Genome-wide transcriptional profiling of Legionella Pneumophila-infected A549 cells transfected with siRNA against TNFAIP2

ABSTRACT: In this study we transfected A549 cells with siRNA against TNFAIP2, infected them with L. pneumophila and performed transcriptional profiling. We found enrichment of genes in pro-inflammatory pathways by Pathway Over-respresentation analysis upon infection. There was no significant change in gene expression that we could attribute specifically to the knockdown of TNFAIP2. Transcription profiling by array of Legionella pneumophila ppGpp knockout and relA over expression mutants.

Project description:Effect of ppGpp on transcript level after RelA overexpression (90 min). The cells are constructed in our lab. They are knock-out mutants in the production of ppGpp(guanosine tetraphosphate) of Legionella, complemented with an over-expression vector containing the relA gene to express the RelA protein (responsible for the ppGpp production). The aim is to follow the gene response in Legionella pneumophila after induction of ppGpp at different time points.

Transcriptional profiling was done on cultures grown to exponential phase (OD=0.8). Three independent biological replicates were analysed.

Transcription profiling of Dictyostelium discoideum wild type and dupA null after infection with Legionella pneumophila

Project description:The social amoeba Dictyostelium discoideum is vulnerable to infection by the pathogen that causes Legionnaire's Disease, Legionella pneumophila. Dictystelium cells lacking the dual-specificity phospahatase DupA are at least partially resistant to infection, and strikingly the expression profile of uninfected dupA null cells mirrors the profile of infected wild-type cells to a considerable extent. This suggests that DupA has a key role in regulating host defence systems.
