DNA-seq of Candida glabrata BG2 derived amphotericin B adaptors

ABSTRACT: In this study, BG2 is the wild type Candida glabrata strain. Approximately 1 million cells of BG2 were spread on YPD plate supplemented with 4 ug/ml and 5 ug/ml amphotericin B, respectively. Randomly 18 and 6 adaptors were chosen from 4ug/ml (TJ2873-TJ2890) and 5ug/ml (TJ2891-TJ2896) drug plate, respectively.These adaptors were sequenced.

Whole genome sequencing of BG2 derived adaptors selected from sub-inhibitory amount of tunicamycin

Project description:Candida albicans lab strain BG2 was grown in YPD broth supplemented with 0.5ug/ml tunicamycin at 37C for 24h. We used three techinical repeats. The culture was washed, and diluted with distilled water. Then approximately 200 cells from each of the three cultures were spread on YPD plates. Randomly 99 colonies from each culture were tetsted for tolerance to tunicamycin, as compared to the parent BG2. In total we got 6 tolerant adaptors. These 6 adaptors as well as the parent were sequenced.

Whole genome sequencing of caspofungin adaptors in Candida auris

Project description:Candida auris clade III isolate B12039 was spread on YPD plate supplemented with 250 ng/ml, 500 ng/ml, 1000 ng/ml, and 2000 ng/ml of caspofungin. In total, 33 adaptors were chosen for further analysis. We did sequencing of them as as well as the parent. 9 adaptors (250-1 - 250-9) were obtained from YPD+250ng/ml caspofugin. 6 adaptors (500-1 - 500-6) were obtained from YPD+250ng/ml caspofugin. 6 adaptors (1000-1 - 1000-6) were obtained from YPD+250ng/ml caspofugin. 12 adaptors (2000-1 - 2000-12) were obtained from YPD+250ng/ml caspofugin

H99 derived uniconzole (0.25ug/mL and 2ug/mL) adaptors

Project description:H99 derived 2 ug/mL uniconazole adaptors: Approximately 1 million cells of Crypotoccus neoformans lab strain H99 were spread on YPD-agar plate supplemented with 2 ug/mL uniconazole. After 5 days incubation of the plate at 30℃, randomly 30 adaptors were chosen. 21 adaptors were resistant and were sequenced. H99 derived 0.25 ug/mL uniconazole adaptors: Cells of H99 were adjusted to 2500 cells/mL. The culture was supplemented with 0.25 ug/mL uniconazole. After 72 h incubation, the culture was washed, diluted and approximately 200 cells were spread on YPD-agar plate. Randomly 95 colonies were tested for resistance to uniconazole. One colony (TJ3832) was resistant and was sequenced.

Randomly 96 adaptors derived from TJ926 and 96 adaptors derived from TJ927 were tested with disk diffusion assay. Among the resistant adaptors, randomly 15 adaptors (A4-A18) and 48 adaptors (A19-A66) derived from TJ926 were sequenced.
