Androgen Deprivation Alters the Transcriptome of Endothelial Cells More Dramatically in Human Prostate Cancer Tissue than in Benign Prostate Tissue

Project description:Purpose: Examine the cell type specific (Epithelial, Endothelial, and Stromal) transcriptome profile between prostate primary tissue xenografts of prostate tissue tumor tissue and uninvolved tissue (named non-tumor hereafter) in response testosterone (T) deprivation. Additional cell type specific untreated fresh tissue from both tumor and non-tumor samples is included for additional validation. Related samples previously uploaded to GEO are found under accession number GSE145641. Method: Nude-Fox1nu mice were castrated and silastic tubing containing dry T was inserted providing “humanized” levels of circulating T. 5-8 pieces of fresh human prostate tissue was then inserted subcutaneously along the flank of the animal. For samples not subjected to T-deprivation, after 30 days the animal was euthanized, and tissue was harvested. For samples undergoing T-deprivation, after 30 days with humanized circulating T levels, T-deprivation was performed by surgical removal of silastic tubing after which serum T levels dropped below the level of detection within 24 hours. Samples were then harvested at Pre T-deprivation time point, an early T-deprivation time point (3-days) or late T-deprivation timepoint (15-17 days) post-surgery. Tissue from fresh human prostate or tissue from xenograft and was then enriched into endothelial, epithelial and the remaining stromal compartments using magnetic bead enrichment. RNA-Seq was then preformed on cell types from both origins and the results compared to determine the cell type specific effects of T-deprivation on the human prostate transcriptome.

Suppression of LPS-induced IFNg and NO in splenocytes by select E2-regulated miRNA: A novel mechanism of immune mod.

Project description:microRNA (miRNA), recently identified, non-coding, small RNA, are emerging as key regulators in homeostasis of the immune system. Therefore, aberrant expression of miRNA may be linked to immune dysfunction, such as in chronic inflammation and autoimmunity. In this study, we investigated the potential role of miRNA in estrogen-mediated regulation of innate immune responses, as indicated by upregulation of LPS-induced IFNg, inducible nitric oxide synthase (iNOS), and nitric oxide in splenic lymphocytes from estrogen-treated mice. We found that miR-146a, a negative regulator of Toll-like receptor (TLR) signaling, was decreased in freshly-isolated splenic lymphocytes from estrogen-treated mice compared to placebo controls. Increasing the activity of miR-146a significantly inhibited LPS-induced IFNg and iNOS expression in mouse splenic lymphocytes. Further, miRNA microarray and Real-time RT-PCR analysis revealed that estrogen selectively upregulates/downregulates the expression of miRNA in mouse splenic lymphocytes. miR-223, which is highly upregulated by estrogen, regulates LPS-induced IFNg, but not iNOS or nitric oxide in splenic lymphocytes. Inhibition of miR-223 activity decreased LPS-induced IFNg in splenic lymphocytes from estrogen-treated mice. Our data are the first demonstrating selective regulation of miRNA expression in immune cells by estrogen and are indicative of an important role of miRNA in estrogen-mediated immune regulation. Transcription profiling of rat anterior pituitary gland of the ACI treated with diethylstilbestrol (DES) from Copenhagen and Brown Norway strains.

Project description:Anterior pituitary glands were isolated from 21 week old male rats either untreated or treated for 12 weeks with the synthetic estrogen diethylstilbestrol (DES). Three biological replicates were prepared for untreated, control animals and four biological replicates were prepared for DES-treated animals. This was done for each of 3 inbred rat strains: ACI, Copenhagen, and Brown Norway. Expression profiles were determined using Affymetrix Rat Genome 230 v. 2.0 arrays. Comparisons of untreated vs. treated animals within a strain will allow identification of estrogen responsive genes. Comparisons between strains, either treated or untreated, will identify strain (i.e., genetic) differences in expression. Experiment Overall Design: 21 samples from anterior pituitary gland were analyzed. For each of 3 inbred rat strains (ACI, Copenhagen, and Brown Norway) there were 3 biological replicates of untreated, control animals, and 4 biological replicates of DES-treated animals.

Estrogen Induced DNA Methylation Changes in Mouse Hippocampus

Project description:Estrogen, specifically estradiol, has been shown to mediate DNA methylation changes at mutiple locations downstream of estrogen response elements (EREs). We identified differentially methylated regions associated with long-term estradiol exposure in the hippocampus of C57BL/6J mice. Hippocampus was collected from ovariectomized (OVX) C57BL/6J mice treated with estradiol (n=5) and vehicle controls (n=5). Genomic DNA was extracted and treated with methylation sensitive restriction enzymes HpaII and HinP1I to enrich for the unmethylated fraction. This fraction was interrogated using the Affymetrix GeneChip® Mouse Tiling Promoter 1.0R Array

The organizational effects of testosterone on brain DNA methylome

Project description:Analysis of DNA methylation in the bed nucleus of the stria terminalis/preoptic area and striatum in response to perinatal testosterone exposure. The hypothesis tested was that treatment of females with testosterone on the day of birth would lead to masculinization of the methylome in adulthood. There were three experimental groups: males, females, and females treated with T on the day of birth. The methylation patterns in each group was determined using reduced representation bisulfite sequencing. Two brain regions and two time points (day 4 and day 60) were surveyed in each group. Each biological replicate is a pool of tissue from three animals.
