Single cell sequencing of wild-type and mutant iPSC-derived hypothalamic arcuate-like neurons cells (deletion of hg19:chr16:53,796,691-53,832,771 / hsFTOΔ36)

ABSTRACT: In this experiment, we deleted 36,100 bp of the first intron of the human FTO gene to test whether this region contains transcriptional enhancers for RPGRIP1L, FTO, IRX3, IRX5, and IRX6 genes. The deletion was performed in iPSC and we differentiated wild-type and hsFTOΔ36 cells into hypothalamic progenitor cells. Because differentiation often results in various cell populations, we performed scRNA-seq of single cells to better identify the differential expression genes between deleted and wild-type cells.
