Project description:Synovial fluid was obtained weekly from nine horses throughout a 70-day study period and subjected to small RNA sequencing. Nine skeletally mature Standardbred trotters (seven mares and two geldings, 2.5-7 years old, weighing 397-528 kg) were included in this study. OA was surgically induced in the left middle carpal joint and the right middle carpal joint underwent sham surgery. Under arthroscopic guidance, an osteochondral fragment was created in the left middle carpal joint by chiselling off a fragment on the dorsodistal surface of the radial carpal bone using an 8 mm curved osteotome. The fragment was left adherent to the joint capsule proximally, and an incongruent defect bed of approximately 15 mm in width was created in the exposed subchondral bone between the fragment and radial carpal bone by drilling with an arthroscopic burr. Debris from the procedure was left inside the joint. Sham surgery (arthroscopy alone) was performed in the right middle carpal joint. All portals were sutured using 2-0 propylene (Surgipro, Medtronic Danmark A/S, Copenhagen, Denmark). Both carpi were bandaged with sterile dressings. Bandages were changed at day 2 and removed at day 5. Sutures were removed 10 days after surgery. From 2 weeks following OA induction and onwards, horses were exercised in 2 min of trot (4.4-5.3 m/sec) , 2 min of fast trot/gallop (9 m/sec) and 2 min of trot (4.4-5.3 m/sec) 5 days a week on a treadmill. The horses were euthanized on day 71 or 72 with an overdose of pentobarbital sodium (140 mg/kg, Euthasol Vet, Dechra Veterinary Products, Uldum, Denmark). One horse, Horse 5, was euthanized according to predefined humane endpoints at day 49, as it became lame at the walk.

Project description:Osteoarthritis (OA) is characterised by loss of articular cartilage, synovial membrane dysfunction and subchondral sclerosis. Few studies have used a global approach to stratify equine synovial fluid (SF) molecular profiles according to OA severity. SF was collected from 58 metacarpophalangeal (MCP) and metatarsophalangeal joints of racing Thoroughbred horses (Hong Kong Jockey Club; HKJC) and 83 MCP joints of mixed breed horses from an abattoir and equine hospital (biobank). Joints were histologically and macroscopically assessed for OA severity. For proteomic analysis, native SF and SF loaded onto ProteoMiner™ equalisation columns, to deplete high abundant proteins, were analysed using liquid chromatography-tandem mass spectrometry (LC-MS/MS) and label-free quantification. Validation of selected differentially expressed proteins was undertaken using clinical SF collected during diagnostic investigations. Native SF metabolites were analysed using 1D 1H Nuclear Magnetic Resonance (NMR). 1,834 proteins and 40 metabolites were identified in equine SF. Afamin levels decreased with synovitis severity and four uncharacterised proteins decreased with OA severity. Gelsolin and lipoprotein binding protein decreased with OA severity and apolipoprotein A1 levels increased for mild and moderate OA. Within the biobank, glutamate levels decreased with OA severity and for the HKJC, 2-aminobutyrate, alanine and creatine increased with severity. Proteomic and metabolomic integration was undertaken using linear regression via Lasso penalisation modelling, incorporating 29 variables (R2=0.82) with principal component 2 able to discriminate advanced OA from earlier stages, predominantly driven by H9GZQ9, F6ZR63 and alanine. Combining biobank and HKJC datasets, discriminant analysis of principal components modelling prediction was good for mild OA (90%). This study has stratified equine OA using both metabolomic and proteomic SF profiles and identified a panel of markers of interest which may be applicable to grading OA severity. This is also the first study to undertake computational integration of NMR metabolomic and LC-MS/MS proteomic datasets of any biological system.

Project description:Equine atypical myopathy (AM) is a severe environmental intoxication linked to the ingestion of protoxins contained in seeds and seedlings of the sycamore maple (Acer pseudoplatanus) in Europe. The toxic metabolites cause a frequently fatal rhabdomyolysis syndrome in grazing horses. Since toxic metabolites can also be present in co-grazing horses, it is still unclear as to why, in a similar environmental context, some horses show signs of AM whereas others remain clinically healthy. Label-free proteomics analyses on the serum of 26 diseased AM, 23 co-grazers and 11 control horses were performed to provide insights into biological processes and pathways. A total of 43 and 44 differentially expressed proteins between “AM vs co-grazing horses” and “AM vs control horses” were found. Disease-linked changes in the proteome of different groups were found to correlate with detected amounts of toxins and principal component analysis was performed to identify the 29 proteins with the greatest impact on the proteomic differences between groups. Among the pathway-specific changes the glycolysis/gluconeogenesis pathway, the coagulation/complement cascade, and the biosynthesis of amino acid were affected. Sycamore maple poisoning results in a combination of inflammation, oxidative stress, and high energy demand, which is trying to be met by enhanced glycolysis.

Project description:The aim of the study was to characterise the protein complement of synovial fluid (SF) in health and osteoarthritis (OA) using liquid chromatography mass spectrometry (LC-MS/MS) following peptide-based depletion of high abundance proteins. SF was used from nine normal and nine OA Thoroughbred horses. Samples were analysed with LC-MS/MS using a NanoAcquity™ LC coupled to a LTQ Orbitrap Velos. In order to enrich the lower-abundance protein fractions protein equalisation was first undertaken using ProteoMiner™. Progenesis-QI™ LC-MS software was used for label-free quantification. In addition immunohistochemistry, western blotting and mRNA expression analysis was undertaken on selected joint tissues.The number of protein identifications was increased by 33% in the Proteominer™ treated SF compared to undepleted SF. A total of 754 proteins were identified in SF. A subset of 10 proteins were identified which were differentially expressed in OA SF. S100-A10, a calcium binding protein was upregulated in OA and validated with western blotting and immunohistochemistry. Several new OA specific peptide fragments (neopeptides) were identified.The protein equalisation method compressed the dynamic range of the synovial proteins identifying the most comprehensive SF proteome to date. A number of proteins were identified for the first time in SF which may be involved in the pathogenesis of OA. We identified a distinct set of proteins and neopeptides that may act as potential biomarkers to distinguish between normal and OA joints.

Project description:Osteoarthritis (OA) causes persistent joint deterioration, severe morbidity, and reduced mobility in both humans and horses. Extracellular vesicles (EVs) are key players in cell-to-cell communication and are thus ideal for biomarker discovery. Here, we used a unique multi-omics strategy to find possible EV biomarkers from the synovial fluid of healthy horses compared to naturally occurring OA and advanced OA. We discovered that SF-EVs’ proteome and phospholipidome are correlated. Furthermore, we found that the quantity of EVs present in SF was unaffected by OA. We discovered crucial signalling pathways involving integrins (p=2.72x10-24), the actin cytoskeleton (p=3.88x10-32), Rho family GTPases (p=1.24x10-19), and the clathrin-mediated endocytosis pathway (p=1.30x10-24). Specifically, the advanced OA group’s immunological response (macrophage binding and interaction) was upregulated. These findings are suggestive that the membrane and protein cargo of SF-EVs are altered in response to OA pathology and may be used as biomarkers of disease. Consequently, SF-EVs could have the potential to be employed in the future to help with OA stratification and prognosis. In addition, this study provides a framework for future research using a larger cohort.

Project description:Myofibrillar myopathy (MFM) in horses is a late onset disease that affects performance and athleticism. It is characterized by myofibrillar disarray and protein aggregation with no known cause. The objective of this study was to elucidate the molecular drivers of MFM in Warmblood (WB) horses by proteomic profiling (5 MFM WB, 4 non-MFM WB) of gluteal muscle. MFM horses used in this study had a chronic history of poor performance and exercise intolerance as well as accumulation of desmin aggregates in > 4 myofibers per muscle sample. The Equine Neuromuscular Diagnostic Laboratory database at Michigan State University was queried to identify WB horses with snap frozen gluteus medius biopsies available for analysis. Non-MFM control horses were defined as horses with no history of exercise intolerance and no evidence of desmin accumulation or other histopathology in muscle biopsies. Muscle biopsy samples were obtained at rest from horses that had not undertaken strenuous exercise in the preceding 48 hours.

Project description:The purpose of this experiment was to further our understanding of gene expression in the central nervous system (thalamus and cerebrum) after exposure to West Nile virus. To that end, three different analyses were performed. The first examined differences in gene expression between horses not vaccinated and exposed to WNV and normal control horses (exposure). The second examined differences in gene expression between horses not vaccinated and exposed to WNV and horses vaccinated and exposed to WNV (survival). And the third examined differences between the nonvaccinated cerebrum and nonvaccinated thalamus of horses exposed to WNV (location). Six conditions- Gene expression in the thalamus and cerebrum of three different groups of horses (Non-vaccinated horses exposed to West Nile virus, Vaccinated horses exposed to West Nile virus, normal horses not exposed to West Nile virus). Biological replicates- 6 normal cerebrums, 6 normal thalamus, 6 vaccinated and exposed cerebrums, 6 vaccinated and exposed thalamus, 6 non-vaccinated and exposed cerebrum, 6 non-vaccinated and exposed thalamus.

Project description:Retinal Müller glial cells (RMG) play a central role in retinal neuroinflammation, actively participating in the immune response and adopting a dynamic inflammatory phenotype. However, the precise functions of RMG in retinal inflammation remain incompletely understood. In this study, we investigated the proteome of RMG isolated from healthy horses and horses affected by equine recurrent uveitis (ERU). ERU serves as a unique spontaneous model of autoimmune uveitis, exhibiting key clinical and pathophysiological features observed in humans, such as a relapsing-remitting course of disease and a CD4+ T cell-driven autoimmune etiology [1, 2]. To gain insights into the molecular landscape of RMG in a spontaneous animal model of autoimmune uveitis and their potential contribution to retinal autoimmune neuroinflammation, we used discovery proteomics for the analysis of RMG from healthy horses and horses with ERU. Our data are not only relevant to veterinary medicine but also hold significant translational value for human ophthalmology as well. Project description:Anoplocephala perfoliata is a common tapeworm in horse with an ability to cause colic and increase mortality. At present, current diagnostic methods are not sensitive enough to detect A. perfoliata and immunoreactive excretory/secretory (E/S) proteins produced by this parasite, which comprise the most promising candidates for diagnostic tests. Here, we compared the E/S proteins produced by A. perfoliata worms of two different physiological states (small- and large-sized worms) in vitro by label-free quantitative (LFQ) proteomics with protein database composed of proteins encoded by Hymenolepis diminuta, Echinococcus multilocularis/granulosus and Taenia aseatica for protein identifications. The quantitative proteomics data was complemented with one-dimensional gel electrophoresis combined with immunoblotting using antisera from horses diagnosed positive and negative for this tapeworm. Protein bands cross-reacting most efficiently with the anti-A. perfoliata antibodies were subjected to in-gel digestion with LC-MS/MS identification. Both the non-gel and gel-based proteomic results were compared to propose a list of candidate proteins for further studies. These proteins will be validated for their suitability as diagnostic markers specific to cestodes and A. perfoliata.
