ABSTRACT: To compare The pattern of Gene expression by 7-hydroxymethotrexate-resistant human leukemia cells reveals changes designed to preserve intracellular levels of folate and nucleotide biosynthesis; We used microarrays to detail the global programme of gene expression underlying cellularisation and identified distinct classes of up-regulated genes during this process. Experiment Overall Design: Treatment of human T-cell leukemia lines to the major plasma metabolite of methotrexate (MTX), 7-hydroxymethotrexate (7-OHMTX), resulted in an acquired resistance as a results of complete loss of folypolyglutamate synthetase (FPGS) activity. To identify the genes associated with MTX and 7-OHMTX resistance, we analyzed the patterns of gene expression profiles by leukemia sublines and compared with parent cells.
