Transcription profiling by array of mouse Col8a1-Col8a2 deficient and wild type mesangial cells stimulated with TGF-beta1

ABSTRACT: Col8a1/Col8a2-deficient mesangial cells and wildtype cells are treated with TGFbeta-1 for 24h. This experiment was replicated once.

Project description:Extrarenal viral infections commonly trigger glomerulonephritis mostly in association with immune complex disease. The immunoglobulin component of immune complexes can activate glomerular cell Fc receptors but whether complexed viral nucleic acids contribute to glomerular inflammation remains unknown. Glomerular mesangial cells express TLR3 but lack TLR7-9, hence, it is unclear whether mesangial cells can recognize and respond to viral ssRNA or DNA. Here we studied the immune responses activated by 3P-RNA (5'Triphosphate RNA) and Non-CpG DNA (Double stranded DNA) in primary mesangial cells (PMC). We used microarrays to detail the global programme of gene expression that induced by 3P-RNA and Non-CpG DNA. Experiment Overall Design: PMC were stimulated with 3P-RNA and Non-CpG DNA for 6 hours and then total RNA was isolated for hybridization to MOE430_2 arrays.

Gene expression analysis of high-glucose stimulated human mesangial cells and tubular epithelial cells

Project description:we performed genome-wide transcriptome profiling in mesangial cells and tubular epithelial cells (TECs), which were stimulated by high glucose (HG) and detected the expression of inflammation associated genes. HG increased the mRNA expression of oxidative stress, inflammasome and mammalian target of rapamycin (mTOR) related genes in mesangial cells. Pro-inflammatory/Th1 gene expression was upregulated, but Th2 related gene expression was downregulated in mesangial cells. In TECs, HG stimulation increased pro-inflammatory/Th1/Th2 gene expression. mRNA profiles of human mesangial cells and tubular epithelial cells, which were stimuated HG for 24 hours or mannitol contorol.

Heme oxygenase-1 knocked down rat mesangial cells treated with hydrogen peroxide

Project description:HO-1 cells denote the cultured rat mesangial cells with heme oxygenase-1 knocked down by RNA interference (using lentiviral vector). GFP cells denote the cultured rat mesangial cells that are transfected with empty lentiviral vector containing GFP cassette. Cells are treated with hydrogen peroxide 100 micromolar for 2 hours, or without. RNA are then harvested for array analysis. Biological replicates are performed (two independent experiment sets). GFP cell and HO-1 cell are untreated, or treated with hydrogen peroxide (100 micromolar for 2 hours).

Project description:Previous data have demonstrated the attenuation of platelet-derived growth factor (PDGF)-induced hyperproliferation and migration in primary human mesangial cells by lipoxin A4. In these experiments we aimed to find out what the effect of Lipoxin A4 would be on the global genomic changes associated with PDGF. We treated cells with PDGF (10ng/ml) ± LXA4 (1nM) for a 24h period and examined genomic differences, we could derive from our results that lipoxin A4 was diminishing the pro-inflammatory, pro-proliferative and pro-fibrotic responses induced by PDGF.

Transcription profiling of human pulp and odontoblasts reponse to TGF-beta1

Project description:Transforming growth factor beta 1 (TGF-Î²1) is the most extensively studied growth factor in dentin-pulp complex, with pleiotropic effects on pulp response and healing. Our main objective was to analyze the expression profile of pulp tissue and odontoblasts, and the effects of TGF-Î²1 on these profiles in cultured human pulp and odontoblasts with a specific interest in the anti- and pro-inflammatory cytokines. Experiment Overall Design: Pulp tissues and odontoblasts were cultured for different time periods, and microarray was performed to both cultured and native samples to detect the effects of TGF-Î²1. Expression of various interleukins (IL) were confirmed by RT-PCR, and in +/- TGF-Î²1 treated pulps also by antibody array.

Transcription profiling of human tongue carcinoma cells treated with emdogain and TGF beta1

Project description:Background: The composition of Emdogain® (EMD) is unclear, but it has been postulated to contain transforming growth factor-beta-1 (TGF-beta1) as the main functioning cytokine. The purpose of this study was to compare target genes of EMD and TGF-beta1 on invasive oral carcinoma HSC-3 cells. Particular focus was on matrix metalloproteinase (MMP) family genes. Methodology/Principal Findings: Affymetrix microarrays were conducted to study differentially (P<0.05) expressed genes in HSC-3 cells after 6h and 24h of EMD (200 ?g/ml) or TGF-beta1 (10 ng/ml) incubations. Gene Ontology (GO) enrichment analyses from the regulated genes were also conducted. After 6h of EMD or TGF-?1 treatment, 48 and 393 genes, respectively, were regulated. After 24h incubations, only 12 genes by EMD but 346 genes by TGF-beta1 were regulated. Among the most regulated genes by both of the study reagents were several genes commonly regulated in carcinomas, including MMP-9 and -10. The expression of MMP-10 by EMD treated carcinoma cells was also verified in protein level. Conclusions/Significance: Our results show that EMD and TGF-beta1 can regulate genes related to carcinogenesis, but TGF-beta1 regulates significantly greater number of genes. This suggests that TGF-beta1 cannot be present in EMD at the studied concentration, if present at all.

Single glomerular mesangial cells RNA-seq analysis

Project description:Available single-cell RNA-seq analyses have revealed that individual cells of the same type differ substantially in gene expression. We wonder whether glomerular mesangial cells, the cell type that support the unique structure of glomeruli, also exhibit big difference in gene expression among individual cells; and what biological information could be obtained from the single-mesangial cell RNA-seq data. Therefore, we isolated mouse glomeruli by Dynabead/magnetic concentration method, and digested them with enzymes to dissociate the cells. We loaded the single cell suspension to a Fluidigm C1 Single-Cell Auto Prep System for single cell cDNA preparation. The cDNA samples were analyzed with mesangial cell marker, Gata3,and the Gata3 positive samples were amplified and underwent sequecing using Illumina Highseq 2000 system.
