Single-cell and single-nucleus RNAseq of the human and mouse skeletal muscle across age

Project description:This study aims to characterize the diversity of cell types in human skeletal muscle across age using two complementary technologies: single-cell and single-nucleus sequencing, which provide a comprehensive coverage of cell types in the muscle. We leveraged the aforementioned datasets to study change in cell type composition and gene expression between young (n= 8, approx. 20-40 yrs) and old (n = 9, approx. 60-80 yrs) adults, highlighting changes in the major skeletal muscle compartments, muscle satellite cells, myofiber and muscle microenvironment including stromal, immune and vascular cell types. Additionally, we generated a complementary mouse muscle aging dataset by profiling hindlimb muscles from young (n = 5, 3 months) versus old mice (n = 3, 19 months), using single-cell and single-nucleus sequencing for comparison.

Near-whole-genome transcriptome analysis of gene expression in human skeletal muscle tissue at baseline in obese individuals with Type 2 Diabetes

Project description:Analysis of gene expression associated with exercise response. The hypothesis tested was that individuals with Type 2 Diabetes that failed to demonstrate exercise-induced metabolic improvements would also reflect this lack of response in their skeletal muscle transcriptional profile at baseline. Of 186 genes identified by microarray analysis, 70% were upregulated in Responders and downregulated in Non-responders. Several genes involved in substrate metabolism and mitochondrial biogenesis differed significantly between the groups at baseline. This differential baseline gene expression indicated that Non-responders had blunted oxidative capacity. Total RNA extracted from baseline samples of skeletal muscle of obese individuals with Type 2 Diabetes who were characterized as either Responders or Non-responders was examined for differential expression of exercise response-assocated genes.

HIV Infection and Antiretroviral Therapy Have Divergent Effects on Mitochondria in Adipose Tissue

Project description:Exploratory microarray analysis identified significant changes in gene expression in adipose tissue. These included changes in genes regulating lipid and steroid metabolic processes and electron carrier activity in HIV-infected patients receiving antiretroviral therapy (ART). Additional genes involved in metabolic processes and mitochondrial function were found to be up-regulated in the adipose tissue of HIV-positive patients compared with HIV-negative controls. To identify differential gene expression in different tissues (PBMC, muscle, adipose tissue) between HIV patient groups (HIV negative, HIV positive with ART, HIV positive without ART).

Characterization of the skeletal muscle proteome in under-nourished old rats

Project description:Therefore, undernutrition is common in the elderly where its prevalence varies from 4 to 10% at home, from 15 to 30% in care homes and reaches 30 to 70% in hospitals de-pending on the diagnostic criteria used [10]. Most of all, aging impairs the ability to adapt to undernutrition leading to severe loss of body weight and muscle [11-14]. In particular, old rats exhibit defective adaptation to long-term 50% dietary restriction initiated at an advanced age, resulting in dramatic body weight loss and deregulation of protein metabolism in terms of nitrogen balance and protein content, especially at skeletal muscle level [12]. Therefore, in elderly patients, undernutrition exacerbates the state of frailty and dependence, greatly increasing the risk of morbidity and mortality [15]. This is even more critical that the world aging population is growing up. Accord-ing to United Nations statistics on the demographic trend in aging population, the proportion of persons aged 65 or over in the world is projected to reach nearly 16% in 2050, compared to 9% in 2019 [16]. Specific studies are needed to better understand the undernutrition-induced skeletal muscle alterations in order to identify new therapeu-tic targets and prevent sarcopenia in the old population

Effects of acute exercise on gene expression in exercising and non-exercising human skeletal muscle

Project description:Background: Exercising is know to have an effect on exercising skeletal muscle, but unkown is the effect on non-exercising skeletal muscle. Gene expression changes in the non-exercising skeletal muscle would point to a signalling role of skeletal muscle 9 healthy middle-aged men performed 1 hour of one-legged exercise, before and afterwards muscle biopsies were taken from both legs. Skeletal muscle biopsies were analyzed by microarray.

Effects of 30 days resveratrol supplementation on adipose tissue morphology and gene expression patterns in obese men

Project description:Polyphenolic compounds, such as resveratrol, have recently received widespread interest due to their ability to mimic effects of calorie restriction. The objective of the present study was to gain more insight into the effects of 30 days resveratrol supplementation on adipose tissue morphology and underlying processes. Nine healthy obese men were supplemented with placebo and 150mg/day resveratrol for 30 days, separated by a 4-week washout period. A postprandial abdominal subcutaneous adipose tissue biopsy was collected to assess adipose tissue morphology and gene expression using microarray analysis. Resveratrol significantly decreased adipocyte size, with a shift towards a reduction in the proportion of large and very large adipocytes and an increase in small adipocytes. Microarray analysis revealed downregulation of Wnt, Notch and BMP/TGF-β signaling pathways and upregulation of pathways involved in cell cycle after resveratrol supplementation, suggesting enhanced adipogenesis. Furthermore, the lysosomal/phagosomal pathway and the transcription factor EB were upregulated, reflecting an alternative pathway of lipid breakdown by autophagy. These data suggest that adipose tissue is an important target tissue for the effects of resveratrol in humans, but further research is necessary to investigate whether it translates into an improved adipose tissue function. In a double-blind randomized crossover study, 9 healthy obese men were supplemented with placebo and 150 mg/day resveratrol for 30 days, with a 4-week washout period in between. At the end of each intervention period, a biopsy was taken from the abdominal subcutaneous adipose tissue, which was subjected to gene expression profiling.
