metabolic crosstalk. LECs co-cultured with breast cancer cells exhibited cell-line dependent altered metabolic profiles, including significant changes in lactate concentration in breast cancer co-culture. Cell metabolic phenotype analysis using Seahorse showed LECs in co-culture exhibited reduced mitochondrial respiration, increased reliance on glycolysis and reduced metabolic flexibility. Optical redox ratio measurements revealed reduced NAD(P)H levels in LECs potentially due to increased NAD(P)H utilization to maintain redox homeostasis. 13C-labeled glucose experiments did not reveal lactate shuttling into LECs from breast cancer cells, yet showed other 13C signals in LECs suggesting internalized metabolites and metabolic exchange between the two cell types. We also determined that breast cancer co-culture stimulated lymphangiogenic signaling in LECs, yet activation was not stimulated by lactate alone. Increased lymphangiogenic signaling suggests paracrine signaling between LECs and breast cancer cells which could have a pro-metastatic role.</p>
