Expression analysis of seeds and seedlings of tomato growing on regular MS medium or MS medium supplemented with Helical multi-walled carbon nanotubes (Helical MWCNTs) in concentration 25 ug/ml

ABSTRACT: To investigate effects of Helical multi-walled carbon nanotubes (Helical MWCNTs) on total expression profile of tomato seeds and young sedlings we introduced Helical MWCNTs in MS agar medium in concentrationd of 25 ug/ml. Objectives for this study included the identification of genes that were up or down-regulated at the transcriptional level in tomato seeds and seedlings in response to application of Helical MWCNTs Tomato seeds (1 day after placement of medium) and 11-days old seedlings growing on regular MS agar medium or MS medium supplemented with Helical MWCNTs (25 ug/ml) were selected for RNA extraction and hybridization on Affymetrix microarrays.

Project description:Objectives for this study included the identification of genes that were up or down-regulated at the transcriptional level in tomato InsP 5-ptase transgenic lines exposed to continous light for 7 days (light stress). The RNA was isolated from seedlings of wild-type (WT) and Ins P 5-ptase transgenic line 7 that were germinated on MS medium in a growth chamber with 50 umol m-2 s-1 light intencity at 12 h day-light and 12 h dark for 10 days and then exposed to continous light for 7 days. Affymetrix platform was used for expression analysis.

Project description:In the present work, experiments were carried out to understand the molecular mechanism underlying phyA’ silencing.The EMS-mutagenesis of phyA’ line, L17, generated suppressor mutations that activate phyA’ locus. Detailed study of one suppressor line, sps1, indicated that methylation of specific CG site within the phyA coding region is critical for maintaining phyA’ silencing. In order to study the possible genomic targets of the putative SPS1 factor, microarray analysis using ATH1 genome array chip was done on sps1, L17 and Col-0. 9 samples in total were analyzed. Each line (WT, sps1, L17) were analyzed in triplicate.
