Differential expression in the cortex and white matter of patients with cerebral small vessel disease (SVD)

ABSTRACT: In this study, we performed the first genome-wide expression profiling of post-mortem brains of a cohort of patients deceased from SVD and compared them to age-matched normal controls. Normal-appearing frontal temporal and occipital cortical and subcortical brain samples were dissected at autopsy from 5 patients diagnosed with pure SVD and 5 control patients without neurological disease and immediately frozen.

Pro-inflammatory monocyte phenotype during acute progression of cerebral small vessel disease

Project description:The aetiology of cerebral small vessel disease (SVD) remains elusive, though currently evidence is accumulating that inflammation contributes to its pathophysiology. In this prospective high-frequency imaging study, we showed that the incidence of acute SVD progression coincides with a functional and transcriptional pro-inflammatory monocyte phenotype. To detect SVD progression, individuals with SVD underwent monthly magnetic resonance imaging (MRI) for 10 consecutive months. RNA sequencing revealed a pro-inflammatory monocyte signature with upregulation of the EGR2 gene. Collectively, we showed that individuals with progressive SVD have pro-inflammatory circulating monocytes characterized by an augmented cytokine production capacity and a pro-inflammatory transcriptional signature.

Transcriptional profiling of human microglia reveals grey-white matter heterogeneity and multiple sclerosis-associated changes

Project description:Microglia are brain-resident, myelin-phagocytosing cells, yet their role in lesion initiation in grey and white matter regions in multiple sclerosis (MS) is unclear. We isolated primary microglia from both, occipital cortex and corpus callosum, of 10 MS and 11 control donors and studied their transcriptional profile by RNA sequencing, thereby identifying regional and MS-associated changes. Identification of pathways underlying regional differences showed a relatively increased type I interferon response in cortical grey matter microglia, while white matter microglia more highly expressed NF-κB pathway genes. In normal-appearing white matter MS tissue, lipid metabolism genes were increased, suggesting processing of myelin by microglia already in areas seemingly devoid of MS pathology. Normal-appearing grey matter MS microglia showed increased activation of glycolysis and metal ion homeostasis, possibly reflecting microglia reacting to iron depositions. Notably, expression of genes associated with microglia homeostasis were hardly changed, suggesting that subtle regional changes in MS-associated microglia do not yet affect their resting state.

Gene expression of white and grey matter demyelinated areas and normal appearing matter from Multiple Sclerosis leukocortial lesions of human post-mortem tissue

Project description:Using laser capture microscopy, white (WM) and grey matter (GM) demyelinated areas and normal appearing matter was collected from histologically verified leukocortical lesions from snap-frozen human post mortem tissuederived from Multiple Sclerosis patients. Our data shows large differences in gene expression in WM and GM demyelinated areas (compared to their respective normal appearing matter) even when the demyelinated areas are spatially connected such as in leukocortical lesions. Thus, we show that WM demyelinated areas and GM demyelinated areas are distinct entities with distinct pathology. Therefore findings observed in WM demyelinated areas cannot be generalized to GM demyelinated areas.

To elucidate the cytotoxic impact and brain-specific host response during coronavirus disease 2019 (COVID-19), we profiled the olfactory mucosa, olfactory bulb, brainstem and cerebellum from deceased COVID-19 patients who underwent rapid autopsy.

Expression data from human brain samples

Project description:Spontaneous intracerebral hemorrhage (ICH) represents about 15% of all strokes and is associated with high mortality rates. Our aim was to identify the gene expression changes and biological pathways altered in the brain following ICH. Twelve brain samples were obtained from four deceased patients who suffered an ICH including perihematomal tissue (PH) and the corresponding contralateral white (CW) and grey (CG) matter. Brain samples from 4 deceased patients who had a supratentorial intracerebral hemorrhage within the previous 4 days were included in the microarray study. This study group included 2 women and 2 men with a median age of 79 (68-92). On autopsy and during macroscopic exam, perihematomal areas suspected to present edema were identified by an experienced neuropathologist using last available neuroradiology images. Samples from perihematomal areas (PH), contralateral grey matter (CG) and contralateral white matter (CW) were obtained within the first 5 h after death and snap frozen in liquid nitrogen and stored at -80ºC until RNA isolation.

Proteome analysis of microvessels from CADASIL patients

Project description:CADASIL, the most frequent and intensely studied monogenic SVD, is characterized by a severe pathology in the cerebral vasculature including the mutation-induced aggregation of the Notch3 extracellular domain (Notch3ECD) and the formation of protein deposits of insufficiently determined composition in the extracellular space of vessel walls. To advance our understanding of protein accumulation in CADASIL-affected tissue, we quantitatively determined the proteome of cerebral vessels isolated from patient and control autopsy samples (n = 6 for each group), obtaining 95 proteins with significantly increased abundance.

Brain-wide alterations revealed by spatial transcriptomics and proteomics in COVID-19 infection

Project description:Understanding the pathological basis of the neurological symptoms observed following SARS-CoV2 infection is essential to optimizing outcomes and developing therapeutics. We performed proteomics profiling across eight cortical and subcortical brain regions, frontal lobe, temporal lobe, occipital lobe, hippocampus, thalamus, basal ganglia, midbrain, and pons, using postmortem brain samples from severe acute COVID-19 patients and matched controls (n=16), all preserved as formalin-fixed paraffin-embedded (FFPE) tissue. Integrating proteomics and additional transcriptomic analyses, we identified board dysregulation of mitochondrial and synaptic pathways in deep-layer excitatory neurons and changes exhibited similarities with those seen in various age-related neurodegenerative diseases, such as Parkinson's disease (PD) and Alzheimer's disease (AD).
