Project description:In this experiment, cattle from 2 breeds Boran and N'Dama were infected with Trypanosoma Congolense, IL1180 clone, African sleeping sickness, a disease which affects cattle in sub-saharan Africa. These breeds were chosen because they differ in their tolerance to infection, with Boran being highly susceptible, and N'Dama being more tolerant to infection. Three tissues Liver, Spleen and Lymph Node were harvested from individuals at various timepoints. Liver, Spleen and Lymph Node were collected post mortem at days 0 (naive), 21 and 35 post infection. In addition, further liver samples were collected by biopsy at days 0 (naive), 12, 15, 18, 26, 29, 32 days post infection. Each condition thus comprises: Time, Tissue, Breed. For each condition, RNA extracts from individuals were hybridised on individual arrays.<br>Infection experiment, tissue harvesting and RNA extraction were carried out at ILRI Nairobi Kenya.<br>Hybridizations were carried out at the Microarray facility, Roslin Institute, UK.<br>Data analysis was carried out at Liverpool University and Manchester University, UK.<br>

Project description:glioblastoma multiforme genomic profiling by single nucleotide polymorphism microarray<br><br>Human GBM (glioblastoma multiforme)cell lines (U87, U118, U138, U343, U373, T98G) were maintained in Dulbecco's modified Eagle's medium with 10 % fetal calf serum, 10 U/ml penicillin-G, and 10 mg/ml streptomycin. All cells were incubated at 37 oC in 5% CO2.<br><br>Four primary GBM explants were established from patients with glioblastoma multiforme undergoing surgery as following described: Tumor specimens were immediately transported to the laboratory, finely minced to single cell suspension and cultured in complete medium [Ham's F-12/DME High Glucose medium containing 10% fetal calf serum, 10 U/ml penicillin-G, and 10 mg/ml streptomycin and 2 mM glutamax-1 into 100 cm2 tissue culture plastic dishes the second passage. All cells were incubated at 37 oC in 5% CO2.<br><br>GBM (glioblastoma multiforme) tissue samples were quick frozen. <br><br>Standard proteinase K-phenol-chloroform extraction method was used to extract DNA from GBM samples, cell lines and explants.<br><br>The matched peripheral blood data can be used as normalized data for their matched tumor tissue data. <br><br>The cell lines samples and two explants without normalized data, but they can be normalized by one of the peripheral blood DNA data.

Project description:Gene expression in early animal embryogenesis is in large part controlled post-transcriptionally. Maternally-contributed microRNAs may therefore play important roles in early development. We have elucidated a major biological role of the nematode mir-35 family of maternally-contributed, essential microRNAs. We show that this microRNA family regulates the sex determination pathway at multiple levels, acting both upstream and downstream of her-1 to prevent aberrantly activated male developmental programs in hermaphrodite embryos. The predicted target genes that act downstream of the mir-35 family in this process, sup-26 and nhl-2, both encode RNA binding proteins, thus delineating a previously unknown post-transcriptional regulatory subnetwork within the well-studied sex determination pathway of C. elegans. Repression of nhl-2 by the mir-35 family is not only required for proper sex determination but also for viability, showing that a single microRNA target site can be essential. Since sex determination in C. elegans requires zygotic gene expression to read the sex chromosome karyotype, early embryos must remain gender-naïve; our findings show that the mir-35 family microRNAs act in the early embryo to function as a developmental timer that preserves naïveté and prevents premature deleterious developmental decisions. The mir-35 family of microRNAs is essential for development. The mir-35-41(nDf50) allele deleted 7 of 8 mir-35 family members, and presents a hypomorphic phenotype in which embryonic lethality is temperature sensitive. To characterize the molecular phenotype associated with loss of mir-35 family function, we profiled gene expression in mir-35-41(nDf50) mutant embryos at both permissive (20) and restrictive (25) temperatures. Project description:Gene expression was studied from the blood derived RNAs of the Finnish family members as well as from 10 controls using GeneChip Human Genome U133 Plus2 (Affymetrix). Eight out of 10 family members in the expression analysis are heterozygous for the NPAT c.2437-2438delAG, three of which are NLPHL cases. NPAT expression was studied in the NLPHL patients and the carriers of the deletion c.2437-2438delAG (8) compared to the controls and the non-mutation carriers (12). Note: Case 2 not submitted. Project description:The long-lived sacred lotus (Nelumbo nucifera) fruit provides a good platform for uncovering aging-related mechanism in extension of seed-life and improving of food quality of economic crops during storage. Here, we investigated the proteome of artificial aging lotus seed to uncover the aging -resistance mechanism. Proteome was performed on the seeds embryo axis treated at 80C for 0, 12 and 24 Hours. We identified 4667 proteins, and found 383 and 37 proteins (folds>1.3, FDR<0.01) were differentially expressed from 0-12h and 12-24h treatment, respectively. This proteome data will provide a better understanding and give further insights on lotus seed aging-resistance.
