Cucurbit[10]uril.

ABSTRACT: Melamine diamine 1 is able to displace CB[5] from the CB[10].CB[5] complex resulting in CB[10].12 and precipitated CB[5].1. We were able to isolate free CB[10] by treatment of CB[10].1 with acetic anhydride followed by washing with MeOH, DMSO, and water. The spacious cavity of CB[10] is able to complex large guests, including a cationic calix[4]arene derivative in its 1,3-alternate form (CB[10].1,3-alt-3). The addition of adamantane carboxylic acid (4) to CB[10].3 triggers a conformational change during the formation of termolecular complex CB[10].cone-3.4.

Project description:The synthesis of acyclic cucurbit[n]uril dendrimers G1-G3 that bear four dendrons on their aromatic sidewalls via thiolate S(N)2 chemistry is reported. G1-G3 are polycationic and can bind to pEGFP plasmid DNA as shown by dynamic light scattering (DLS), gel electrophoresis, and scanning electron microscopy (SEM). The gene delivery ability of G1-G3 is presented.

Expected and unexpected photoreactions of 9-(10-)substituted anthracene derivatives in cucurbit[<i>n</i>]uril hosts.

Project description:By arranging substrates in a "reaction ready" state through noncovalent interactions, supramolecular nanoreactors/catalysts show high selectivity and/or rate acceleration features. Herein, we report the host-guest complexation of 9-(10-)substituted anthracene derivatives (G1-G3) with cucurbit[n]uril (CB[n], n = 8, 10), and the photoreactions of these derivatives in the presence of CB[n] hosts. Both CB[10] and CB[8] showed no obvious effects on the photoreaction of 9,10-disubstituted derivative G1. For G2 and G3, CB[10] operated as either a nanoreactor or catalyst (10%) for the photodimerization of two compounds with high selectivity and high yield. However, although CB[8] formed a 1 : 2 complex with G2, as also observed with CB[10], the photosolvolysis product (9-anthracenemethanol) was obtained quantitatively after photoirradiation of the CB[8]·2G2 complex. This unexpected photosolvolysis was rationalized by a plausible catalytic cycle in which anthracene acts as a photoremovable protecting group (PPG) and the carbonium ion intermediate is stabilized by CB[8].

Cucurbit[7]uril-based high-performance catalytic microreactors.

Project description:Catalytic microreactors manufactured using microfluidic devices have received significant research interest in recent years. However, little attention has been paid to immobilising metallic nanoparticles (NPs) onto microchannel walls for high efficiency catalytic reactions. We demonstrate a facile preparation of cucurbit[7]uril-based catalytic microreactors, where metallic NPs are immobilised onto microchannels via supramolecular complexation with methyl viologen@cucurbit[7]uril (CB[7]). These microreactors exhibit a remarkable catalytic activity owing to the substantially high surface area to volume ratio of the microchannels and metallic NPs. Superior to most conventional heterogeneous catalytic reactions, separation post reaction and complicated recycling steps of the catalysts are not required. Moreover, CB[7] can complex a variety of metallic NPs to its portal, providing a multifunctional high-performance in situ catalytic platform.

4-Sulfocalix[4]arene/Cucurbit[7]uril-Based Supramolecular Assemblies through the Outer Surface Interactions of Cucurbit[<i>n</i>]uril.

Project description:Upon mixing of aqueous solutions of the freely soluble building blocks cucurbit[7]uril (Q[7]) and 4-sulfocalix[4]arene (SC[4]A), white microcrystals instantly separate in near-quantitative yield. The driving force for this assembly is suggested to be the outer-surface interaction of the Q[n]. Dynamic light scattering, scanning electron microscopy, and NMR (diffusion-ordered NMR spectroscopy) analyses have confirmed the supramolecular aggregation of Q[7] and SC[4]A. Titration 1H NMR spectroscopy and isothermal titration calorimetry have shown that the interaction ratio of Q[7] and SC[4]A is close to 3:1. Moreover, the Q[7]/SC[4]A-based supramolecular assembly can accommodate molecules of some volatile compounds or luminescent dyes. Thus, this work offers a simple and highly efficient means of preparing adsorbent or solid fluorescent materials.

pH-Mediated Single Molecule Conductance of Cucurbit[7]uril.

Project description:Recognition tunneling technique owns the capability for investigating and characterizing molecules at single molecule level. Here, we investigated the conductance value of cucurbit[7]uril (CB[7]) and melphalan@CB[7] (Mel@CB[7]) complex molecular junctions by using recognition tunneling technique. The conductances of CB[7] and Mel@CB[7] with different pH values were studied in aqueous media as well as organic solvent. Both pH value and guest molecule have an impact on the conductance of CB[7] molecular junction. The conductances of CB[7] and Mel@CB[7] both showed slightly difference on the conductance under different measurement systems. This work extends the molecular conductance measurement to aqueous media and provides new insights of pH-responsive host-guest system for single molecule detection through electrical measurements.

Supramolecular Complexation of Azobenzene Dyes by Cucurbit[7]uril.

Project description:This report describes cucurbit[7]uril (CB7) complexation of azobenzene dyes that have a 4-(N,N'-dimethylamino) or 4-amino substituent. Absorption and NMR data show that CB7 encapsulates the protonated form of the azobenzene and that the complexed dye exists as its azonium tautomer with a trans azo conformation and substantial quinoid resonance character. Because CB7 complexation stabilizes the dye conjugate acid, there is an upward shift in its pKa, and in one specific case, the pKa of the protonated azobenzene is increased from 3.09 to 4.47. Molecular modeling indicates that the CB7/azobenzene complex is stabilized by three major noncovalent factors: (i) ion-dipole interactions between the partially cationic 4-(N,N'-dimethylamino) or 4-amino group on the encapsulated protonated azobenzene and the electronegative carbonyl oxygens on CB7, (ii) inclusion of the upper aryl ring of the azobenzene within the hydrophobic CB7 cavity, and (iii) a hydrogen bond between the proton on the azo nitrogen and CB7 carbonyls. CB7 complexation enhances azobenzene stability and increases azobenzene hydrophilicity; thus, it is a promising way to improve azobenzene performance as a pigment or prodrug. In addition, the striking yellow/pink color change that accompanies CB7 complexation can be exploited to create azobenzene dye displacement assays with naked eye detection.

Cucurbit[6]uril p-xylylenediammonium diiodide deca-hydrate inclusion complex.

Project description:The title inclusion complex, C(36)H(36)N(24)O(12)·C(8)H(14)N(2) (2+)·2I(-)·10H(2)O, displays a large ellipsoidal deformation of the cucurbit[6]uril (CB[6]) skeleton upon complex formation. The benzene ring of the cation is rotationally disordered between two orientations in a ratio of 3:1. The solvent H(2)O mol-ecules form a hydrogen-bonded network by inter-action with the carbonyl groups of CB[6] and the I(-) counterions. The crystal studied exhibited non-merohedral twinning. Both CB[6] and the cation are centrosymmetric.

Host-guest complexation of cucurbit[8]uril with two enantiomers.

Project description:Host-guest complexation of cucurbit[8]uril (Q[8]) with two enantiomers, D-3-(2-naphthyl)-alanine (D-NA) and L-3-(2-naphthyl)-alanine (L-NA), has been fully investigated. Experimental data indicate that double guests reside within the cavity of Q[8] in both aqueous solution and solid state, generating highly stable homoternary complexes D-NA2@Q[8] and L-NA2@Q[8].

Evaluation of the Immunosafety of Cucurbit[n]uril In Vivo.

Project description:Cucurbiturils are a family of macrocyclic oligomers capable of forming host-guest complexes with various molecules. Due to noncovalent binding to drug molecules and low toxicity, cucurbiturils has been extensively investigated as potential carriers for drug delivery. However, the immune system's interactions with different drug carriers, including cucurbiturils, are still under investigation. In this study, we focused on cucurbiturils' immunosafety and immunomodulation properties in vivo. We measured blood counts and lymphocyte subpopulations in blood, spleen, and bone marrow, and assessed the in vivo toxicity to spleen and bone marrow cells after intraperitoneal administration to BALB/c mice. When assessing the effect of cucurbit[6]uril on blood parameters after three intraperitoneal injections within a week in laboratory animals, a decrease in white blood cells was found in mice after injections of cucurbit[6]util, but the observed decrease in the number of white blood cells was within the normal range. At the same time, cucurbit[7]uril and cucurbit[8]uril did not affect the leukocyte counts of mice after three injections. Changes in the number of platelets, erythrocytes, and monocytes, as well as in several other indicators, such as hematocrit or erythrocyte volumetric dispersion, were not detected. We show that cucurbiturils do not have immunotoxicity in vivo, with the exception of a cytotoxic effect on spleen cells after сucurbit[7]uril administration at a high dosage. We also evaluated the effect of cucurbiturils on cellular and humoral immune responses. We founded that cucurbiturils in high concentrations affect the immune system in vivo, and the action of various cucurbiturils differs in different homologues, which is apparently associated with different interactions in the internal environment of the body.

Synthesis and self-assembly processes of monofunctionalized cucurbit[7]uril.

Project description:We present a building-block approach toward functionalized CB[7] derivatives by the condensation of methylene-bridged glycoluril hexamer 1 and glycoluril bis(cyclic ethers) 2 and 12. The CB[7] derivatives Me2CB[7] and CyCB[7] are highly soluble in water (264 mM and 181 mM, respectively). As a result of the high intrinsic solubility of Me2CB[7], it is able to solubilize the insoluble benzimidazole drug albendazole. The reaction of hexamer 1 with glycoluril derivative 12, which bears a primary alkyl chloride group, gives CB[7] derivative 18 in 16% isolated yield. Compound 18 reacts with NaN3 to yield azide-substituted CB[7] 19 in 81% yield, which subsequently undergoes click reaction with propargylammonium chloride (21) to yield CB[7] derivative 20 in 95% yield, which bears a covalently attached triazolyl ammonium group along its equator. The results of NMR spectroscopy ((1)H, variable-temperature, and DOSY) and electrospray mass spectrometry establish that 20 undergoes self-assembly to form a cyclic tetrameric assembly (204) in aqueous solution. CB[7] derivatives bearing reactive functional groups (e.g., N3, Cl) are now available for incorporation into more complex functional systems.
