Whole genome sequencing of TF-AID-mutagenized flies

ABSTRACT: In this study, we stably expressed TF-AID in the germline of Drosophila melanogaster to induce mutations around specific transcription factor binding sites across the genome. To identify the mutations, we sequenced: (1) individual flies from the TF-AID-expressing populations, sampled across generations and populations; (2) all the background stocks, including TF-AID stocks, NGT40, all balancers, and w1118; (3) 3rd-chromosome isogenic lines derived from TF-AID populations, with 2 flies per isogenic lines sequenced, as replicates; (4) flies from lines with eye phenotypes (pooling 6-12 flies per line) and their 3rd-chromosome isogenic descendants.
