Developmental Transcription Profiles of Wild-Type and WHIRLY Knockdown Barley Leaves

ABSTRACT: Wild-type barley cv Golden Promise and two independent RNAi knockdown lines (W1-1 And W1-7) of the WHIRLY1 gene were grown under controlled environments. Leaves were harvested after 7 or 14 days and sampled along a developmental gradient of base, mid and tip. Microarray analysis was conducted from three biological replicates using a custom designed Agilent barley microarray. Analysis of transcript abundance suggests developmental delay in WHIRLY knockdown lines with significant disruption of plastid development and transcription.
