Selective FRET nano probe based on carbon dots and naphthalimide-isatin for the ratiometric detection of peroxynitrite in drug-induced liver injury.

ABSTRACT: Drug-induced liver injury (DILI) is the most common cause for acute liver failure in the USA and Europe. However, most of DILI cases can recover or be prevented if treatment by the offending drug is discontinued. Recent research indicates that peroxynitrite (ONOO^-) can be a potential indicator to diagnose DILI at an early stage. Therefore, the establishment of an assay to detect and track ONOO^- in DILI cases is urgently needed. Here, a FRET-based ratiometric nano fluorescent probe CD-N-I was developed to detect ONOO^- with high selectivity and excellent sensitivity. This probe consists of carbon dots and a naphthalimide-isatin peroxynitrite sensing system assembled based on electrostatic interactions. Using CD-N-I we were able to detect exogenous ONOO^- in live cells and endogenous ONOO^- in APAP-induced liver injury of HepG2 cells.

A Highly Sensitive and Selective Nano-Fluorescent Probe for Ratiometric and Visual Detection of Oxytetracycline Benefiting from Dual Roles of Nitrogen-Doped Carbon Dots.

Project description:The specific detection of oxytetracycline (OTC) residues is significant for food safety and environmental monitoring. However, rapid specific determination of OTC from various tetracyclines is still challenging due to their similar chemical structures. Here, nitrogen-doped carbon dots (NCDs) with excitation and pH-dependent optical properties and a high-fluorescence quantum yield were successfully synthesized, which were directly employed to fabricate a dual-response fluorescence probe by self-assembly with Eu3+ (NCDs/Eu3+) for the ratiometric determination of OTC. The addition of OTC into the probe greatly enhances the characteristic emission of Eu3+ due to the "antenna effect", and the incorporation of NCDs into the probe further improves the Eu3+ fluorescence by remarkably weakening the quenching effect caused by H2O molecules and efficiently shortening the distance of energy transfer from OTC to Eu3+. Meanwhile, the fluorescence of NCDs apparently decreases due to aggregation-caused quenching. The results demonstrate that a ratiometric detection of OTC (0.1-25 µM) with a detection limit of 29 nM based on the double response signals is achieved. Additionally, visual semi-quantitative assay of OTC can be realized with the naked eye under a 365 nm UV lamp according to the fluorescence color change of the as-fabricated probe. This probe exhibits acceptable specificity and anti-interference for OTC assay, holding promise for the fast detection of OTC in real water and milk samples.

Carbon Dot/Naphthalimide Based Ratiometric Fluorescence Biosensor for Hyaluronidase Detection.

Project description:Bladder cancer is the leading cause of death in patients with genitourinary cancer. An elevated level of hyaluronidase (HAase) was found in bladder cancer, which acts as an important biomarker for the early diagnosis of bladder cancer. Hence, there is a need to develop a simple enzymatic assay for the early recognition of HAase. Herein, we report a simple, sensitive, and ratiometric fluorescence assay for HAase detection under physiological conditions. The fluorescence assay was constructed by the adsorption of cationic carbon dots and positively charged naphthalimide on negatively charged hyaluronic acid and the development of a Förster resonance energy transfer (FRET) mechanism from carbon dots to a naphthalimide fluorophores. The hyaluronidase enzyme cleaves the hyaluronic acid in this assay, and breaking down the FRET mechanism induces ratiometric changes. A detection limit of 0.09 U/mL was achieved, which is less than the HAase level found in normal human body fluids. Moreover, this assay may be used for diagnosing HAase-related diseases.

A ratiometric fluorescent probe for pH detection based on Ag2S quantum dots-carbon dots nanohybrids.

Project description:In this study, a novel ratiometric fluorescent nanoprobe for pH monitoring has been developed by synthesizing red fluorescent Ag2S quantum dots (Ag2S QDs) and green fluorescent carbon dots (CDs) nanohybrids (Ag2S CDs) in one pot using CDs as templates. The nanoprobe exhibits dual-emission peaks at 500 and 670 nm under a single-excitation wavelength of 450 nm. The red fluorescence can be selectively quenched by increasing pH, while the green fluorescence is an internal reference. Therefore, the change of the relative fluorescence intensity (I500/I670) in the ratiometric Ag2S CDs probes can be used for pH sensing. The results revealed that I500/I670 of Ag2S CDs probes was linearly related to pH variation between pH 5.4 and 6.8. Meanwhile, the Ag2S CDs probes possessed a good reversibility along with pH changing between 5.0 and 7.0 without any interruption from common metal ions, proteins and other interferences.

Utilising a 1,8-naphthalimide probe for the ratiometric fluorescent visualisation of caspase-3.

Project description:The development of selective and sensitive probes for monitoring caspase-3 activity-a critical enzyme involved in apoptosis-remains an area of significant interest in biomedical research. Herein, we report the synthesis and characterisation of a novel ratiometric fluorescent probe, Ac-DEVD-PABC-Naph, designed to detect caspase-3 activity. The probe utilises a 1,8-naphthalimide fluorophore covalently linked to a peptide sequence via a self-immolative p-aminobenzyl alcohol (PABA) linker. Upon enzymatic cleavage by caspase-3, the probe undergoes spontaneous degradation, releasing the free naphthalimide fluorophore, resulting in a ratiometric change in fluorescence emission. Spectroscopic studies revealed a time-dependent ratiometric fluorescent response, demonstrating the probe's ability to visualise caspase-3 activity with high sensitivity. Enzyme kinetics such as K m (Michaelis constant), k cat (turnover number), and LOD (Limit of Detection) were obtained, suggesting that the probe possesses comparable kinetic data to other probes in literature, but with the added benefits of ratiometric detection. Selectivity studies also demonstrated the probe's specificity for caspase-3 over other endogenous species and enzymes. Ac-DEVD-PABC-Naph may be a promising tool for the quantitative detection and fluorescent visualisation of caspase-3 activity in biological systems, with potential applications in apoptosis research and drug development.

A naphthalimide-based turn-on fluorescence probe for peroxynitrite detection and imaging in living cells.

Project description:Peroxynitrite (ONOO-) is a potent biological oxidant that plays a significant role in diverse physiological and pathological processes. A novel fluorescent probe HCA-OH was developed for specific and sensitive detection of peroxynitrite, and displayed a significant fluorescence turn-on signal. With low cytotoxicity and good photostability, the probe HCA-OH could be applied in imaging ONOO- distribution in HepG2 cells and live C. elegans in real time. Therefore, the probe can be a promising tool for imaging in vivo.

Nanoscale metal-organic frameworks coated with poly(vinyl alcohol) for ratiometric peroxynitrite sensing through FRET.

Project description:This work describes a facile yet powerful approach to energy-transfer NMOF (nanoscale metal-organic framework) fabrication for ratiometric peroxynitrite (ONOO-) sensing. Poly(vinyl alcohol) (PVA) is chosen to organize the energy donor (NMOF) and acceptor (molecular probes). PVA can conveniently graft onto the NMOF surface and bind to the molecular probes bearing the arylboronic acid group through multiple weak coordination interactions. Due to efficient Förster resonance energy transfer (FRET), the bright blue fluorescence of the NMOF is quenched while the green or red emission from the acceptor is enhanced. Upon reacting with ONOO-, the ONOO- sensors depart from the NMOF and the FRET is interrupted and the fluorescence of the NMOF recovered. Based on this strategy, we developed two ratiometric ONOO- nanosensors for the detection of ONOO- in solutions and living cells. This work is the first report of NMOF ONOO- sensors through FRET and could inspire the design of other NMOF based chemical sensors and biosensors.

BODIPY-Based Ratiometric Fluorescent Probe for Sensing Peroxynitrite in Inflammatory Cells and Tissues.

Project description:Peroxynitrite (ONOO-) plays an important role in many physiological and pathological processes. Excessive ONOO- in cells leads to oxidative stress and inflammation. However, precise monitoring of ONOO- levels in specific organelles (e.g., mitochondria) is still lacking and urgently needed. Herein, we rationally designed a mitochondria-targeted ratiometric fluorescent probe, MOBDP-I, for imaging of ONOO- in the mitochondria of inflammatory cells and model mice. This probe, MOBDP-I, was synthesized by conjugating a BODIPY fluorophore to a mitochondria-targeting moiety-indole-salt group by a carbon-carbon double bond (C=C). In the presence of ONOO-, the C=C bond between the BODIPY backbone and the indole-salt group was oxidized and broken, leading to an 18-fold enhancement of fluorescence at 510 nm, along with a significant fluorescence decrease at 596 nm. The ratiometric response property bestowed the probe with advantages in the precise quantification of ONOO- in cells, thus allowing estimation of the extent of inflammation in living cells and mouse models of rheumatoid arthritis, peritonitis, and brain inflammation. MOBDP-I could act as an effective molecular tool to study the relationship between ONOO- and the occurrence and development of inflammatory diseases.

Ratiometric fluorescence probe of Cu2+ and biothiols by using carbon dots and copper nanoclusters.

Project description:A novel dual-emission ratiometric fluorescent probe based on N-doped yellow fluorescent carbon dots (y-CDs) and blue fluorescent copper nanoclusters (CuNCs) was established for quantitative determination of Cu2+ and biothiols. In this work, the Cu2+-(y-CDs) complexes formed by the chelation of y-CDs with Cu2+, showed an absorption peak at 430 nm that not only enhanced the fluorescence of y-CDs through inhibiting photoinduced electron transfer (PET) but also effectively quenched the fluorescence of CuNCs due to Förster resonance energy transfer (FRET). In addition, the chelation of y-CDs with Cu2+ could be inhibited by biothiols that prevented the fluorescence of y-CDs from being enhanced and the fluorescence of CuNCs from being quenched. On account of the changes of ratiometric signal, a dual-emission fluorescence probe for Cu2+ and biothiols determination was achieved. The proposed method exhibited high sensitivity for Cu2+ and biothiols in the ranges of 0.5-100 μM and 0.8-50 μM and the limits of detection (LODs) of Cu2+, glutathione (GSH), cysteine (Cys) and homocysteine (Hcy) were 0.21 μM, 0.33 μM, 0.39 μM and 0.46 μM, respectively. Subsequently, the established strategy presented an application prospect for the detection of Cu2+ and biothiols in real samples.

Multi-carbon dots and aptamer based signal amplification ratiometric fluorescence probe for protein tyrosine kinase 7 detection.

Project description:BackgroundProtein tyrosine kinase 7 (PTK 7) is a membrane receptor, which can be found in various kinds of cancers. In view of this, detection of PTK 7 in the peripheral circulation would be an effective way for the early diagnosis of cancer.ResultsIn this work, a multi-carbon dots and aptamer-based signal amplification ratiometric fluorescence probe was developed. The fluorescence of the aptamer-modified y-CDs and b-CDs were respectively chosen as the detection signal and interior label. The fluorescence of y-CDs was quenched by Fe3O4 and cDNA (complement to aptamer) compound without PTK 7, but recovered by the addition of PTK 7. Then, the free aptamer was cut by DNase I, which amplified the detection signal. The ratiometric fluorescence sensor for PTK 7 was established with the LOD of 0.016 ng mL-1.ConclusionsSummary, a multi-carbon dots and aptamer-based signal amplification ratiometric fluorescence probe was developed for the detection of protein tyrosine kinase 7. The developed probe was applied to PTK 7 detection in MCF-7 cells and human serum with satisfying results, thus indicating that this probe has huge potential in clinical practice.

Ratiometric Polymer Probe for Detection of Peroxynitrite and the Application for Live-Cell Imaging.

Project description:Peroxynitrite (ONOO-) is one of the sources of oxidation stress involved in many biological signaling pathways. The role of ONOO- being a double-edged sword in biological systems drives the development of effective detection tools. In this work, a boronate-based polymeric fluorescent probe PB-PVA was synthesized and the probe performance was evaluated. The probe exhibits ratiometric sensing of ONOO- in a range of 0-6 µM. There is good linear relationship between the probe fluorescence intensity ratio and ONOO- concentration. The probe also displays moderate selectivity towards ONOO- over other ROS. Moreover, it is water-soluble and possesses good biocompatibility which aids the imaging of ONOO- in living cells. These properties could make the probe a promising tool in in vitro study related to ONOO-.
