Project description:Custom microarrays were used to examine differential gene expression between pyrethroid resistant vs pyrethroid susceptible phenotypes of the dengue vector mosquito Aedes aegypti. Pyrethroid resistant population were from Cayenne (French Guiana, GUY), Baie Mahault (Guadeloupe, GUA) and Noumea (New Caledonia, CAL) whilst New Orleans lab colony represented the lab susceptible strain Pools of total RNA was extracted from the whole bodies of 3 day old female mosquitoes that had survived exposure to 0.06% deltamethrin (for GUY, GUA, CAL) . Single colour hybridization experiments were performed using labelled cDNA on the Agilent 'Aedes aegypti detox chip plus': A-MTAB-574. Four unique biological replicates per population were used in the study

Project description:Bacillus thuringiensis israelensis (Bti) toxins are increasingly used for mosquito control, but little is known about the precise mode of action of each of these toxins, and how they interact to kill mosquito larvae. By using RNA sequencing, we investigated change in gene transcription level and polymorphismvariations associatedwith resistance to each Bti Cry toxin and to the full Bti toxin mixture in the dengue vector Aedes aegypti. The upregulation of genes related to chitin metabolismin all selected strain suggests a generalist, non-toxin-specific response to Bti selection in Aedes aegypti. Changes in the transcription level and/or protein sequences of several putative Cry toxin receptors (APNs, ALPs, α-amylases, glucoside hydrolases, ABC transporters) were specific to each Cry toxin. Selective sweeps associated with Cry4Aa resistancewere detected in 2 ALP and 1 APNgenes. The lack of selection of toxin-specific receptors in the Bti-selected strain supports the hypothesis that Cyt toxin acts as a receptor for Cry toxins in mosquitoes.

Transcriptome profiling of Bemisia tabaci reveals stage-specific gene expression signatures for thiamethoxam resistance

Project description:To investigated the stage-specific gene expression response to thiamethoxam in the Bemisia tabaci, we have designed the Agilent eArray platform to identify stage-regulated gene expression towards thiamethoxam exposure. All the B biotype Bemisia tabaci were maintained on cabbage. Thiamethoxam susceptible (TH-S) was cultured without exposure to any chemical insecticides, Thiamethoxam resistance (TH-R) strain exhibited >70-fold resistance to thiamethoxam in comparision to the TH-S strain. Eggs were incubated in 24hours collected as one sample. The fourth nymphs were collected as another sample. The one-day-old unmated adult females were collected as the third samples. Both of the samples were collected from the TH-R, TH-S strains, respectively

Proteomic and transcriptomic responses to thiamethoxam in the Sweetpotato whitefly, Bemisia tabaci (Hemiptera: Aleyrodidae)

Project description:We investigated the transcriptional response to thiamethoxam in the Bemisia tabaci using Illumina sequencing technology. A total of 1,338 genes were differently expressed in the thiamethoxam-resistant whiteflies. All the B biotype whiteflies were maintained on cabbage. The susceptible strain (TH-S) was cultured without exposure to any chemical insecticides. Before the sample collected, almost 10,000 thiamethoxam-resistant adult whiteflies were treated with 2,000 mg/L thiamethoxam (~LC80) to eliminate the heterozygous individuals. Then the survivors were collected after 48 hours and designated as the TH-2000 sample. Then approximately 1,500 adult TH-S and TH-2000 were collected, respectively, The RNA was extracted and sequenced using Illumina HiSeq 2000.

Laboratory Jeddah strain (F5) without exposure to insecticides Laboratory Jeddah strains selected for deltamethrin resistance (F5). For each comparison (field, lab selected or unselected vs susceptible), three biological replicates were used. The two susceptible strains, New Orleans and Rockefeller served as reference.

Project description:We investigate the functional complexity of the Plutella xylostella transcriptome in defending against a Bt toxin using Illumina sequencing technology. Over 2,900 differentially expressed unigenes were obtained in resistant P. xylostella comparison to their susceptible counterpart. All the P. xylostella were maintained on cabbage.The susceptible strain (MM) was cultured without exposure to any Bt toxins.Before the sample collected, Cry1Ac-resistant P. xylostella were treated with 750μg/mL Bt toxin Cry1Ac to eliminate the heterozygous individuals. Then the survivors were collected after 48 hours and designed as the resistant sample (MK and GK). Then fourth-instars larvae midgut tissues of MK,GK and MM were collected, respectively, The RNA was extracted and sequenced using Illunima HiSeq 2000.
