Transcriptomics analysis of E. faecalis V583 exposed to subinhibitory concentration of chlorhexidine

ABSTRACT: The vancomycin resistant strain V583 was exposed to subinhibitory concentration of chlorhexidine. E. faecalis V583 cells were collected at 15 minutes after chlorhexidine addition and without chlorhexidine, for RNA extraction and hybridization on Affymetrix microarrays.

Transcriptomic analysis of chlorhexidine shock in Acinetobacter baumannii

Project description:In this study the transcriptomic changes occurring in A. baumannii ATCC 17978 in response to shock treament using the biocide chlorhexdine were determined. A. baumannii ATCC 17978 cells were grown at 37Â°C with shaking (200rpm) in 35mL cultures in MH broth to OD600=0.75, at which time they were split into 15mL cultures. One 15mL sample was treated with 4Âµg/mL chlorhexidine (0.5x MIC), whereas the other was not treated and used as a reference. Cultures were allowed to grow for a further 30 mins (to an average final OD600= ~1.35 for untreated cells and 1.25 for chlorhexidine treated samples), when cells were harvested by centrifugation and immediately suspended in Trizol reagent (Invitrogen). Total RNA was extracted using the PureLinkTM Micro-to-Midi Total RNA Purification kit (Invitrogen), incorporating an on-column DNAseI (Invitrogen) digestion. The experiment included using four grids of a custom Agilent 8 x 15000 spot microarray. Three biological replicates and one flip dye experiment.

Transcriptomic comparison of chlorhexidine tolerant and sensitive Salmonella Typhimurium

Project description:Transcriptional profiles of a chlorhexidine tolerant Salmonella Typhimurium were compared to its, chlorhexidine sensitive, isogenic progenitor isolate. RNA was extracted from mid-log phase cells from both isolates, without chlorhexidine exposure and following exposure to 1 µg/ ml of chlorhexidine for 30 minutes. Transcriptional profiles of the tolerant isolate were compared to the sensitive isolate, with and without chlorhexidine exposure.

Project description:In the present study, we employed Affymetrix Pseudomonas aeruginosa GeneChip arrays to investigate the dynamics of global gene expression profiles during the cellular response of Pseudomonas aeruginosa to Chlorhexidine diacetate, which involved initial growth inhibition and metabolism. Experiment Overall Design: We conducted three independent microarray experiments (biological replicates) in the absence (control) and the presence (experimental) of Chlorhexidine diacetate. We calculated fold change as the ratio between the signal averages of three untreated (control) and three chlorhexidine diacetate-treated (experimental) cultures for 0, 10 and 60 min exposures.

The Enterococcus faecalis V583 transcriptional profile during survival within macrophages

Project description:Transcriptional profiling of E. faecalis V583 during intracellular survival compared to V583 grown to mid-log phase (OD 600 = 0.05) in THB + 1% glucose. In this study we report the complete intracellular E. faecalis V583 transcriptome following infection of RAW264.7 macrophages. During intracellular survival, approximately 45% of the V583 genome was differentially regulated including numerous genes involved in the oxidative stress, heat shock and SOS responses. We observed that the E. faecalis-containing phagosome was limited for glycolytic substrates, nucleotides, amino acids and numerous ions necessary for growth and protein function. Approximately 35% of the genes differentially regulated during survival within macrophages were of hypothetical/unknown function, suggesting that the V583 response to phagocytosis involves many previously unstudied loci. Here, we provide the first comprehensive study elucidating the transcriptional response of E. faecalis to phagocytosis, which may provide new targets for future studies. Bacterial RNA was obtained from RAW264.7 macrophage infected with E. faecalis V583 4, 8 and 12 h post-infection and compared to E. faecalis V583 RNA extracted from mid log growth (OD 600 = 0.05). Control cultures were grown in Todd-Hewett Broth supplemented with 1% glucose.

Project description:The aim was to study the transcriptional profiling of the tdc cluster delection mutant E. faecalis V583 Î?tdc (non-tyramine producer) compared to the wild type strain E. faecalis V583 (tyramine producer). We compared the expression profile of the strains grown in M17 medium with glucose as carbon source and suplemented with tyrosine. E. faecalis V583 Î?tdc cells (test) compared with E. faecalis V583 cells (reference). Both strains grown in GM17 medium suplemented 15 mM tyrosine.
