mRNA expression pattern of KrasG12D mice with acute pancreatitis compared with wild-type mice without acute pancreatitis

Project description:Transgenic KrasG12D mice can recapitulate pancreas intra-epithelial neoplasia (PanIN). Caerulein is a cholecystokinin analogue and induces acute pancreatitis when injected intra-abdominally. Caerulein-induced acute pancreatitis will accelerate PanIN progression in KrasG12D mice. We compared mRNA profile changes between KrasG12D mice with acute caerulein-induced pancreatitis and wild-type mice without acute pancreatitis. The experiment had two groups. Experiment group: KrasG12D mice with acute caerulein-induced pancreatitis (N=6). Three mice in experiment group received 1-week caerulein injection, and the other three mice received 2-week caerulein injection. All experiment group mice started to receive caerulein injection at 1-month of age, and were sacrificed at the last day of caerulein injection. Control group: wild-type mice without acute pancreatitis (N=6). The mice were sacrificed at 1.5-month of age. Whole pancreas tissue lysate samples were subjected to mRNA array assay.

Project description:Autoimmune pancreatitis (AIP) is a disease with unclear immunologic triggers. This study shows that the pancreatic stellate cells(PSCs) are involved in the regulation of the immune response and can cause autoimmunity when the NF-κB signalling in these cells is disrupted. The PSCs were isolated from animals which show autoimmune pancreatitis (NEMO knockout group) or chronic pancreatitis (NEMO wildtype group).

Global quantitative proteomics analysis reveals the downstream signaling networks of Msx1 and Msx2 in skeletal muscle development

Project description:The homeoprotein Msx1 and Msx2 involved in normal skeletal muscle development and also contribute to muscle defects if altered during development. Deciphering the downstream signaling networks of Msx1 and Msx2 in myoblasts differentiation will help us to understand the molecular events that contribute to muscle defects. The objective of this study was to evaluate the proteomics characteristics in Msx1 and Msx2 mediated myoblasts differentiation, using isobaric tags for the relative and absolute quantification labelling technique (iTRAQ). The results showed that 1535 proteins with quantitative information were obtained. Volcano plots illustrated, in undifferentiated stage, 32 common downstream regulatory proteins for Msx1 and Msx2, 39 specific regulatory proteins for Msx1, and 13 specific for Msx2. While, in differentiated stage, 17 common downstream regulatory proteins for Msx1 and Msx2, 10 specific regulatory proteins for Msx1, and 21 specific for Msx2. Gene ontology, KEGG pathway and protein-protein interaction networks analyses revealed these proteins primarily associated with Arginine and proline metabolism, Glycolysis/Gluconeogenesis, Fatty acid degradation, Metabolism of xenobiotics by cytochrome P450 and Apoptosis. In addition, our data shows Acta1 was probably a core of the downstream regulatory networks of Msx1 and Msx2 in skeletal muscle development. The findings will help us to understand the molecular roles of Msx1 and Msx2 during muscle development as well as regeneration, and to understand the molecular events that contribute to muscle defects.

Expression data of caerulein-treated wild-type mice and KrasG12D-mutated mice at different stages of pancreatic regeneration after inflammatory injury

Project description:Consecutive caerulein injections induce an acute pancreatitis in mice. Here, we recorded gene expression levels at different stages of pancreatic regeneration in wild-type mice as well as KrasG12D-mutated mice. Tissue was collected from mice pancreata, cell sorting was not performed. t=0h refers to the time where caerulein was injected. control referes to NaCl-treated samples (no caerulein). 13 time points were used for wild-type mice, 9 time points were used for KrasG12D-mutated mice. Multiple replicates were generated for each time point. We used Affymetrix GeneChip Mouse Gene 1.0 ST arrays.

Project description:We have studied pancreas samples in pancreas from a mouse model of acute pancreatitis, comparing wild-type animals and TRP14 KO animals. Samples are extracted with N-ethylmaleimide to block reduced cysteines, and no reducing agent is used, with a search including cysteinylated peptides. Acute pancreatitis was induced in 12 weeks-old mice (wild-type and KO for TRP14) by seven intraperitoneal injections of cerulein (50 µg/kg body weight) at 1 h intervals. Then, 1 h after the last injection, animals were euthanized under anesthesia with 3% isoflurane, exsanguinated, and the pancreas was immediately removed and processed.

microRNA expression in Mestenteric lymph samples from rat models of acute pancreatitis

We used Affymetrix microRNA arrays to assess the differences in mesenteric lymph fluid miRNAs in a taurocholate induced rat model of pancreatitis Mesenteric lymph was collected from five biological replicates from control sham operated animals (SHAM), fluid resuscitated taurocholate induced acute pancreatitis (RAP) and non-resuscitated taurocholate induced pancreatitis animals (AP). The latter group represent a more severe form of the disease.

Whole-Genome Wide Quantitative Proteomics Studies Revealed Tissue-Specific Expression and Phosphorylation of Regulatory Proteins in Arabidopsis

Project description:Organogenesis in plants occurs during all stages of the life cycle. Although previous studies have identified many genes important for either vegetative or reproductive development at RNA level, global scale information of translational and post-translational regulatory levels is still limited. Here, six Arabidopsis stages/organs were analyzed using quantitative proteomics and phosphoproteomics, identifying 2,187 non-redundant proteins and 1,194 phosphoproteins. Compared to that in cauline leaves, expression of 1,445, 1,644, and 1,377 proteins showed greater than 1.5-fold alteration in stage 1-9 flowers, stage 10-12 flowers, and open flowers, respectively. In addition, 294 phosphoproteins with 472 phosphorylation sites were newly discovered, including 275 showing differential expression patterns, providing possible candidates for functional studies or molecular markers. In addition, more phosphorylated proteins were identified in reproductive stages (1,149) than in the vegetative process (995). 701 of the proteins differentially phosphorylated between flowers and leaves were predicted to localize to the nucleus, including 22 transcription factors. Moreover, phosphorylation of CDPK6 and MAPK6 and their interacting proteins, were enriched in reproductive tissues. Overall, our study yielded extensive data on protein expression and phosphorylation at six stages and provides an important resource for understanding regulatory mechanisms during plant development.
