TLR signalling induced by bacterial RNA vs R848


ABSTRACT: Murine dendritic cells were derived from bone-marrow of 4 mice using GM-CSF. The DC were treated with RNA from gram-positive bacteria Listeria monocytogenes packaged in DOTAP or TLR7 agonist R848. Negative controls were DOTAP with no RNA or mock.

Bacterial RNA (bRNA) can induce cytokine production in macrophages and dendritic cells (DCs) through a previously unidentified receptor. Gene expression analysis of murine DCs showed that bRNA induced gene regulation similar to that induced by stimulation of TLR7 with R848. Although TLR7 was dispensable for cytokine induction by bRNA, TLR-associated proteins MyD88 and UNC93B were required.
